ILVBL is a 632 amino acid single-pass membrane protein that belongs to the TPP enzyme family. Expressed in the majority of tissues, ILVBL has the highest level of expression in heart, pancreas and placenta. ILVBL is highly homologous to several bacterial enzymes, including the B isozyme of the large catalytic subunit of E. coli acetohydroxy-acid synthase (AHAS) and the oxalyl-coA decarboxylase of O. formigenes, that utilize thiamine pyrophosphate as a cofactor. ILVBL binds one magnesium ion and one thiamine pyrophosphate per subunit, and may catalyze the initial step in branched-chain amino acid biosynthesis. The family of guanylin regulatory peptides, including guanylin and uroguanylin, are strongly expressed in intestinal mucosa and regulate intestinal fluid secretion during digestion. Guanylins are also involved in acid neutralization and the regulation of membrane-bound guanylate cyclase signaling molecules. Guanylin and uroguanylin are secreted primarily in the stomach, intestine, and colon. Guanylin is also detected in plasma. Guanylin is an endogenous activator of intestinal guanylate cyclase. It stimulates intestinal guanylate cyclase through the same receptor binding region as the heat-stable enterotoxins. Gut enterochromaffin cells synthesize guanylin to be a prohormone of 115 amino acids which is then is processed to the molecular form of 94 amino acids. This 10kDa form is found circulating in the blood.

HDAC6 is a member of the class II mammalian histone deacetylases. Human HDAC6 is composed of 1215 amino acid residues. It possesses two separate putative catalytic domains. Both catalytic domains are fully functional HDACs and contribute independently to the overall activity of HDAC6 protein. A very potent NES is present at the amino-terminus of HDAC6, which was found to play an important role in regulating the shuttling of HDAC6 protein between cytoplasm and nucleus. The shuttling process may be a critical regulatory mechanism of HDAC6 function. The expression of HDAC6 is tightly linked to the state of cell differentiation. HDAC6 may participate in coordinating expression of a group of genes involved in the remodelling of chromatin during cell differentiation.
