Cleaved by the protease thrombin to yield monomers which, together with fibrinogen beta (FGB) and fibrinogen gamma (FGG), polymerize to form an insoluble fibrin matrix. Fibrin has a major function in hemostasis as one of the primary components of blood clots. In addition, functions during the early stages of wound repair to stabilize the lesion and guide cell migration during re-epithelialization. Was originally thought to be essential for platelet aggregation, based on in vitro studies using anticoagulated blood. However, subsequent studies have shown that it is not absolutely required for thrombus formation in vivo. Enhances expression of SELP in activated platelets via an ITGB3-dependent pathway. Maternal fibrinogen is essential for successful pregnancy. Fibrin deposition is also associated with infection, where it protects against IFNG-mediated hemorrhage. May also facilitate the immune response via both innate and T-cell mediated pathways.

Protects against apoptosis mediated by BAX.Apoptosis, or programmed cell death, is related to many diseases, such as cancer. Apoptosis is triggered by a variety of stimuli including members in the TNF family and prevented by the inhibitor of apoptosis (IAP) proteins. IAP proteins form a conserved gene family including IAP, XIAP/ILP-1/MIHA, and Livin/KIAP that bind to and inhibits specific proteases. A novel member in the IAP protein family was recently identified and designated ILP-2 for IAP-like protein-2 (1). ILP-2 has high homology to ILP-1, but is encoded by a distinct gene that is solely expressed in testis of tested normal human tissues (1). ILP-2, unlike ILP-1, has no inhibitory effect on Fas and TNF induced apoptosis, but potently inhibits apoptosis induced by overexpression of Bax or by coexpression of caspase-9 with Apaf-1. ILP-2 interacts with the processed caspase-9. These results suggest that ILP-2 is a novel IAP family member with restricted specificity for caspase-9.

CCDC116 is a 515 amino acid protein that exists as two alternatively spliced isoforms. Encoded by a gene that maps to human chromosome 22q11.21, CCDC116 is induced by curcumin (diferulolylmethane), although its role is unclear. CCDC116 is significantly affected by dietary curcumin, which may have a protective role in inflammatory bowel disease (IBD) and may reduce the relapse rate in human ulcerative colitis (UC). As the second smallest human chromosome, chromosome 22 contains over 500 genes and about 49 million bases. Phelan-McDermid syndrome, Neurofibromatosis type 2 and autism are associated with chromosome 22. A schizophrenia susceptibility locus has been identified on chromosome 22 and studies show that 22q11 deletion symptoms include a high incidence of schizophrenia. Translocation between chromosomes 9 and 22 may lead to the formation of Philadelphia Chromosome and subsequent production of a novel fusion protein known as BCR-Abl, a potent cell proliferation activator found in several types of leukemia.

Guanidine Hydrochloride is a protein denaturant and thus having an important role in molecular weight determinations.
Guanidine Hydrochloride is a strong chaotropic agent useful for the denaturation and subsequent refolding of proteins. This strong denaturant can solubilize insoluble or denatured proteins such as inclusion bodies. This can be used as the first step in refolding proteins or enzymes into their active form. Urea and dithiothreitol (DTT) may also be necessary. Guanidine HCl is used in the isolation of RNA to dissociate the nucleoprotein into its nucleic acid and protein moieties. It is an inhibitor of RNase. Highly concentrated (6 - 8 M) Guanidine HCl solutions are used to denature native globular proteins. It apparently disrupts hydrogen bonds which hold the protein in its unique structure. However, there also is evidence suggesting that guanidine hydrocholoride may disrupt hydrophobic interactions by promoting the solubility of hydrophobic residues in aqueous solutions.

Guanidine hydrochloride consists of a network of guanidinium cations and chloride anions linked by N–H•••Cl hydrogen bonds, it is a strong chaotropic agent. Guanidine HCl may agglomerate upon storage. It may appear as a free-flowing crystalline powder, a free flowing powder with solid material dispersed throughout, or a solid. The quality of the product does not appear to be affected and solutions prepared from the free-flowing and lumpy guanidine HCl appear identical.
