The protein encoded by this gene is a member of the Ser/Thr protein kinase family. This protein kinase is highly similar to the gene products of S. cerevisiae cdc28, and S. pombe cdc2. It is a catalytic subunit of the cyclin-dependent protein kinase complex, whose activity is restricted to the G1-S phase, and essential for cell cycle G1/S phase transition. This protein associates with and regulated by the regulatory subunits of the complex including cyclin A or E, CDK inhibitor p21Cip1 (CDKN1A) and p27Kip1 (CDKN1B). Its activity is also regulated by its protein phosphorylation. Two alternatively spliced variants and multiple transcription initiation sites of this gene have been reported.

This gene encodes an enzyme belonging to the protein tyrosine kinase family, and it plays a role in T-cell development and lymphocyte activation. This enzyme, which is phosphorylated on tyrosine residues upon T-cell antigen receptor (TCR) stimulation, functions in the initial step of TCR-mediated signal transduction in combination with the Src family kinases, Lck and Fyn. This enzyme is also essential for thymocyte development. Mutations in this gene cause selective T-cell defect, a severe combined immunodeficiency disease characterized by a selective absence of CD8-positive T-cells. UGT1A9 is a UDP glucuronosyltransferase, an enzyme of the glucuronidation pathway that transforms small lipophilic molecules, such as steroids, bilirubin, hormones, and drugs, into water soluble, excretable metabolites. This gene is part of a complex locus that encodes several UDP glucuronosyltransferases. The locus includes thirteen unique alternate first exons followed by four common exons. Four of the alternate first exons are considered pseudogenes. Each of the remaining nine 5' exons may be spliced to the four common exons, resulting in nine proteins with different N termini and identical C termini. Each first exon encodes the substrate binding site, and is regulated by its own promoter. UGT1A9 is active on phenols.

The Src family of protein tyrosine kinases act in signal transduction pathways. In Src-transformed fibroblasts and in normal cells treated with certain growth factors fish is tyrosine-phosphorylated. Treatment of cells with cytochalasin D results in rapid tyrosine phosphorylation of fish, along with activation of Src. Fish is likely to be involved in tyrosine kinase Signalling and may have a role in cytoskeletal changes.

Activation of TAL1 characterizes up to 60% of cases of human T cell acute lymphoblastic leukemia, making it the most frequent gain-of-function mutation observed in this disorder. TAL1 (also designated SCL) is a serine phosphoprotein and basic helix-loop-helix transcription factor known to regulate embryonic hematopoiesis. This transcription factor binds as a heterodimer with E2A and HEB/HTF4 to a nucleotide sequence motif termed the E-box. A multi-pass membrane protein, GDE3 localizes to cell membrane and cytoplasm, and colocalizes with actin in the cytoskeleton. GDE3 contains one GDPD domain, binds calcium as a cofactor and is encoded by a gene mapping to human chromosome Xq13.1.

The encoded protein and decorin are thought to be the result of a gene duplication. Decorin contains one attached glycosaminoglycan chain, while this protein probably contains two chains. For this reason, this protein is called biglycan. This protein plays a role in assembly of collagen fibrils and muscle regeneration. TAL1 disruption at 1p32, a common rearrangement in the T-cell acute lymphoblastic leukemia, usually results in the formation of a SCL interrupting locus (SIL)-TAL1 fusion product. SIL is an immediate early gene whose expression is associated with cell proliferation. The Sil protein exhibits ubiquitous expression in hematopoietic cell lines and tissues. However, Sil protein levels remain tightly regulated during the cell cycle, achieving peak levels in mitosis and diminishing on transition to G1 phase. Overexpression of Sil in primary adenocarcinomas predicts metastatic spread, especially in lung tumors with increased mitotic activity.
