This gene encodes a cell surface tyrosine kinase receptor for members of the platelet-derived growth factor family. These growth factors are mitogens for cells of mesenchymal origin. The identity of the growth factor bound to a receptor monomer determines whether the functional receptor is a homodimer or a heterodimer, composed of both platelet-derived growth factor receptor alpha and beta polypeptides. This gene is flanked on chromosome 5 by the genes for granulocyte-macrophage colony-stimulating factor and macrophage-colony stimulating factor receptor; all three genes may be implicated in the 5-q syndrome. A translocation between chromosomes 5 and 12, that fuses this gene to that of the translocation, ETV6, leukemia gene, results in chronic myeloproliferative disorder with eosinophilia. FGFR1OP2 belongs to the SIKE family. The FGFR1OP2 (FGFR1 oncogene partner 2) gene was identified through its involvement in a fusion with the FGFR1 gene. FGFR1OP2 may be involved in the wound healing pathway. It is expressed in bone marrow, spleen and thymus. A chromosomal aberration involving FGFR1OP2 may be a cause of stem cell myeloproliferative disorder (MPD). Insertion ins(12;8)(p11;p11p22) with FGFR1. MPD is characterised by myeloid hyperplasia, eosinophilia and T cell or B cell lymphoblastic lymphoma. In general it progresses to acute myeloid leukemia. The fusion protein FGFR1OP2-FGFR1 may exhibit constitutive kinase activity and be responsible for the transforming activity.

Prickle1 is an 831 amino acid protein that contains one PET domain and three LIM zinc-binding domains and localizes to the cytoplasm, as well as to the nuclear membrane. Expressed at higher levels in placenta and at lower levels in liver, brain, kidney, lung and pancreas, Prickle1 is thought to function as a nuclear receptor that interacts with NRSF, a silencer protein that binds the DNA sequence element NRSE (neuron-restrictive silencer element). Defects in the gene encoding Prickle1 are associated with autosomal recessive progressive myoclonic epilepsy-1B, which is characterized by quick jerks of the arms, shoulders or legs. The gene encoding Prickle1 maps to human chromosome 12, which encodes over 1,100 genes and comprises approximately 4.5% of the human genome.
