Mono-ADP-ribosylation is one of the posttranslational protein modifications regulating cellular metabolism (e.g. nitrogen fixation) in prokaryotes. Mono-ADP-ribosylation is a process in which the ADP-ribose moiety of nicotinamide adenine dinucleotide is transferred to an acceptor amino acid. Five mammalian ADP-ribosyltransferases (ART1-ART5) have been cloned, and expression is restricted to tissues such as cardiac and skeletal muscle, leukocytes, brain and testis. ART3 (ADP-ribosyltransferase 3), also known as Ecto-ADP-ribosyltransferase 3, is a testis specific membrane protein that does not appear to have ADP-ribosyltransferase activity. It lacks the R-S-EXE active site motif and is therefore unable to catalyze the reaction. ART3 is predominantly found in spermatocytes and may play a role in spermatogenesis.

PCSK1N is a 260 amino acid protein that is both secreted and localized to the trans-Golgi network. Expressed in pancreas and brain, PCSK1N is thought to play a role in the control of the neuroendocrine secretory pathway and may also be involved in PCSK1 inhibition. The gene encoding PCSK1N maps to human chromosome X, which contains nearly 153 million base pairs and houses over 1,000 genes. In conjunction with chromosome Y, chromosome X is responsible for sex determination, as an X and a Y chromosome lead to normal male development, while two copies of an X chromosome lead to normal female development. There are a number of conditions related to an abnormal number and combination of sex chromosomes, some of which include Turner's syndrome, color blindness, hemophilia and Duchenne muscular dystrophy.

P55 contains a PDZ/DHR domain, a conserved SH-3 domain that appears to suppress tyrosine kinase activity of various oncoproteins, a 39-amino acid motif that binds to cytoskeletal protein 4.1R, and a guanylate kinase-like domain. Interaction with glycophorin C (GPC) and 4.1R suggests that p55 may play a role in the dynamic regulation in the erythrocyte membrane. In addition, p55 gene expression <i>in vivo</i> may be associated with a CpG island. P55 is constitutively expressed in K562 erythroleukemia cells during erythropoiesis and undergoes a 2-fold amplification after induction.

The DCDC2 gene encodes the DCDC2 protein (Doublecortin-containing protein 2, RU2, RU2S) which contains two Doublecortin peptide domains similar to those in the Doublecortin gene. DCDC2 is transcribed as a “normal†gene, which results in a sense transcript (RU2S), but when it is transcribed in the opposite direction, a shorter antisense transcript (RU2AS), which is found in tumors, results. The DCDC2 protein demonstrates ubiquitous expression, whereas RU2AS expression is restricted to normal kidney, bladder, liver and testis, and to tumors of various histologic origins.
