Beschreibung:   GPAT1 Antibody: Glycerol-3-phosphate acyltransferase 1 (GPAT1), one of four known GPAT isoforms, is located on the mitochondrial outer membrane, allowing reciprocal regulation with carnitine palmitoyltransferase-1. It is thought to be critical for the development of hepatic steatosis; steatosis triggered by GPAT1 overexpression leads to hepatic and possibly peripheral insulin resistance. GPAT1 is transcriptionally upregulated by insulin and sterol regulatory element binding protein (SREBP-1) and downregulated by AMP-activated protein kinase. Mice deficient in GPAT1 exhibit decreased triacylglycerol (TAG) in cardiomyocytes even in high-fat diets, suggesting that GPAT1 contributes significantly to TAG accumulation in heart tissue during lipogenic or high fat diets. Beschreibung:   LKB1 Antibody: The LKB1 serine/threonine protein kinase was initially identified as a tumor suppressor gene mutated in human Peutz-Jeghers syndrome (PJS), a condition resulting in the growth of numerous intestinal polyps classed as hamartomas. LKB1 exists as a heterotrimeric complex with two other proteins, Ste20-related adaptor protein (STRAD) and MO25. Together, this complex can phsophorylate and activate the AMP-activate protein kinase (AMPK). Following AMPK activation by LKB1, AMPK then phosphorylates TSC1 and TSC2, key components of the metabolism-regulating TOR signaling pathway, which antagonizes the activation for the TOR pathway. The principal biologically active forms of pantothenic acid are coenzyme A (CoA) and acyl carrier protein (ACP). In CoA, the business center of the molecule is the pantothenic acid metabolite 4'-phosphopantetheine. Coenzyme A is comprised of 4'-phosphopantetheine linked by an anhydride bond to the nucloetide adenosine 5'-monophosphate. 4'-Phosphopantetheine itself is comprised of pantothenic acid linked at one end, via an amide bond, to beta-mercaptoethylamine, derived from L-cysteine, and at the other end to a phosphate group. The sulfhydryl group of 4'-phosphopantetheine, which is the business end of the coenzyme, forms thioesters with acyl groups producing acyl-CoA derivatives, including acetyl-CoA.
Coenzyme A facilitates removal of lipid peroxides by increasing mobilization of fatty acids, and promote repair of plasma membranes by activating phospholipid synthesis.

Beschreibung:   BRSK2 Antibody: BRSK2 was initially identified through a computer screen of the human genome and shows significant homology to the C. elegans neuronal cell polarity regulator SAD1. BRSK2 is expressed in the brain and to a lesser extent in the testes. BRSK2 is a member of the AMP-activated protein kinase subfamily and can be activated by the tumor suppressor kinase LKB1. More recently, it has been shown that both BRSK2 and the related protein BRSK1 are required for mammalian neuronal polarization. While BRSK1- and BRSK2-null mice were viable, double-mutant mice died within two hours of birth. Neurons from these mice showed uniformly-sized neurites as opposed to the normal long axon and multiple shorter dendrites. These neurites also displayed both axonal and dendritic markers. BRSK2 has also been shown to be an autoantigen in paraneoplastic limbic encephalitis. At least four isoforms of BRSK2 are known to exist.

The POU family is so named because the first 3 members identified were PIT1 and OCT1 of mammals, and Unc-86 of C. elegans. PIT1 contains 2 protein domains, termed POU-specific and POU-homeo, which are both necessary for high affinity DNA binding on genes encoding growth hormone and prolactin. PIT1 is also important for regulation of the genes encoding prolactin and thyroid-stimulating hormone beta subunit by thyrotropin-releasing hormone and cyclic AMP. This gene encodes a member of the POU family of transcription factors that regulate mammalian development. The protein regulates expression of several genes involved in pituitary development and hormone expression. Mutations in this genes result in combined pituitary hormone deficiency. Multiple transcript variants encoding different isoforms have been found for this gene.
