Golgin 245 is also known as p230, GCP2, GOLG or golgi autoantigen and is a 2,230 amino acid protein that is expressed as three isoforms. Golgin 245 is localized to the cytoplasm in cells and is a member of the golgin family, all of which are peripheral membrane proteins associated with the Golgi complex. Golgin 245 has a carboxyl-terminal GRIP domain, which attaches to the trans-Golgi network (TGN) and TGN-derived vesicles. It is thought that the interaction that takes place between golgin 245 and MACF1, which cross-links microtubules to the Actin cytoskeleton, allows proteins to be transported from the TGN to the cell periphery. against golgin 245 are associated with two chronic disorders known as Sjé°ƒren's syndrome and Hepatitis B.

When mutations are introduced in obese gene, mice with impaired function of leptin are massively obese and in high risk of diabetes. Leptin deficiency reduces metablic rate. Leptin deficient mice are less active and with lower body temperature than normal animals. Human Leptin shares approximately 84% sequence identity with the mouse protein. Human Leptin consists of 167 amino acid residue including a 21 amino acid residue signal sequence and 146 amino acid residue mature protein sequence.

Protocadherin-20 (PCDH20), also known as protocadherin-13, is a 924 amino acid protein containing 6 cadherin domains and potentially functioning as a calcium-dependent cell-adhesion protein. In non-small cell lung cancer cell lines, a homozygous loss of PCDH20 was identified through either deletion of one allele and methylation of the other or methylation of both alleles. Hypermethylation of PCDH20 is associated with worse prognosis and clinical outcome, suggesting that PCDH20 may function as a tumor suppressor.

Sulpho-NHS reacts primarily with primary amines to convert carboxyl groups to amine reactive sulfo-NHS esters. It is ideal for cross-linking, chemical labelling and solid support immobilisation. It is used to generate amine reactive esters from carboxylate groups and is primarily used to increase the efficiency of the carbodiimide EDC cross-linking. EDC reactions do not require Sulpho-NHS; however, its presence greatly enhances cross-linking efficiency and allows for a two-step process. EDC reacts with a carboxylate group to form an active ester, which is subject to rapid hydrolysis in aqueous buffers. The Sulfo-NHS increases the stability of the active intermediate and the resulting coupled product is identical to EDC reactions without sulpho-NHS.

HAS1, HAS2 and HAS3 are HA (hyaluronan or hyaluronic acid) synthase proteins. The extracellular matrix in most vertebrates express HA, which is a high molecular weight linear polysaccharide composed of alternating glucuronic acid and N-acetylglucosamine residues linked by b-1,3 and b-1,4 glycosidic bonds. The three HAS genes show distinct patterns of expression during development and their protein products play significantly different roles in the formation of the HA matrix. Both HAS1 and HAS2 synthesize high molecular-weight HA, whereas HAS3 produces lower molecular weight HA. The expression of the three HAS isoforms is more prominent in growing cells than in resting cells and is differentially regulated by various stimuli, suggesting distinct functional roles of the three proteins. HAS3 produces both secreted and cell-associated forms of hyaluronan and is the most active of the three isoforms of this enzyme in adults. HAS3 gene expression plays a crucial role in the regulation of hyaluronan synthesis in the epidermis. Specifically, IFN-g markedly upregulates HAS3 mRNA, whereas TGF Beta downregulates HAS3 transcript levels. The human HAS3 gene maps to chromosome 16q22.1.

SLAM Family Member 6 (SLAMF6) is a 60 kD single-pass type I membrane protein that belongs to the SLAM subgroup of the CD2 family. Human SLAMF6/ NTB-A contains a 205 amino acid extracellular domain (ECD) with one Ig-like V-set and one Ig-like C2-set domain, a 21 amino acid transmembrane segment and an 84 amino acid cytoplasmic domain, with two immunoreceptor tyrosine-based switch motifs. SLAMF6 is a homodimer. SLAMF6 can interact with PTN6 and, upon phosphorylation, with PTN11 and SH2D1A/SAP. Phosphorylation-dependent NTB-A association with SAP is required for full production of IFN- gamma by NK cells and independent of EAT-2 binding. It Triggers cytolytic activity only in natural killer cells (NK) expressing high surface densities of natural cytotoxicity receptors. On B cells, NTB-A modulates immunoglobulin class switching and the balance between tolerance and autoimmunity.
