Creatinine, the end product of creatine catabolism is a normal constituent of urine; daily output about 25 mg/kg body weight. The increased amounts in the urine are typically associated with substantially impaired renal function. Also found together with creatine in muscle tissues and blood. It reacts with picric acid under alkaline conditions to form a Janovski complex. The rate of formation of the colored complex, measured at 480-520 nm is proportional to the creatinine concentration.
Creatinine is a useful marker for normalizing levels of other molecules found in urine and the altered creatinine levels may be associated with conditions that result in decreased renal blood flow, such as diabetes and cardiovascular disease.

DL-Dithiothreitol is also known as Clelands reagent; Protective agent for sulfhydryl groups (-SH). Quantitatively reduces disulfides (-S-S- to -SH). In this reaction the DTT is oxidized to the cyclic disulfide which ensures the reduction of other disulfides in solution. Disulfide reduction occurs quickly at pH 8.
Dithiothreitol is useful for stabilizing sulfhydryl containing enzymes. Effective in sample buffers for reducing protein disulfide bonds prior to SDS-PAGE. DTT can also be used for reducing the disulfide bridge of the cross-linker N,N'-bis(acryloyl)cystamine to break apart the matrix of a polyacrylamide gel. DTT is less pungent and is less toxic than 2-mercaptoethanol. Useful for stabilizing sulfhydryl-containing enzymes.

Dimethyl Sulfoxide is a dipola, aprotic solvent. It has been shown to accelerate strand renaturation (1-10% concentration) and is believed to give the nucleic acid thermal stability against depurination. Dimethyl Sulfoxide is used to enhance dermal absorption of many chemicals, as a solvent for many organic and inorganic compounds including fats, carbohydrates, dyes, resins, and polymers, in antifreeze or hydraulic fluids, as a cryopreservative for cell cultures, oxidation of thiols and disulfides to sulfonic acids, as a PCR cosolvent to help improve yields, especially in long PCR. DMSO is routinely used in polymerase chan reaction (PCR), amplification of cDNA libraries, DNA sequencing, column-loading buffers for poly (A)+ RNA selection, buffers for the transformation of competent E. coli, and transfection protocols. To prepare sterile solutions use a teflon or nylon membrane to sterile-filter the DMSO - do not use a cellulose acetate membrane.

2,3,5-Triphenyltetrazolium Chloride is a redox indicator. In the TTC assay (also known as TTC test or tetrazolium test), TTC is used to differentiate between metabolically active and inactive tissues. The white compound is enzymatically reduced to red TPF (1,3,5-triphenylformazan) in living tissues due to the activity of various dehydrogenases (enzymes important in oxidation of organic compounds and thus cellular metabolism), while it remains as white TTC in areas of necrosis since these enzymes have been either denatured or degraded.
2,3,5-Triphenyltetrazolium Chloride is commonly used in biochemical experiments especially to indicate cellular respiration. TTC has been employed in autopsy pathology to assist identification of post-mortem myocardial infarctions. Glycine is a non-essential amino acid. It is only amino acid with no asymmetric carbon and thus is not chiral. It is the major inhibitory neurotransmitter. It is involved in the biosynthesis of the porphyrin rings of hemes and chlorophylls. Glycine is commonly used in buffer solutions, in electrophoresis and preparative chromatography. A study of the folding of monoclonal antibodies in the presence of glycine and their subsequent purification has been published. The use of glycine in the purification of lipopolysaccharides, lipooligosaccharides, and lipid A has been reported. It is an amino acid for use in cell culture media development applications and existing media formulations. Glycine is commonly used as a component in Tris-glycine and Tris-glycine-SDS running buffers for polyacrylamide gel electrophoresis, a component of Towbin's transfer buffer for Western blots, a buffer substance in cryoenzymology, in osmotic pressure maintenance in isoelectric focusing of erythrocytes, salting-in effect in protein chemistry, and as a buffer component in the coupled phosphatase-kinase reaction for end labelling of restriction fragments. Methotrexate is a cell cycle arresting agent with varying effects. Methotrexate has been reported to arrest the cell cycle in late G1/S thus leading to the inhibition of the synthesis of DNA, RNA, thymidylates, and proteins. The main mechanism of action is reported to involve the inhibition of enzymes involved in purine metabolism which leads to the accumulation of adenosine or the suppression of intercellular adhesion molecule expression by T cells. Additionally, this compound has been observed to inhibit DHFR.
Methotrexate is used for chemotherapy either alone or in combination with other agents. It is effective for the treatment of a number of cancers including: breast, head and neck, leukemia, lymphoma, lung, osteosarcoma, bladder, and trophoblastic neoplasms. It is also used in treatment of autoimmune diseases, ectopic pregnancy, and for the induction of medical abortions. It is used to inhibit dihydrofolate reductase in DHFR-based protein expression systems. Also effective in treatment of pyrimethamine-resistant Plasmodium vivax malaria parasites. Potent inhibitor of dihydrofolate reductase and agent for antitumor studies. Use to inhibit dihydrofolate reductase in DHFR-based protein expression systems. Also shows immunosuppressive effects in, e.g., rheumatoid arthritis. Methotrexate is an allosteric inhibititor of dihydrofolate reductase (DHFR), the enzyme that catalyzes the conversion of dihydrofolate to tetrahydrofolate. Since tetrahydrolfolate is required for purine and pyrimidine synthesis, methotrexate treatment results in the inhibition of DNA and RNA synthesis.
