The RING-type zinc finger motif is present in a number of viral and eukaryotic proteins and is made of a conserved cysteine-rich domain that is able to bind two zinc atoms. Proteins that contain this conserved domain are generally involved in the ubiquitination pathway of protein degradation. MARCH10 (membrane-associated ring finger (C3HC4) 10), also known as RNF190 (ring finger protein 190) or MARCH-X, is an 808 amino acid protein with one RING-CH-type zinc finger domain. MARCH10 may function as an E3 ubiquitin-protein ligase that accepts a ubiquitin residue from an E2 ubiquitin-conjugating enzyme and immediately transfers that residue to a protein that is targeted for degradation.

This gene encodes a member of the F-box protein family which is characterized by an approximately 40 amino acid motif, the F-box. The F-box proteins constitute one of the four subunits of ubiquitin protein ligase complex called SCFs (SKP1-cullin-F-box), which function in phosphorylation-dependent ubiquitination. The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. The protein encoded by this gene belongs to the Fbls class and, in addition to an F-box, contains at least 9 tandem leucine-rich repeats. [provided by RefSeq, Jul 2008].

FIG4 is a 907 amino acid protein that contains one SAC domain through which it is thought to function as a phosphoinositide phosphatase that may play an important role in signal transduction and vesicle trafficking. Defects in the gene encoding FIG4 are the cause of Charcot-Marie-Tooth disease type 4J (CMT4J) and amyotrophic lateral sclerosis type 11 (ALS11), both of which negatively affect the nervous system. CMT4J is a recessive demyelinating disorder of the peripheral nervous system and is characterized by reduced motor nerve conduction velocities and axonal degeneration. Unlike CMT4J, ALS11 is characterized by the degeneration of upper motor neurons in the brain and lower neurons in the spinal cord, causing paralysis and, ultimately, death.

The PSD-95/SAP 90 family of proteins, which are known to bind to and cluster various membrane proteins, are involved in the organization of synaptic structure. SynGAP (for synaptic GTPase activating protein), a novel synaptic RasGAP, is a brain-specific protein abundant in the cortex, hippocampus and olfactory bulb. SynGAP interacts with all three PDZ domains within PSD-95/ SAP 90 proteins. It represents one of three classes of mammalian RasGAPs and is specifically localized to excitatory synapses possessing NMDA receptors. SynGAP may be involved in the regulation of BDNF as well as Ras signaling. Its activity is inhibited by phosphorylation by CaMKII, which could result in the activation of the MAP kinase pathway.

GMEB-1 is a 573 amino acid protein that contains one SAND domain and is a member of the KDWK family of combinatorial transcription modulators. Localized to both the cytoplasm and the nucleus, GMEB-1 forms a heterodimer with GMEB-2 (Glucocorticoid modulatory element-binding protein 2) and, once associated with GMEB-2, plays a key role in parvovirus DNA replication. In addition, GMEB-1 functions alone as a trans-acting factor that, by binding to glucocorticoid modulatory elements (GMEs) in TAT (tyrosine aminotransferase) promoters, increases intracellular sensitivity to glucocorticoid concentrations. GMEB-1 also interacts with initiator procaspases and, via this interaction, can inhibit caspase-induced apoptosis. Due to alternative splicing events, GMEB-1 is expressed as two isoforms.

FBXO36 is a 188 amino acid protein that contains one forty amino acid F-box region, making it a member of the F-box family. F-box proteins are critical components of the SCF (Skp1-CUL-1-F-box protein) type E3 ubiquitin ligase complex and are involved in substrate recognition and recruitment for ubiquitination. F-box proteins are members of a large family that regulates cell cycle, immune response, signaling cascades and developmental programs by targeting proteins, such as cyclins, cyclin-dependent kinase inhibitors, IkB-å and ∫-catenin, for degradation by the proteasome after ubiquitination. Functioning as a component of the SCF complex, FBXO36 is thought to recognize and bind to select phosphorylated proteins, thereby promoting their ubiquitination and subsequent degradation.

Hip (HSP70-interacting protein), also known as ST13 (suppression of tumorigenicity protein 13), is one of several co-chaperones that regulate activities of the HSP70 chaperone family (1,2). The homo-oligomeric protein Hip cooperates with HSP70 in protein folding by stabilizing the ADP-bound state of HSP70. Hip directly binds to the ATPase domain of HSP70 when it is converted to the ADP-bound state by proteins of the HSP40 family (3). By collaborating with other positive co-factors such as HSP40 and Hop, or competing with negative co-factors such as Bag1, Hip may facilitate the chaperone function of HSP70 in protein folding and repair, and in controlling the activity of regulatory proteins such as steroid receptors and various regulators of proliferation or apoptosis (4-8).

Importin 7 is a 1,038 amino acid protein encoded by the human gene IPO7. Importin-7 belongs to the importin b family and contains one importin N-terminal domain. Importin-7 functions in nuclear protein import, either by acting as an autonomous nuclear transport receptor or as an adapter-like protein in association with the Importin b subunit KPNB1. Acting autonomously, Importin-7 is thought to serve itself as receptor for nuclear localization signals (NLS) and to promote translocation of import substrates through the nuclear pore complex (NPC) by an energy requiring, Ran-dependent mechanism. At the nucleoplasmic side of the NPC, Ran binds to Importin-7, the Importin-7/substrate complex dissociates and Importin-7 is re-exported from the nucleus to the cytoplasm where GTP hydrolysis releases Ran. Importin-7 is a nuclear protein that is expressed in most tissues.
