SKAP55 and SKAP55-R bind to Fyb through their SH3 domains and function as substrates for p59Fyn in resting T cells (1–3). SKAP55 contains an amino-terminal pleckstrin homology domain and a carboxy-terminal SH3 domain binding motif of adjacent arginine and lysine residues followed by tandem tyrosines (i.e. RKxxYxxY) (4,5). SKAP55-R, similar in overall structure to SKAP55, contains a coiled-coil N-terminal domain (1,2). SKAP55 associates with SLAP-130, another component of the Fyn complex, which plays a role in the regulation of signaling events initiated by lymphocyte antigen receptors leading up to T cell activation (6). The human Fyb gene maps to chromosome 5p13.1 and encodes a 783 amino acid protein (7).

The RING-type zinc finger motif is present in a number of viral and eukaryotic proteins and is made of a conserved cysteine-rich domain that is able to bind two zinc atoms. Proteins that contain this conserved domain are generally involved in the ubiquitination pathway of protein degradation. RNF10 (ring finger protein 10), also known as RIE2, is an 811 amino acid protein that localizes to the cytoplasm and contains one RING-type zinc finger. Existing as multiple alternatively spliced isoforms, RNF10 interacts with MOX-2 and is thought to regulate its transcription in schwann cells, possibly playing a role in myelin formation. The gene encoding RNF10 maps to human chromosome 12, which encodes over 1,100 genes and comprises approximately 4.5% of the human genome. Chromosome 12 is associated with a variety of diseases and afflictions, including hypochondrogenesis, achondrogenesis, Kniest dysplasia, Noonan syndrome and Trisomy 12p, which causes facial developmental defects and seizure disorders.

The breast cancer susceptibility gene (BRCA1) localizes to chromosome 17q. Mutations within this gene account for approximately 45% of families with high incidence of breast cancer and at least 80% of families with increased incidence of both early-onset breast cancer and ovarian cancer. A second breast cancer susceptibility gene, BRCA2, located on chromosome 13q12-13, also confers a high incidence of breast cancer, but unlike BRCA1, BRCA2 does not confer a substantially elevated risk of ovarian cancer. The BRCA2-Associated Factor 35 (BRAF35) protein forms a complex with BRCA2, which associates with condensed chromatin during histone H3 phosphorylation. BRAF35 expression levels are highest in proliferating tissues and parallel BRCA2 expression patterns. The structure of BRAF35 includes a kinesin-like coiled coil domain and a nonspecific DNA binding HMG domain. The chromatin localization of BRAF35 and antibody microinjection studies indicate a role for the BRAF35/BRCA2 complex in cell cycle regulation.

Recognizes a 53 kDa protein, which is identified as p53 suppressor gene product. It reacts with the mutant as well as the wild form of p53. Its epitope maps within the N-terminus (aa 37-45) of p53. Monoclonal antibody PAb1801 does not block the binding of DO-7 MAb to p53 in an ELISA test. p53 is a tumor suppressor gene expressed in a wide variety of tissue types and is involved in regulating cell growth, replication, and apoptosis. It binds to MDM2, SV40 T antigen and human papilloma virus E6 protein. Positive nuclear staining with p53 antibody has been reported to be a negative prognostic factor in breast carcinoma, lung carcinoma, colorectal, and urothelial carcinoma. Anti-p53 positivity has also been used to differentiate uterine serous carcinoma from endometrioid carcinoma as well as to detect intratubular germ cell neoplasia. Mutations involving p53 are found in a wide variety of malignant tumors, including breast, ovarian, bladder, colon, lung, and melanoma.

Heterotrimeric G proteins function to relay information from cell surface receptors to intracellular effectors. Each of a very broad range of receptors specifically detects an extracellular stimulus (a photon, pheromone, odorant, hormone or neurotransmitter) while the effectors (i.e., adenyl cyclase), which act to generate one or more intracellular messengers, are less numerous. In mammals, G protein Alpha, Beta and Gamma polypeptides are encoded by at least 16, 4 and 7 genes, respectively. Most interest in G proteins has been focused on their Alpha subunits, since these proteins bind and hydrolyze GTP and most obviously regulate the activity of the best studied effectors. Four distinct classes of G Alpha subunits have been identified; these include G Alpha s, G Alpha i, G Alpha q and G Alpha 12/13. The two members of the fourth class of G Alpha subunit proteins, G Alpha 12 and G Alpha 13, are insensitive to ADP-ribosylation by pertussis toxin, share 67% identity with each other and less than 45% identity with other G Alpha subunits and are widely expressed in a broad range of tissues.
