MP21 is a member of the EMP24/GP25L/p24 cargo family of proteins that regulates vesicular trafficking in the early secretory pathway. TMP21 is a ubiquitously expressed single-pass type I membrane protein localizing to the Golgi cisternae and the plasma membrane. It contains one GOLD (Golgi dynamics) domain and participates in protein transport and quality control between the endoplasmic reticulum (ER) and the Golgi complex. In addition, TMP21 is a component of the heteromeric secretase complex (or Presenilin complex) and functions to regulate the ?secretase activity. Two isoforms, namely TMP21-I and TMP21-II, exist for this protein.

Albumin is a soluble, monomeric protein which comprises about one-half of the blood serum protein. Albumin functions primarily as a carrier protein for steroids, fatty acids, and thyroid hormones and plays a role in stabilizing extracellular fluid volume. Albumin is a globular unglycosylated serum protein of molecular weight 65,000. Albumin is synthesized in the liver as preproalbumin which has an N-terminal peptide that is removed before the nascent protein is released from the rough endoplasmic reticulum. The synaptogyrin family of proteins are integral membrane proteins containing four transmembrane regions. Synaptogyrins are tyrosine-phosphorylated proteins with two neuronal (Synaptogyrins 1 and 3) and one ubiquitous (Synaptogyrin-2) isoform. Synaptophysin and synaptogyrin represent the major constituents of synaptic vesicles. The 26kDa protein Synaptogyrin-1 is associated with presynaptic vesicles in neuronal cells. Synaptogyrin-2, also known as Cellugyrin has a tyrosine phosphorylated C-terminal cytoplasmic tail, and is involved in the regulation of membrane traffic in non-neuronal cells. Synaptogyrin-3 is expressed mainly in brain and placenta. The SYNGR4 gene encodes for the 234 amino acid protein Synaptogyrin-4.

FBXO25 is a member of the F box protein family which is characterized by an approximately 40 amino acid motif, the F box. The F box proteins constitute one of the four subunits of ubiquitin protein ligase complex called SCFs (SKP1 cullin F box), which function in phosphorylation dependent ubiquitination. The F box proteins are divided into 3 classes: Fbws containing WD 40 domains, Fbls containing leucine rich repeats, and Fbxs containing either different protein protein interaction modules or no recognizable motifs. FBXO25 belongs to the Fbxs class. There are three named isoforms produced by alternative splicing.FBXO25, also known as FBX25, is a 367 amino acid protein that contains one C-terminal F-box domain and belongs to the Fbx class of the F-box family of proteins. F-box proteins are critical components of the SCF (Skp1-CUL-1-F-box protein) type E3 ubiquitin ligase complex and are involved in substrate recognition and recruitment for ubiquitination. They are members of a larger family of proteins that are involved in the regulation of a wide variety of cellular processes (including the cell cycle, immune response, signaling cascades and developmental processes) through the targeting of proteins, such as cyclins, cyclin-dependent kinase inhibitors, IkB-a and b-catenin, for degradation by the proteasome after ubiquitination. Expressed at high levels in brain, FBXO25 localizes predominantly to the nucleus and directly interacts with Skp1 p19 and CUL-1. Disruption of the gene encoding FBXO25 can lead to X-linked mental retardation.

Xylenol orange tetrasodium salt is a fluorochrome acid dye with complexing and chelating properties that make it useful as an indicator in the spectrophotometric detection and assay of metal ions such as calcium, cadmium, bismuth, zinc, lead, and mercury. Its tetrasodium salt especially is used as an indicator in metal titrations. It changes color from yellow to red between pH 6.4 to 10.4. It has six potentially anionic substituents, one being sulfonate, and is anionic under staining conditions and during most laboratory applications.

This MAb is specific to kappa light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.

CLN8, a 286 amino acid transmembrane protein, localizes mainly to the endoplasmic reticulum, but also partially to the ER-Golgi intermediate compartment (ERGIC). Mutations in the CLN8 gene cause neuronal ceroid lipofuscinosis 8 and progressive epilepsy with mental retardation (EPMR). Both disorders are forms of neuronal ceroid-lipofuscinose (NCL), a group of progressive neurodegenerative diseases found in children, characterised by failure of psychomotor development, impaired vision, seizures and premature death. The CLN8 protein is one of eight proteins in the CLN family, including CLN1-CLN7, which are associated with NCL.
