The MDC gene is one of several Cys-Cys (CC) cytokine genes which are clustered on the q arm of chromosome 16. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. The cytokine encoded by MDC displays chemotactic activity for natural killer cells, chronically activated T lymphocytes, monocytes and dendritic cells. It has no chemoattractant activity for eosinophils, neutrophils and resting T lymphocytes and also displays a mild activity for primary activated T lymphocytes. The product of this gene binds to chemokine receptor CCR4. This chemokine may play a role in the trafficking of activated T lymphocytes to inflammatory sites and other aspects of activated T lymphocyte physiology.

Hip (HSP70-interacting protein), also known as ST13 (suppression of tumorigenicity protein 13), is one of several co-chaperones that regulate activities of the HSP70 chaperone family (1,2). The homo-oligomeric protein Hip cooperates with HSP70 in protein folding by stabilizing the ADP-bound state of HSP70. Hip directly binds to the ATPase domain of HSP70 when it is converted to the ADP-bound state by proteins of the HSP40 family (3). By collaborating with other positive co-factors such as HSP40 and Hop, or competing with negative co-factors such as Bag1, Hip may facilitate the chaperone function of HSP70 in protein folding and repair, and in controlling the activity of regulatory proteins such as steroid receptors and various regulators of proliferation or apoptosis (4-8).

This gene is a proto-oncogene that encodes a RING finger E3 ubiquitin ligase. The encoded protein is one of the enzymes required for targeting substrates for degradation by the proteasome. This protein mediates the transfer of ubiquitin from ubiquitin conjugating enzymes (E2) to specific substrates. This protein also contains an N-terminal phosphotyrosine binding domain that allows it to interact with numerous tyrosine-phosphorylated substrates and target them for proteasome degradation. As such it functions as a negative regulator of many signal transduction pathways. This gene has been found to be mutated or translocated in many cancers including acute myeloid leukaemia. Adducins are a family of cytoskeleton proteins encoded by three genes (alpha, beta and gamma). Adducin is a protein associated with the inner leaflet of the plasma membrane and is one of the proteins localized at the spectrin-Actin junction of the membrane skeleton. The cortical Actin cytoskeletal network is lost during apoptosis and Adducins are central in the cortical Actin network organization. Adducin alpha is a cytoskeletal protein involved with sodium-pump activity in the renal tubule and is associated with hypertension. The expression of Adducin alpha and Adducin gamma is ubiquitous in contrast to the restricted expression of Adducin beta . Adducin beta is expressed at high levels in brain and hematopoietic tissues, such as bone marrow in humans and spleen in mice.

This antibody neutralizes the activity of cyclin D1 in vivo. About 60% of mantle cell lymphomas (MCL) contain a t(11; 14)(q13; q32) translocation resulting in over-expression of cyclin D1. This antibody is useful in identifying mantle cell lymphomas (cyclin D1 positive) from CLL/SLL and follicular lymphomas (cyclin D1 negative). Occasionally, hairy cell leukemia and plasma cell myeloma weakly express Cyclin D1.

This MAb is specific to kappa light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. It recognizes human Ig kappa light chains of both secreted and cell surface immunoglobulin. It detects also free kappa light chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.
