The coupling of ubiquitin conjugation to endoplasmic reticulum (ER) degradation (CUE) domain functions as a ubiquitin (UB) binding domain that is approximately 40 amino acids in length. Present in eukaryotic proteins that are involved in ubiquitination and protein trafficking pathways, CUE domains can bind monoubiquitin and may be required for ubiquitination of the proteins in which they are found. CUEDC1 (CUE domain-containing protein 1) is a 386 amino acid protein that contains one CUE domain, suggesting a possible role in protein trafficking and degradation pathways. Defects in the gene encoding CUEDC1 may be associated with early stage cervical cancer, implicating CUEDC1 as a potential tumor marker. Two isoforms of CUEDC1 exist due to alternative splicing events.

It recognizes human Ig kappa light chains of both secreted and cell surface immunoglobulin. It detects also free kappa light chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.

ZBED1 is a 694 amino acid protein that localizes specifically to granular structures within the nucleus. Expressed ubiquitously at low levels and present at higher levels in heart, placenta, spleen and skeletal muscle, ZBED1 is thought to function as a transcription factor that regulates a number of ribosomal protein (RP) encoding genes, thereby playing a role in the cell cycle and in cell proliferation events. ZBED1 contains one BED-type zinc finger and binds specifically to 5'-TGTCG[CT]GA[CT]A-3' DNA regions found in RP promotors. Additionally, ZBED1 binds strongly to the promotor region of Histone H1 (a protein required for the condensation of nucleosomes into higher order structures), subsequently activating H1 transcription.

Copine 6 is a 557 amino acid member of the copine family of evolutionarily conserved, soluble, calcium-dependent, membrane-binding proteins. Members of the copine family are involved in signal transduction and membrane trafficking. Arabidopsis thaliana mutants lacking copine proteins exhibit reduced cell number and smaller cell size, effects which may be due to a defect in vesicle fusion or transport. Copine 6 contains two N-terminal C2 domains and one C-terminal VWFA (von Willebrand factor A) domain, which is also referred to as the A domain or the core domain. As is characteristic of the copine family, copine 6 functions in membrane trafficking and is capable of binding phospholipids in a calcium-dependent manner. Copine 6 may also play a role in synaptic plasticity.

This gene is a proto-oncogene that encodes a RING finger E3 ubiquitin ligase. The encoded protein is one of the enzymes required for targeting substrates for degradation by the proteasome. This protein mediates the transfer of ubiquitin from ubiquitin conjugating enzymes (E2) to specific substrates. This protein also contains an N-terminal phosphotyrosine binding domain that allows it to interact with numerous tyrosine-phosphorylated substrates and target them for proteasome degradation. Adducins are a family of cytoskeleton proteins encoded by three genes (alpha, beta, gamma). Adducin is a protein associated with the inner leaflet of the plasma membrane and is one of the proteins localised at the spectrin-actin junction of the membrane skeleton. The cortical actin cytoskeletal network is lost during apoptosis and Adducins are central in the cortical actin network organisation. Adducin Alpha is a cytoskeletal protein involved with sodium-pump activity in the renal tubule and is associated with hypertension. The expression of Adducin Alpha and Adducin Gamma is ubiquitous in contrast to the restricted expression of Adducin Beta. Adducin Beta is expressed at high levels in brain and hematopoietic tissues, such as bone marrow, in humans, and in spleen in mice.

Treacher Collins syndrome is also chromosome 5 associated and is caused by insertions or deletions within the TCOF1 gene. Deletion of the p arm of chromosome 5 leads to Cri du chat syndrome. Deletion of 5q or chromosome 5 altogether is common in therapy-related acute myelogenous leukemias and myelodysplastic syndrome.
