Enteroviruses, such as enterovirus 71, are classified to be in the picornavirus family, pico [small] + RNA [ribonucleic acid] + virus. Picornaviruses are among the smallest and simplest ribonucleic acid containing viruses known (1). The RNA for many enteroviruses have now been cloned and complete genomic sequences have been obtained. The RNA from all sequenced enteroviruses are similar in length, about 7400 nucleotides, and have identical organization (1). The human alimentary tract is the predominant site of enterovirus replication and these viruses were first isolated from enteric specimens. These viruses are the cause of paralytic poliomyelitis, aseptic meningitis-encephalitis, myocarditis, pleurodynia, hand-foot-and-mouth disease, conjunctivitis, and numerous other syndromes associated with extra-intestinal target organs. There are 67 numbered types of enteroviruses in the enterovirus family (1): three polioviruses, twenty-three coxsackieviruses A, six coxsackieviruses B, thirty-one echoviruses, and four other enteroviruses.

HLA-A belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is anchored in the membrane. Class I molecules play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. They are expressed in nearly all cells. The heavy chain is approximately 45 kDa and its gene contains 8 exons. Exon 1 encodes the leader peptide, exons 2 and 3 encode the alpha1 and alpha2 domains, which both bind the peptide, exon 4 encodes the alpha3 domain, exon 5 encodes the transmembrane region, and exons 6 and 7 encode the cytoplasmic tail. Polymorphisms within exon 2 and exon 3 are responsible for the peptide binding specificity of each class one molecule. In Drosophila, neuronal cell fate decisions are directed by NUMB, a Signalling adapter protein with two protein-protein interaction domains, namely a phosphotyrosine-binding domain and a proline-rich SH3-binding region (PRR). The mammalian NUMB homolog plays a role in the determination of cell fate during development and binds with a variety of proteins, including Eps15, LNX1 and Notch 1. NumbL (NUMB-like protein), also known as Numb-R, NBL, CAG3A, CTG3a, NUMBLIKE or TNRC23, is a 609 amino acid cytoplasmic protein that, like NUMB, is thought to play a role in cell fate. Expressed at high levels in developing brain tissue, NumbL contains one PID (phosphotyrosine interaction domain) and plays an important role in neuronal differentiation, possibly associating with Eps15 and Notch 1. In mice, deletion of the NumbL gene is associated with early embryonic death, suggesting an essential role for NumbL in early development.

Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDHB12 (Protocadherin beta 12) is a 795 amino acid protein that is one of 16 proteins in the protocadherin beta cluster. Unlike the alpha and gamma gene clusters whose genes are spliced to downstream constant region exons during transcription, members of the beta cluster (such as PCDHB12) do not use constant-region exons to produce mRNAs. As a result, each protocadherin beta gene encodes the transmembrane, extracellular and short cytoplasmic domains of the protein. Localized to the cell membrane, PCDHB12 is a single-pass type I membrane protein that contains six cadherin domains. PCDHB12 may participate in the establishment and maintenance of specific neuronal connections in the brain.

Cell cycle progression is subject to arrest at G1 and G2 checkpoints in response to DNA damage, presumably to allow time for DNA repair prior to entry into S and M phase, respectively. PEG-3 (progression elevated gene-3) shares significant homology with GADD 34 and is inducible by DNA damage. An additional GADD related gene, PA26, is a possible target of p53. Three isoforms of PA26 have been identified as PA26-T1, PA26-T2 and PA26-T3.

The SH3TC2 protein is expressed in adult heart, testis, spinal cord, and brain as well as in fetal brain and liver. Mild mononeuropathy of the median nerve (MNMN) is caused by heterozygous mutation in the SH3TC2 gene. Also, Charcot-Marie-Tooth disease type 4C (CMT4C) is a more severe neuropathy caused by homozygous or compound heterozygous mutation in the SH3TC2 gene. Existing as four alternatively spliced isoforms and containing 18 exons, the SH3TC2 gene is conserved in chimpanzee, dog, cow, mouse, rat, chicken and zebrafish, and maps to human chromosome 5q32.

PDZK4, also known as PDZRN4L (PDZ domain-containing RING finger protein 4-like protein) or LU1, is a 769 amino acid coiled-coil protein that contains one PDZ (DHR) domain. Encoded by a gene that maps to human chromosome Xq28, PDZK4 is conserved in dog, cow, mouse, rat and zebrafish. PDZK4 localizes to cytoplasm and is expressed specifically in adult and fetal brain. PDZK4 functions as an oncogene and is up-regulated in synovial carcinomas. Treatment of synovial sarcoma cells with small interfering RNA (siRNA) inhibits PDZK4 expression, resulting in tumor-cell growth suppression, suggesting that inappropriate expression of PDZK4 may play a role synovial sarcoma cell proliferation.The exact function of FRMPD2 is unknown. The protein contains a FERM domain: such structures are often involved in signal transduction pathways. Alternatively spliced transcript variants encoding different isoforms have been identified.

ANKMY1 (ankyrin repeat and MYND domain containing 1), also known as ZMYND13 or TSAL1, is a 941 amino acid protein that contains seven ANK repeats, three MORN repeats and one MYND-type zinc finger. MORN repeats were first identified in junctophilins, cytoplasmic proteins involved in junctions between the plasma membrane and the ER/SR membrane. The presence of MORN repeats suggests that ANKMY1 may interact with the plasma membrane. The MYND domain consists of a cluster of cysteine and histidine residues, arranged with an invariant spacing to form a potential zinc-binding motif which may be involved in protein-protein interactions. Three isoforms of ANKMY1 exists due to alternative splicing events.

ODF3B, also known as ODF3L3 (outer dense fiber protein 3-like protein 3), is a 253 amino acid protein belonging to the ODF3 family. Existing as two isoforms produced by alternative splicing, ODF3B contains one DUF1309 repeat. The gene that encodes ODF3B maps to human chromosome 22, which contains over 500 genes and about 49 million bases. Being the second smallest human chromosome, 22 contains a surprising variety of interesting genes. Phelan-McDermid syndrome, Neurofibromatosis type 2 and autism are associated with chromosome 22. A schizophrenia susceptibility locus has been identified on chromosome 22 and studies show that 22q11 deletion symptoms include a high incidence of schizophrenia. Translocations between chromosomes 9 and 22 may lead to the formation of the Philadelphia Chromosome and the subsequent production of the novel fusion protein, BCR-Abl, a potent cell proliferation activator found in several types of leukemia.

The mammalian homologues of the key cell death gene CED 4 in C. elegans has been identified recently from human and mouse and designated Apaf1 (for apoptosis protease activating factor 1). Apaf1 binds to cytochrome c (Apaf2) and caspase 9 (Apaf3), which leads to caspase 9 activation. Activated caspase 9 in turn cleaves and activates caspase 3 that is one of the key proteases, being responsible for the proteolytic cleavage of many key proteins in apoptosis. A new Apaf1 Interacting Protein (APIP) also known as CG129 and MMRP19, has been identified as a negative regulator of ischemic injury. APIP competes with Caspase 9 binding site of Apaf1. APIP is predicted to code for a 204 amino acid. An isoform of APIP, APIP2 encodes a 242 amino acid protein, which is an alternative splicing variant differing in its N terminus from APIP. APIP transcript is ubiquitously expressed in most adult tissue with high expression in skeletal muscle, heart, and kidney.
