Septin 14, also known as SEPT14, is a 432 amino acid protein that belongs to the septin family. The highly conserved septin family of GTP-binding cytoskeletal proteins is implicated in membrane transport, apoptosis, cell polarity, cell cycle regulation, cytokinesis and other cellular functions. Septins polymerize into heterooligomeric protein complexes that form filaments, and can associate with cellular membranes, actin filaments and microtubules. Septin 14 has a GTPase domain followed by a C-terminal coiled-coil domain characteristic of group II septins. Septin 14 interacted with all septins except those that are members of its phylogenetic cluster, including itself. Septin 14 shares highest identity (67%) with Septin 10. Expressed in testis and more weakly in fetal liver, tonsil and thymus, the Septin 14 protein is not detected in testicular cancer or other normal or cancer cell lines. The Septin 14 gene is conserved in chimpanzee, dog, cow, mouse and rat, and maps to human chromosome 7p11.2.

G0S2 is a 103 amino acid novel target of peroxisome proliferator-activated receptors (PPARs) and regulator of latent HIV. G0S2 may be involved in adipocyte differentiation and its expression is essential for committing cells to enter the G1 phase of the cell cycle. G0S2 contains a CpG-rich island and multiple sites for potential phosphorylation by casein kinase II and protein kinase C. The gene encoding G0S2 maps to human chromosome 1, which is the largest human chromosome. Chromosome 1 spans about 260 million base pairs and makes up 8% of the human genome. There are about 3,000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1. Notably, the rare aging disease Hutchinson-Gilford progeria is associated with the LMNA gene which encodes lamin A. Stickler syndrome, Parkinsons, Gaucher disease and Usher syndrome are also associated with chromosome 1. Aberrations in chromosome 1 are found in a variety of cancers including head and neck cancer, malignant melanoma and multiple myeloma.

Vasopressin, also known as arginine vasopressin (AVP) or antidiuretic hormone (ADH), is a posterior pituitary hormone that is synthesised in the hypothalamus. Vasopressin is synthesised as a precursor protein that consists of arginine vasopressin and two associated proteins, neurophysin 2 and the glycopeptide copeptin. Vasopressin, together with its carrier protein neurophysin II, is packaged into neurosecretory vesicles and transported axonally to the nerve endings in the neurohypophysis, where it is either stored or secreted into the bloodstream. Vasopressin acts as a growth factor by enhancing pH regulation through acid-base transport systems. It has a direct antidiuretic action on the kidney and also causes vasoconstriction of the peripheral vessels. Vasopressin can also contract smooth muscle during parturition and lactation. It also plays a role in cognition, tolerance, adaptation and complex sexual and maternal behaviour, as well as in the regulation of water excretion and cardiovascular functions. Mutations in the vasopressin precursor cause autosomal dominant neurohypophyseal diabetes insipidus (ADNDI), which is characterised by persistant thirst, polydipsia and polyuria.

This gene is a member of the beta-1,3-galactosyltransferase (beta3GalT) gene family. This family encodes type II membrane-bound glycoproteins with diverse enzymatic functions using different donor substrates (UDP-galactose and UDP-N-acetylglucosamine) and different acceptor sugars (N-acetylglucosamine, galactose, N-acetylgalactosamine). The beta3GalT genes are distantly related to the Drosophila Brainiac gene and have the protein coding sequence contained in a single exon. The beta3GalT proteins also contain conserved sequences not found in the beta4GalT or alpha3GalT proteins. The carbohydrate chains synthesized by these enzymes are designated as type 1, whereas beta4GalT enzymes synthesize type 2 carbohydrate chains. The ratio of type 1:type 2 chains changes during embryogenesis. By sequence similarity, the beta3GalT genes fall into at least two groups: beta3GalT4 and 4 other beta3GalT genes (beta3GalT1-3, beta3GalT5). The encoded protein of this gene does not use N-acetylglucosamine as an acceptor sugar at all. In mammalian cells, transcription is regulated in part by high molecular weight coactivating complexes that mediate signals between transcriptional activators and RNA polymerase (1). These complexes include CRSP (for cofactor required for Sp1 activation), which is required, in conjunction with TAFIIs, for transcriptional activation by Sp1 (2). Cytokeratin 10 is a heterotetramer of two type I and two type II keratins. Cytokeratin 10 is generally associated with keratin 1. It is seen in all suprabasal cell layers including stratum corneum. A number of alleles are known that mainly differ in the Gly-rich region (positions 490-560). Defects in cytokeratin 10 are a cause of epidermolytic hyperkeratosis (EHK), also known as bullous congenital ichthyosiform erythroderma (BCIE) or bullous erythroderma ichthyosiformis congenita of Brocq. EHK is an hereditary skin disorder characterized by blistering and a marked thickening of the stratum corneum. At birth, affected individuals usually present with redness, blisters and superficial erosions due to cytolysis. Within a few weeks, the erythroderma and blister formation diminish and hyperkeratoses develop. Transmission is autosomal dominant, but most cases are sporadic. Defects in cytokeratin 10 are also a cause of annular epidermolytic ichthyosis (AEI), also known as cyclic ichthyosis with epidermolytic hyperkeratosis. AEI resembles clinical and histologic features of both epidermolytic hyperkeratosis and ichthyosis bullosa of Siemens.
