FLIP Antibody: Apoptosis is related to many diseases and induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain (DD)-containing adapter molecules and members of the ICE/CED-3 protease family. Caspases-8 (FLICE) and -10 (FLICE2) are two pivotal members in the ICE/CED-3 protease family. FLICE-inhibitory proteins were identified in virus and human and designated v-FLIPs and c-FLIPs, respectively. The human FLIPs were also cloned by several labs independently and termed Casper, I-FLICE, FLAME-1, CASH, CLARP and usurpin. FLIP contains two death effector domains (DEDs) and a caspase-like domain. FLIP interacts with adapter protein FADD and caspase-8 and 10, and potently inhibits apoptosis induced by all known death receptors CD95, DR3, TRAIL-R and TNFR1. Four splice variants of c-FLIPs have been identified and termed FLIPalpha, beta, gamma, and delta, respectively.

The modification of proteins with ubiquitin is an important cellular mechanism for targeting abnormal or short-lived proteins for degradation. Ubiquitination involves at least three classes of enzymes: ubiquitin-activating enzymes, or E1s, ubiquitin-conjugating enzymes, or E2s, and ubiquitin-protein ligases, or E3s. UBE2D1 is a member of the E2 ubiquitin-conjugating enzyme family. This enzyme is closely related to a stimulator of iron transport (SFT), and is up-regulated in hereditary hemochromatosis. It also functions in the ubiquitination of the tumor-suppressor protein p53 and the hypoxia-inducible transcription factor HIF1alpha by interacting with the E1 ubiquitin-activating enzyme and the E3 ubiquitin-protein ligases.

LZTS2 Antibody: The leucine zipper putative tumor suppressor 2 (LZTS2, also known as LAPSER1), a member of the LZTS family, is a negative regulator of the Wnt signaling pathway. It represses beta-catenin-mediated transcriptional activation by promoting the nuclear exclusion of beta-catenin. LZTS2 is involved in cytokinesis and regulates katanin-mediated microtubule severing and release from the centrosome. LZTS2 may negatively regulate axonal outgrowth by preventing the formation of microtubule bundles that are necessary for transport within the elongating axon. LZTS2 is highly expressed in testis and prostate, but it is deleted in multiple cancers, including prostate tumors, suggesting that LZTS2 acts as a potential tumor suppressor.

DARC Antibody: DARC, also known as the Duffy antigen/chemokine receptor, is a seven-transmembrane protein homologous to the classical chemokine G-protein coupled receptors (GPCRs) with the exception of the motif required for G protein coupling. DARC can bind with high affinity several chemokines without transducing any signal, suggesting it may modulate the signals normally induced by these chemokines. Recently, DARC was found to interact with KAI1, a four transmembrane protein recently identified as a tumor metastasis suppressor protein. It is thought that tumor cells dislodged from the primary tumor and expressing KAI1 interact with DARC proteins expressed on vascular cells, transmitting a senescent signal to the tumor cells, while tumor cells that have lost KAI1 expression can proliferate and potentially give rise to metastases. At least three isoforms of DARC are known to exist.

DRAM Antibody: Damage-regulated autophagy modulator (DRAM) is a p53 target gene encoding a lysosomal protein that induces autophagy, a process that degrades cytosolic proteins and organelles. It has been suggested that activation of DRAM by p53 is simultaneous to the activation by p53 of one or more proapoptotic genes such as PUMA, Bax, etc., and that the signaling pathways regulated by these genes together promote a full cell death response. By itself, DRAM cannot induce apoptosis, but the fact that it is inactivated in certain cancers highlights the importance of DRAM and suggests that autophagy may play a more important role in cancer than initially suspected. At least two different isoforms of DRAM are known to exist.
