Myomegalin, is a 2346 amino acid protein that contains one NBPF domain and localizes to the nucleus, cytoplasm, centrosome and Golgi apparatus. Expressed at high levels in fetal and adult heart and at lower levels in brain and placenta, myomegalin is thought to function as an anchoring protein that sequesters members of the cAMP-dependent pathway to the Golgi and to centrosomes, thereby mediating cAMP pathway dynamics. Translocations in the gene that encodes myomegalin are associated with myeloproliferative disorders (MBDs), a group of diseases caused by an overproduction of blood cells. Myomegalin exists as twelve isoforms due to alternative splicing events.

Recognizes a protein of 36 kDa, identified as cyclin D1. Cyclin D1, one of the key cell cycle regulators, is a putative proto-oncogene overexpressed in a wide variety of human neoplasms. This antibody neutralizes the activity of cyclin D1 in vivo. About 60% of mantle cell lymphomas (MCL) contain a t(11; 14)(q13; q32) translocation resulting in over-expression of cyclin D1. This antibody is useful in identifying mantle cell lymphomas (cyclin D1 positive) from CLL/SLL and follicular lymphomas (cyclin D1 negative). Occasionally, hairy cell leukemia and plasma cell myeloma weakly express Cyclin D1.

Tyrosinase is a copper-containing metalloglycoprotein that catalyzes several steps in the melanin pigment biosynthetic pathway; the hydroxylation of tyrosine to L-3,4-dihydroxy-phenylalanine (dopa), and the subsequent oxidation of dopa to dopaquinone. Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Staining of melanomas with this MAb shows tyrosinase in melanotic as well as amelanotic variants. This MAb is a useful marker for melanocytes and melanomas.

Fibulin-7 is a 439 amino acid extracellular matrix protein that belongs to the Fibulin family. Containing two EGF-like domains and one sushi (CCP/SCR) domain, Fibulin-7 exists as four alternatively spliced isoforms. Fibulin-7 is considered an adhesion protein that interacts with extracellular matrix molecules in developing teeth, and may be involved in differentiation and maintenance of odontoblasts as well as in dentin formation. Fibulin-7 is post-translationally glycosylated with N-linked oligosaccharides and interacts with heparin, fibronectin, fibulin-1 and DSP (dentin sialophosphoprotein). Fibulin-7 is encoded by a gene located on human chromosome 2, which consists of 237 million bases, encodes over 1,400 genes and makes up approximately 8% of the human genome.
