CCNT1 belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance through the cell cycle. Cyclins function as regulators of CDK kinases. Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event. This cyclin tightly associates with CDK9 kinase, and was found to be a major subunit of the transcription elongation factor p-TEFb. The kinase complex containing this cyclin and the elongation factor can interact with, and act as a cofactor of human immunodeficiency virus type 1 (HIV-1) Tat protein, and was shown to be both necessary and sufficient for full activation of viral transcription. THEM4 Antibody: THEM4, also known as CTMP, binds specifically to the carboxy-terminal regulatory domain of PKB/Akt at the plasma membrane and acts as a negative regulator, reversing the phenotype of v-Akt-transformed cells. Hypermethylation of the THEM4 promoter and transcriptional downregulation of the gene has been reported in multiple glioblastomas, suggesting that epigenetic regulation of THEM4 may play a role in the progression of this cancer. Bioinformatic analysis, confirmed by in vitro testing, indicates that THEM4 is a broad-range, high activity acyl-CoA thioesterase. Recent reports have also indicated that TMEM4 is a mitochondrial protein whose overexpression is associated with an increase in mitochondrial membrane depolarization and caspase-3 and PARP cleavage, suggesting that THEM4 is involved in the apoptotic program.

UCMA Antibody: UCMA is a secreted cartilage-specific protein that was discovered in a screen for differentially expressed genes in retinoic acid-treated mouse chondrocytes. It was also identified in a human chondrocyte EST screen for candidate genes of skeletal dysplasias. UCMA expression is thought to parallel that of collagen II with its expression decreasing with maturation chrondrocytes mature. UCMA is processed by a furin-like protease into two fragments, an amino-terminal fragment and a carboxy-terminal fragment (UCMA-C). Application of recombinant UCMA-C to primary osteoblasts, mesenchymal stem cells, and MC3T3-E1 pre-osteoblasts interferes with their osteogenic differentiation, but does not affect expression of chondrocyte-specific genes or chondrocyte proliferation, suggesting that UCMA may be involved in the negative control of osteogenic differentiation of osteochondrogenic precursor cells. At least two isoforms of UCMA are known to exist.

BACE Antibody: Accumulation of the amyloid-beta (Abeta) plaque in the cerebral cortex is a critical event in the pathogenesis of Alzheimer's disease. Abeta peptide is generated by proteolytic cleavage of the beta-amyloid protein precursor (APP) at beta- and gamma-sites by two proteases. APP is first cleaved by beta-secretase, producing a soluble derivative of the protein and a membrane anchored 99-amino acid carboxy-terminal fragment (C99). The C99 fragment serves as substrate for gamma-secretase to generate the 4 kDa amyloid-beta peptide, which is deposited in the brains of all suffers of Alzheimer's disease. The long-sought beta-secretase was recently identified by several groups independently and designated beta-site APP cleaving enzyme (BACE) and aspartyl protease 2 (Asp2). BACE/Asp2 is a novel transmembrane aspartic protease and colocalizes with APP.

RKHD3 Antibody: Rkhd3, also known as MEX3B is a member of a novel family of four homologous human MEX3 proteins each containing two heterogeneous nuclear ribonucleoprotein K homology (KH) domains and one carboxy-terminal RING finger module. MEX3 proteins, including Rkhd3, are phosphoproteins that bind RNA through their KH domains and shuttle between the nucleus and the cytoplasm via the CRM1 export pathway. These proteins are a novel family of evolutionarily conserved RNA-binding proteins, differentially recruited to P bodies and potentially involved in post-transcriptional regulatory mechanisms. Rkhd3 is thought to act as a mechanism to fine-tune mRNA regulation in early Xenopus development, and with Rkhd4, but not Rkhd1, will co-localize with both the hDcp1a decapping factor and Argonaute (Ago) proteins in processing bodies (P bodies), recently characterized as centers of mRNA turnover.

Rift Valley Fever Virus Antibody: Rift Valley Fever (RFV) virus is an arthropod-borne virus endemic to Africa that infects humans and animals that is transmitted predominantly by mosquitoes. During human infections, symptoms can range from benign fever to severe encephalitis and fatal hepatitis with hemorrhagic fever. The Bunyaviridae family of viruses to which the RVF virus belongs are spherical enveloped viruses with a tripartite RNA genome of negative or ambisense polarity. The three segments are referred to as the L, M, and S segments. The L and M segments are negative polarity and code fore the L-dependent RNA polymerase and glycoprotein precursor respectively. The S segment is of ambisense polarity and encodes the nucleoprotein and non-structural proteins. This RVF virus antibody was derived from a peptide sequence near the carboxy terminus of the polyprotein precursor translated from the M segment. It will therefore detect both the precursor and the Glycoprotein G2.

TRIM23 is a member of the tripartite motif (TRIM) family. The TRIM motif includes three zinc-binding domains, a RING, a B-box type 1 and a B-box type 2, and a coiled-coil region. This protein is also a member of the ADP ribosylation factor family of guanine nucleotide-binding family of proteins. Its carboxy terminus contains an ADP-ribosylation factor domain and a guanine nucleotide binding site, while the amino terminus contains a GTPase activating protein domain which acts on the guanine nucleotide binding site. The protein localizes to lysosomes and the Golgi apparatus. It plays a role in the formation of intracellular transport vesicles, their movement from one compartment to another, and phopholipase D activation. Three alternatively spliced transcript variants for this gene have been described.

Aak1 Antibody: AP2-associated protein kinase 1 (Aak1) is a member of the Ark1/Prk1 subfamily of Ser/Thr protein kinases that are thought to regulate endocytosis by phosphorylating the accessory endocytic components. Aak1 interacts with and phosphorylates the mu2 subunit of the AP-2 complex, which promotes binding of the AP-2 to tyrosine based (Yxxphi) internalization motif-containing receptors and subsequent receptor endocytosis. At least two isoforms of Aak1 are known to exist; the longer isoform contains an extended carboxy-terminus that contains an additional clathrin-binding domain. Overexpression of this long isoform or Aak1 depletion by RNA interference impairs transferrin recycling from the early/sorting endosome, suggesting that Aak1 functions at multiple steps of the endosomal pathway by regulating transferrin internalization and its recycling back to the plasma membrane.
