Sulfation is an essential conjugation reaction that increases the water solubility of many compounds, thereby influencing their renal excretion and also resulting in the formation of active metabolites. SULT4A1 (sulfotransferase family 4A, member 1), whose alternative names include brain sulfotransferase-like protein, nervous system sulfotransferase, NST, SULTX3, hBR-STL-1, BRSTL1, BR-STL-1, MGC40032 and DJ388M5.3, is a 284 amino acid protein showing cytoplasmic localization. As a member of the sulfotransferase 1 family, SULT4A1 plays a role in elimination of xenobiotics, activation of procarcinogens and regulation of hormones. SULT4A1 is highly expressed in cerebral cortex and frontal lobe, with lower expression in cerebellum, temporal and occipital lobes. Two SULT4A1 isoforms exist to alternative splicing events. The gene encoding SULT4A1 maps to human chromosome 22q13.2, a region which has been implicated in predisposition to schizophrenia.

Saposin-B stimulates the hydrolysis of galacto-cerebroside sulfate by arylsulfatase A (EC 3.1.6.8), GM1 gangliosides by beta-galactosidase (EC 3.2.1.23) and globotriaosylceramide by alpha-galactosidase A (EC 3.2.1.22). Saposin-B forms a solubilizing complex with the substrates of the sphingolipid hydrolases. Saposin-D is a specific sphingomyelin phosphodiesterase activator (EC 3.1.4.12). Prosaposin: Behaves as a myelinotrophic and neurotrophic factor, these effects are mediated by its G-protein-coupled receptors, GPR37 and GPR37L1, undergoing ligand-mediated internalization followed by ERK phosphorylation signaling. Saposins are specific low-molecular mass non-enzymic proteins, they participate in the lysosomal degradation of sphingolipids, which takes place by the sequential action of specific hydrolases.

Small leucine-rich proteoglycans (SLRPs) such as Decorin, Biglycan, Fibromod-ulin, Keratocan, Lumican and Osteoglycin mediate extracellular matrix organization and are binding partners of TGF Beta. The Decorin core protein binds to growth factors, intercellular matrix molecules such as Fibronectin and Throm-bospondin, and to the Decorin endocytosis receptor. Fibromodulin is a collagen-binding keratan sulphate proteoglycan that influences adhesion processes of connective tissue and plays a role in fibrillogenesis by regulating collagen fibril spacing and thickness. Keratocan (KTN) develops corneal transparency and maintains the stromal matrix structure. Keratocan is a secreted protein in the extracellular matrix that binds to keratan sulfate chains. Keratocan is mainly detected in the cornea, but can also be expressed in trachea, intestine, ovary, lung and skeletal muscle. Defects in the gene encoding for Keratocan can cause cornea plana 2 (CNA2), an autosomal recessive disorder where the forward convex curvature of the cornea is flattened.

Cellular receptors for blood proteases regulate chemotaxis, extracellular proteolysis, and growth behavior of normal and malignant cells. Effector cell protease receptor-1 (EPR1) is a receptor for the coagulation protease factor Xa. EPR1 is characterized by a cysteine-rich extracellular module, a single membrane-spanning domain, and a serine-rich cytoplasmic tail featuring at least 15 potential phosphorylation sites. EPR1 also contains 2 N-linked glycosylation sites, 4 O-linked glycosylation sites, and a chondroitin sulfate attachment site, which may provide anchoring for carbohydrate chains, EPR1 transfectants bind to factor Xa in a specific and saturable manner, and in the absence of factor V/Va promote prothrombin activation in a factor Xa concentration-dependent reaction. Activated platelets and megakaryocytes express EPR1. Both EPR1 and membrane-bound factor Va are thought to be required to mediate factor Xa binding to the activated platelet to form a functional prothrombinase complex.
