Retinoic acid (RA; active metabolite of vitamin A) plays a prominent role in regulating the transition of proliferating precursor cells (such as carcinoma cells and neuronal precursors) to postmitotic differentiated cells (Joshi et al., 2005). The retinoid X receptors (RXRs) family (RXRalpha, beta and gamma), preferentially bind 9-cis-RA and regulate gene transcription by forming heterodimers with a second family of RA receptors. RAs have been suggested to potentially play a therapeutic role in cervical cancer (Abu et al., 2005). RAs are known to play key roles in neuronal development and an increasing body of evidence indicates that retinoid signaling may regulate synaptic plasticity and associated learning and memory behaviors (Lane and Bailey, 2005).

Heterogeneous nuclear ribonucleoprotein A1 (hnRNP A1) is a member of the hnRNP A/B family of related RNA binding proteins that bind pre-mRNA and are involved in the processing, metabolism and transport of nuclear pre-mRNA transcripts. hnRNP A1 regulates the alternative splicing of c-Src, c-H-Ras and modifies initiation of translation of the fibroblast growth factor 2 mRNA. hnRNP A1 expression level is elevated in many cancers; knockdown of hnRNP A1 leads to apoptosis in various cancer cells. Although predominantly nuclear, hnRNP A1 is continually transported from the nucleus to the cytoplasm where it disassociates from mRNA and is rapidly re-imported into the nucleus. hnRNP A1 binds to cis-acting repressive sequences (CRS) of HIV-1 to influence HIV-1 production. HIV-1 enhances hnRNP A1 expression and promotes the relocalization of hnRNP A1 to the cytoplasm ,

HOX11, also called homeobox11 or Tcl-3 is located at the 10q24 T-Cell translocation breakpoint region. It encodes a homeobox-domain containing protein, HOX11, containing a glycine and proline-rich amino terminus. HOX11 is required for maintenance of the developing spleen and cell survival. The HOX11protein binds to a specific DNA sequence and localizes to the cell nucleus. It transactivates transcription of a reporter gene linked to a cis-regulatory element, acting as a positive transcription activator. The HOX11 gene is highly expressed in T-ALLs as a result of a t (7:10) (q35;q24) or t(10:14)(q24;qll) translocation. The homeobox gene is deregulated upon translocation into the T-cell receptor locus and activates a specific subset of tumor-associated target genes. HOX 11 gene translocation has been attributed to T-cell leukemia and lymphoid neoplasias.

Mouse myeloid cell surface antigen CD33(CD33) is a member of the immunoglobulin superfamily and SIGLEC (sialic acid binding Ig-like lectin) family. CD33 contains one Ig-like C2-type domain and one Ig-like V-type domain. CD33 is a putative adhesion molecule of myelomonocytic-derived cells that mediates sialic-acid dependent binding to cells. CD33 preferentially binds to alpha-2,6-linked sialic acid. The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface. In the immune response, CD33 may act as an inhibitory receptor upon ligand induced tyrosine phosphorylation by recruiting cytoplasmic phosphatase(s) via their SH2 domain(s) that block signal transduction through dephosphorylation of signalling molecules. CD33 induces apoptosis in acute myeloid leukemia. CD33 is becoming increasingly important as a target of antibody-mediated therapy in acute myeloid leukaemia (AML).

ECH1 is a member of the hydratase/isomerase superfamily. ECH1 shows high sequence similarity to enoyl-coenzyme A (CoA) hydratases of several species, particularly within a conserved domain characteristic of these proteins. The protein contains a C-terminal peroxisomal targeting sequence, localizes to the peroxisome. This enzyme functions in the auxiliary step of the fatty acid beta-oxidation pathway.This gene encodes a member of the hydratase/isomerase superfamily. The gene product shows high sequence similarity to enoyl-coenzyme A (CoA) hydratases of several species, particularly within a conserved domain characteristic of these proteins. The encoded protein, which contains a C-terminal peroxisomal targeting sequence, localizes to the peroxisome. The rat ortholog, which localizes to the matrix of both the peroxisome and mitochondria, can isomerize 3-trans,5-cis-dienoyl-CoA to 2-trans,4-trans-dienoyl-CoA, indicating that it is a delta3,5-delta2,4-dienoyl-CoA isomerase. This enzyme functions in the auxiliary step of the fatty acid beta-oxidation pathway. Expression of the rat gene is induced by peroxisome proliferators.

Protein kinase C inhibitor (IC50 = 50-200 µM depending on assay conditions in MCF-7 cells or IC50 = 100 µM in rat brain). The PKC inhibition is also dependent on the phosopholipid concentration. Also inhibits both calmodulin-dependent and calmodulin-independent Ca2+-, Mg2+-ATPase. Tamoxifen is a selective estrogen response modifier (SERM), anti-angiogenetic factor. DL-Dithiothreitol is also known as Clelands reagent; Protective agent for sulfhydryl groups (-SH). Quantitatively reduces disulfides (-S-S- to -SH). In this reaction the DTT is oxidized to the cyclic disulfide which ensures the reduction of other disulfides in solution. Disulfide reduction occurs quickly at pH 8.
Dithiothreitol is useful for stabilizing sulfhydryl containing enzymes. Effective in sample buffers for reducing protein disulfide bonds prior to SDS-PAGE. DTT can also be used for reducing the disulfide bridge of the cross-linker N,N'-bis(acryloyl)cystamine to break apart the matrix of a polyacrylamide gel. DTT is less pungent and is less toxic than 2-mercaptoethanol. Useful for stabilizing sulfhydryl-containing enzymes.
