Expressed at high levels in kidney, colon, liver, prostate and small intestine, HGD uses iron as a cofactor to catalyse the oxygen-dependent conversion of homogentisate to 4-maleylacetoacetate, a reaction that is the fourth step in the creation of L-phenylalanine from fumarate and acetoacetic acid. Defects in the gene encoding HGD are the cause of alkaptonuria (AKU), an autosomal recessive disorder that is characterised by urine that turns dark on standing and alkalinisation, black ochronotic pigmentation of cartilage and collagenous tissues and spine arthritis.

Tetramethylrhodamine isothiocyanate Isomer R (TRITC) is the R isomer of tetramethylrhodamine isothiocyanate and is also known as 6-tetramethylrhodamine isothiocyanate. It is a strongly fluorescent compound soluble in ethanol, methanol, dimethylformamide, dimethylsulfoxide, and water. TRITC is a moderately-sized aminoxanthene dye carrying carboxylic acid and isothiocyanate substituents on the non-planar pendant phenyl ring. It forms a lipophilic cation in acidic pH and a hydrophilic zwitterion around neutral pH. The isothiocyanate group reacts with nucleophilic substituents (e.g., amino, hydroxyl, thiol) of biomolecules, providing the means of attaching a fluorescent label. TRITC also reacts with water and hydroxide ions and is unstable in aqueous solutions. TRITC is commercially available as a zwitterion or as a chloride.

A furin-like protease processes UCMA into an N-terminal 37 amino acid peptide and a C-terminal 74 amino acid peptide, which is referred to as Unique cartilage matrix-associated protein C-terminal fragment (Ucma-C). Introduction of recombinant Ucma-C interferes with osteogenic differentiation of mesenchymal stem cells, MC3T3-E1 preosteoblasts and primary osteoblasts. This suggests that Ucma may be involved in the negative regulation of osteogenic differentiation of osteochondrogenic precursor cells at the cartilage-bone interface and in peripheral zones of fetal cartilage.
