The death domain (DD) superfamily of proteins share one or more of the following domains: the DD, DED (death-effector domain), CARD (caspase-recruitment domain) and PYD (Pyrin domain). Each of these domains is characterized by a canonical death domain fold, which consists of a bundle of five or six antiparallel α-helices. Recognizes a protein of 30-33 kDa, which is identified as CD20 (Workshop V; Code CD20.12. Workshop IV; Code B17). It recognizes an extracellular domain of CD20. It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. The protein passes through the membrane 4 times with both ends in cytoplasm and exposes one short and one longer loop to the external environment. CD20 is not glycosylated in resting B-cells and its cytoplasmic domains are differentially phosphorylated upon activation. It acts as calcium channel involved in B cell activation and cell cycle progression.

PIPPIN is a Y-box protein (also called cold-shock (CSD) domain-containing protein) and belongs to a family of highly conserved RNA-binding transcriptional regulators. Predominantly expressed in brain cells and localizing to the nucleus and the cytoplasm, PIPPIN contains two RNA-binding motifs, namely PIP1 and PIP2, and one CSD domain. PIPPIN functions as a nucleic acid binding regulatory factor and is believed to participate in brain maturation. More specifically, PIPPIN binds to the 3’-UTR ends of the mRNAs encoding Histone H1 and Histone H3.3. This interaction requires all of the PIPPIN domains to work in concert as one functional protein. In addition, PIPPIN can be sumoylated in a thyroid hormone (T3)-dependent manner. This suggests that PIPPIN modification in response to extracellular stimuli may modulate the regulation of protein synthesis.

PCDHB11 is a member of the protocadherin beta gene cluster, one of three related gene clusters tandemly linked on chromosome five. PCDHB11 clusters demonstrate an unusual genomic organization similar to that of B-cell and T-cell receptor gene clusters. The beta cluster contains 16 genes and 3 pseudogenes, each encoding 6 extracellular cadherin domains and a cytoplasmic tail that deviates from others in the cadherin superfamily. The extracellular domains interact in a homophilic manner to specify differential cell-cell connections. Unlike the alpha and gamma clusters, the transcripts from these genes are made up of only one large exon, not sharing common 3' exons as expected. These neural cadherin-like cell adhesion proteins are integral plasma membrane proteins. Their specific functions are unknown but they most likely play a critical role in the establishment and function of specific cell-cell neural connections.

Glycogen synthesis is initiated by the autoglucosylation of Glycogenin-1. Specifically, Glycogenin-1 glucosylates itself to begin the synthesis of glycogen in mammalian skeletal muscle. It acts as the primer to which further glucose monomers may be added. All of the Glycogenin-1 molecules contain at least one glucosyl residue before autoglucosylation begins. The first step of the glycogen synthesis occurs when a glucose molecule from UDP-glucose binds to the hydroxyl group of Tyr 194 on the Glycogenin-1 molecule. Using its glucosyltransferase activity, Glycogenin-1 adds more glucoses, each one coming from UDP-glucose. The glycosylation process reaches a plateau when five new glucose residues have been added, at which point glycogen synthase (GS) takes over and further elongates the chain. Glycogenin-1 remains covalently attached to the reducing end of the glycogen molecule.
