The cadherins represent a family of Ca²⁺ dependent adhesion molecules that function to mediate cell to cell binding that is critical for the maintenance of structure and morphogenesis. Cadherins each contain a large extracellular domain at the N-terminus, which is characterised by a series of five homologous repeats, the most distal of which is thought to be responsible for binding specificity. The relatively short C-terminal intracellular domain interacts with a variety of cytoplasmic proteins, including -catenin, to regulate cadherin function. The cadherin superfamily includes cadherins, protocadherins, desmogleins and desmocollins. FAT3 (FAT tumour suppressor homolog 3, also known as CDHF15 or CDHR10, is a 4,589 amino acid single-pass type I membrane protein expressed in ES cells, primitive neuroectoderm, fetal brain, infant brain, adult neural tissues and prostate. Containing thirty-three cadherin domains, four EGF-like domains and one laminin G-like domain, FAT3 may participate in the interactions between neurites derived from specific subsets of neurons during development.

The Ca²⁺/calmodulin-dependent protein kinases (CaM kinases) are a structurally related subfamily of serine/threonine kinases that includes CaMKI, CaMKII, CaMKIV and myosin light chain kinases (MYLK, also designated MLCK). The MYLK kinases phosphorylate myosin regulatory light chains to catalyse myosin interaction with actin filaments resulting in contractile activity. Non-muscle, smooth muscle and skeletal/cardiac muscle MYLK isoforms exist. The MYLK gene (also designated MYLK1) encodes both smooth muscle and non-muscle isoforms as well as telokin, a small C-terminal isoform expressed only in smooth muscle with the capacity to stabilize unphosphorylated myosin filaments. Multiple transcript variants are described for the MYLK gene. Smooth-muscle and non-muscle MYLK isoforms are expressed in a wide variety of adult and fetal tissues. The skeletal/cardiac muscle isoforms of MYLK are encoded by a separate gene, MYLK2 (also designated skMLCK). MYLK appears to be a target for PAKs (p21-activated kinases). PAK1 interaction with MYLK results in a decrease in MYLK activity and myosin light chain phosphorylation.

The calpains have papain like activity, thus the pain nomenclature. Both Calpain 1 and Calpain 2 are ubiquitously expressed, and are countered by the endogenous calpain inhibitor, calpastatin. Other calpain family members (calpain 94, ncl2, ncl3, etc) have more limited tissue distribution, and perhaps different functions. The calpain family members consist of a common small subunit (Calpain 4), and a large variable subunit. It is not clear that all calpains contain a small subunit. Domains in the large subunit include the amino terminal domain I, the proteinase domain II, domain III, and the EF hand domain IV. The calpains appear to serve multiple physiological roles, and ideas concerning the functions of these enzymes are in a state of rapid flux.

Replication factor C (RFC) is an essential DNA polymerase accessory protein that is required for numerous aspects of DNA metabolism, including DNA replication, DNA repair and telomere metabolism. RFC is a heteropentameric complex that recognizes a primer on a template DNA, binds to a primer terminus and loads proliferating cell nuclear antigen (PCNA) onto DNA at primer-template junctions in an ATP-dependent reaction. All five of the RFC subunits share a set of related sequences (RFC boxes) that include nucleotide-binding consensus sequences. Four of the five RFC genes (including RFC1, RFC2, RFC3 and RFC4) have consensus ATP-binding motifs. The small RFC proteins, RFC2, RFC3, RFC4 and RFC5, interact with Rad24, whereas the RFC1 subunit does not. RFC1 is a substrate for caspase-3 in vitro and is cleaved by a caspase-3-like protease during FAS-mediated apoptosis. In addition, phosphorylation of the PCNA binding domain of RFC1 by Ca²⁺/calmodulin-dependent protein kinase II (CaMKII) inhibits DNA synthesis. The human RFC1 gene maps to chromosome 4p14 and encodes the RFC1 subunit.

ORAI3 Monoclonal Antibody: Antigen stimulation of immune cells triggers Ca++ entry t hrough Ca++ release-activated Ca++ (CRAC) channels. ORAI3 is one of two mammalian homologs to ORAI1, a recently identified four-transmembrane spanning protein that is an essential component of CRAC. All three homologs have been shown to function as Ca++ plasma membrane channels gated through interactions with STIM1, the store-activated endoplasmic reticulum Ca++ sensor. However, ORAI3 channels failed to produce detectable Ca++ selective currents in cells co-transfected with ORAI3 and STIM1, indicating that ORAI3 channels undergo a lesser degree of depotentiation than ORAI1 or ORAI2. Na+ currents through ORAI1, 2 and 3 channels were equally inhibited by extracellular Ca++, indicating that each have similar affinities for Ca++ within the selectivity filter. This antibody is predicted to have no cross-reactivity to ORAI1 or ORAI2. Larger molecular weight bands are sometimes seen in SDS-PAGE; these may represent post-translationally modified ORAI 3.
