Alpha-1 Antitrypsin or alpha1-antitrypsin (A1AT) is a protease inhibitor belonging to the serpin superfamily. It is generally known as serum trypsin inhibitor. Alpha 1-antitrypsin is also referred to as alpha-1 proteinase inhibitor (A1PI) because it inhibits a wide variety of proteases. It protects tissues from enzymes of inflammatory cells, especially neutrophil elastase, and has a reference range in blood of 1.5 - 3.5 gram/liter (in US the reference range is generally expressed as mg/dL or micromoles), but the concentration can rise manyfold upon acute inflammation. In its absence (such as in alpha 1-antitrypsin deficiency), neutrophil elastase is free to break down elastin, which contributes to the elasticity of the lungs, resulting in respiratory complications such as emphysema, or COPD (chronic obstructive pulmonary disease) in adults and cirrhosis in adults or children.<p>Like all serine protease inhibitors, A1AT has a characteristic secondary structure of beta sheets and alpha helices. Mutations in these areas can lead to non-functional proteins that can polymerise and accumulate in the liver (infantile hepatic cirrhosis). [Wiki].

It is a growth factor for animals as well as microorganisms. Myo-Inositol is the most abundant form of polyols that serves as a structural element of secondary messengers in eukaryotic cells. Inositol or cyclohexane-1,2,3,4,5,6-hexol is a chemical compound, a sixfold alcohol (polyol) of cyclohexane. It exists in nine possible stereoisomers, of which the most prominent form, widely occurring in nature, is cis-1,2,3,5-trans-4,6-cyclohexanehexol, or myo-inositol (former name meso-inositol). Inositol is a carbohydrate, though not a classical sugar. It has a taste which has been assayed at half the sweetness of table sugar. Used as a lipotropic agent. A component of membrane phospholipids, glycosylphosphatidylinositol anchors that bind glycoproteins to cell membranes, and inositol phosphate second messengers. Tweens® are a series of nonionic surfactants derived from sorbitan esters. They are soluble or dispersible in water but differ widely in organic and oil solubilities. Used as oil-in-water emulsifiers in pharmaceuticals, cosmetics, cleaning compounds, etc.
Tween® 80 has been widely used in biochemical applications including: solubilizing proteins, isolating nuclei from cells in culture selective protein extraction growing of tubercule bacilli, and emulsifying and dispersing substances in medicinal and food products. It has little or no activity as an anti-bacterial agent. It has been shown to have an adverse effect on the antibacterial effect of methyl paraben and related compounds. Non-ionic detergent used for selective protein extraction and isolation of nuclei from mammalian cell lines. Soluble/miscible in water to give a clear yellow solution; miscible with alcohol, dioxane, and ethyl acetate; practically insoluble in liquid paraffin and fixed oils (such as mineral oil). Autoclaving of solutions is not recommended. Sterile filtering is suggested with a 0.22 micron filter. Tween® may need to be warmed to about 40 °C and alternated with portions of hot distilled water while being poured through the filter. Store at Room Temperature (15-30 °C).

Antibodies are produced by B lymphocytes, each expressing only one class of light chain. Once set, light chain class remains fixed for the life of the B lymphocyte. In a healthy individual, the total kappa to lambda ratio is roughly 3:1 in serum (measuring intact whole antibodies) or 1:1.5 if measuring free light chains, with a highly divergent ratio indicative of neoplasm.<p>Individual B-cells in lymphoid tissue possess either kappa or lambda light chains, but never both together. Specific rearrangement of lambda light chain of immunoglobulins can lead to loss of some protein coding genes, which does not seem to be functionally relevant (while functionally relevant miR-650 can be overexpressed). Using immunohistochemistry, it is possible to determine the relative abundance of B-cells expressing kappa and lambda light chains. If the lymph node or similar tissue is reactive, or otherwise benign, it should possess a mixture of kappa positive and lambda positive cells. If, however, one type of light chain is significantly more common than the other, the cells are likely all derived from a small clonal population, which may indicate a malignant condition, such as B-cell lymphoma. Glycine is a non-essential amino acid. It is only amino acid with no asymmetric carbon and thus is not chiral. It is the major inhibitory neurotransmitter. It is involved in the biosynthesis of the porphyrin rings of hemes and chlorophylls. Glycine is commonly used in buffer solutions, in electrophoresis and preparative chromatography. A study of the folding of monoclonal antibodies in the presence of glycine and their subsequent purification has been published. The use of glycine in the purification of lipopolysaccharides, lipooligosaccharides, and lipid A has been reported. It is an amino acid for use in cell culture media development applications and existing media formulations. Glycine is commonly used as a component in Tris-glycine and Tris-glycine-SDS running buffers for polyacrylamide gel electrophoresis, a component of Towbin's transfer buffer for Western blots, a buffer substance in cryoenzymology, in osmotic pressure maintenance in isoelectric focusing of erythrocytes, salting-in effect in protein chemistry, and as a buffer component in the coupled phosphatase-kinase reaction for end labelling of restriction fragments. The growth requirements of various microorganisms is reported in the Handbook of Microbiology. Pst1 Antibody: Protoplast secreted protein 1 (Pst1) is an anchored plasma membrane protein. Pst1 was previously identified as a protein secreted by yeast regenerating protoplasts, which suggests a role in cell wall construction. Pst1 is a protein with 444 amino acids and is attached to yeast cell wall via a glycosylphosphatidylinisotol (GPI) anchor. Pst1 contains 15 potential N-linked glycosylation sites and is heavily glycosylated. It migrates at approximately 200 kDa on SDS-PAGE when produced in wild type S. cerevisiae. Our Pst1 protein (Catalog No. 95-151) was expressed in a genetically manipulated triple-mutant (TM) S. cerevisiae stain ( Delta och1 Delta mnn1 Delta mnn4), which results in the production of sole Man8GlcNAc2 carbohydrate structures and Pst1 migration at approximately 100 kDa. When produced in the TM yeast, Pst1 can be recognized by several glycan-specific HIV-1 broadly neutralizing antibodies, including 2G12 and recently identified PGT antibodies. Among several heavily N-glycosylated TM yeast glycoproteins, Pst1 shows higher affinity for 2G12 and efficiently inhibits gp120 interactions with 2G12 and DC-SIGN, and it also blocks 2G12-mediated neutralization of HIV-1 pseudoviruses.

Das E.Z.N.A.® MicroElute Total RNA Kit bietet eine schnelle und einfache Methode zur Isolierung von bis zu 50 µg Gesamt-RNA aus kleinen Mengen an kultivierten eukaryotischen Zellen, Geweben wie lasergeschnittenen Proben (LDS) oder Feinnadelaspiraten (FNA). Normalerweise können bis zu 5×10⁵ eukaryotische Zellen oder 5 mg Gewebe in einem einzigen Experiment verwendet werden, je nach Art des Gewebes. Dieses Kit ermöglicht die Verarbeitung von einer oder mehreren Proben in weniger als 30 Minuten. Es sind keine Phenol-/Chloroformextraktionen und zeitaufwändige Arbeitsschritte wie die CsCl-Gradienten-Ultrazentrifugation erforderlich, und auch die Fällung mit Isopropanol oder LiCl entfällt. Aufgereinigte RNA kann mit 10 bis 15 µl nukleasefreiem Wasser eluiert werden. ACRV1 is a testis-specific, differentiation antigen, acrosomal vesicle protein 1, that arises within the acrosomal vesicle during spermatogenesis, and is associated with the acrosomal membranes and matrix of mature sperm. The acrosomal vesicle protein 1 may be involved in sperm-zona binding or penetration, and it is a potential contraceptive vaccine immunogen for humans. This gene encodes a testis-specific, differentiation antigen, acrosomal vesicle protein 1, that arises within the acrosomal vesicle during spermatogenesis, and is associated with the acrosomal membranes and matrix of mature sperm. This gene consists of 4 exons and its alternative splicing generates multiple distinct transcripts, which encode protein isoforms ranging from 81 to 265 amino acids. The longest transcript is the most abundant, comprising 53-72% of the total acrosomal vesicle protein 1 messages; the second largest transcript comprises 15-32%; the third and the fourth largest transcripts account for 3.4-8.3% and 8.7-12.5%, respectively; and the remaining transcripts combined account for < 1% of the total acrosomal vesicle protein 1 message. It is suggested that phenomena of cryptic splicing and exon skipping occur within this gene. The acrosomal vesicle protein 1 may be involved in sperm-zona binding or penetration, and it is a potential contraceptive vaccine immunogen for humans.
