Collagens are highly conserved throughout evolution and are characterized by an uninterrupted "Glycine-X-Y" triplet repeat that is a necessary part of the triple helical structure. For these reasons, it is often extremely difficult to generate antibodies with specificities to collagens. The development of type specific antibodies is dependent on NON-DENATURED three-dimensional epitopes. Collagens are extensively purified for immunization from human and bovine placenta and cartilage by limited pepsin digestion and selective salt precipitation. This preparation results in a native conformation of the protein. Antibodies are isolated from rabbit antiserum and are extensively cross-adsorbed by immunoaffinity purification to produce 'type' specific antibodies. Greatly diminished reactivity and selectivity of these antibodies will result if denaturing and reducing conditions are used for SDS-PAGE and immunoblotting. This antibody reacts with most mammalian Type IV collagens and has negligible cross-reactivity with type I, II, III, V, or VI collagens.

Transforming growth factor- beta (TGF- beta ) is an essential regulator in the processes of development, cell proliferation, and extracellular matrix deposition. TGF- beta regulates cellular processes by binding to three high-affinity cell surface receptors: TGF- beta receptor type I (TGF- beta -RI), TGF- beta receptor type II (TGF- beta -RII), and TGF- beta beta receptor type III (TGF- beta -RIII). TGF- beta RII is consists of a C-terminal protein kinase domain and an N-terminal ectodomain and belongs to transforming growth factor-beta (TGF- beta ) receptor subfamily. TGF- beta RII has a protein kinase domain which can form a heterodimeric complex with another receptor protein and bind TGF-beta. This receptor/ligand complex phosphorylates protein will enter the nucleus and regulate the transcription of a subset of genes related to cell proliferation.

The autosomal dominant cerebellar ataxias (ADCA) are a heterogeneous group of neurodegenerative disorders characterised by progressive degeneration of the cerebellum, brain stem and spinal cord. Clinically, ADCA has been divided into three groups: ADCA types I-III. ADCAI is genetically heterogeneous, with five genetic loci, designated spinocerebellar ataxia (SCA) 1, 2, 3, 4 and 6, being assigned to five different chromosomes. ADCAII, which always presents with retinal degeneration (SCA7), and ADCAIII often referred to as the `pure' cerebellar syndrome (SCA5), are most likely homogeneous disorders. Several SCA genes have been cloned and shown to contain CAG repeats in their coding regions. ADCA is caused by the expansion of the CAG repeats, producing an elongated polyglutamine tract in the corresponding protein. The expanded repeats are variable in size and unstable, usually increasing in size when transmitted to successive generations. The function of the ataxins is not known. This locus has been mapped to chromosome 6, and it has been determined that the diseased allele contains41-81 CAG repeats, compared to 6-39 in the normal allele, and is associated with spinocerebellar ataxia type 1 (SCA1). At least two transcript variants encoding the same protein have been found for this gene.

TREX1 Antibody: Trex1 is the major human 3' to 5' exonuclease which is required for checkpoint signaling after DNA damage. It is ubiquitously expressed, binds to single stranded DNA coated with replication protein A that accumulates at sites of DNA damage and recruits the ataxia telangiectasia and Rad3 related protein (ATR), a checkpoint kinase, to sites of DNA damage and replication stress. Trex1 is required for ATR expression. This gene uses two different open reading frames. The upstream ORF encodes proteins which interact with ATR and localize to intranuclear foci induced by DNA damage and are essential components of the DNA damage checkpoint. The downstream ORF encodes proteins with 3' to 5' exonuclease activity and may be a subunit of human DNA polymerase III. Multiple transcript variants encoding different isoforms have been found. Mutations in this gene result in Aicardi-Goutieres syndrome, chilblain lupus, and Cree encephalitis.

Fibronectin Leucine Rich Transmembrane Protein 1 (FLRT1) is a member of the Fibronectin Leucine Rich Transmembrane protein (FLRT) family. There are three fibronectin leucine-rich repeat transmembrane (FLRT) proteins: FLRT1, FLRT2 and FLRT3, all contain 10 leucine-rich repeats (LRR), a type III fibronectin (FN) domain, followed by the transmembrane region, and a short cytoplasmic tail. FLRT proteins have dual properties as regulators of cell adhesion and potentiators of fibroblast growth factor (FGF) mediated signalling. The fibronectin domain of all three FLRTs can bind FGF receptors. This binding is thought to regulate FGF signaling during development. The LRR domains are responsible for both the localization of FLRTs in areas of cell contact and homotypic cell association. FLRT1 is expressed at brain compartmental boundaries. FLRT1 is a target for tyrosine phosphorylation mediated by FGFR1 and implicate a non-receptor Src family kinase (SFK).

Human Vascular endothelial growth factor receptor 2(KDR, VEGFR-2) is a member of the class III subfamily of receptor tyrosine kinases (RTKs). KDR is involved in a number of fundamental biological processes such as the regulation of angiogenesis, vascular development, vascular permeability, and embryonic hematopoiesis. It also plays an essential role in promoting proliferation, survival, migration and differentiation of endothelial cells, reorganisation of the actin cytoskeleton. VEGFR2 is identified as the receptor for VEGF and VEGFC and an early marker for endothelial cell progenitors, whose expression is restricted to endothelial cells in vivo.The adaptor protein SHB has been shown to interact with VEGFR2 in receptor tyrosine kinase signalling. In addition, VEGFR2 is able to interact with HIV-1 extracellular Tat protein upon VEGF activation, and seems to enhance angiogenesis in Kaposi's sarcoma lesions. VEGF R2 is thought to be the primary inducer of VEGF-mediated blood vessel growth, while VEGF R3 plays a significant role in VEGF-C and VEGF-D-mediated lymphangiogenesis.

CAPN10 is the calcium-regulated non-lysosomal thiol-protease which catalyzes limited proteolysis of substrates involved in cytoskeletal remodeling and signal transduction.Calpains are ubiquitous, well-conserved family of calcium-dependent, cysteine proteases. The calpain proteins are heterodimers consisting of an invariant small subunit and variable large subunits. The large catalytic subunit has four domains: domain I, the N-terminal regulatory domain that is processed upon calpain activation; domain II, the protease domain; domain III, a linker domain of unknown function; and domain IV, the calmodulin-like calcium-binding domain. This gene encodes a large subunit. It is an atypical calpain in that it lacks the calmodulin-like calcium-binding domain and instead has a divergent C-terminal domain. It is similar in organization to calpains 5 and 6. This gene is associated with type 2 or non-insulin-dependent diabetes mellitus (NIDDM) and located within the NIDDM1 region. Multiple alternative transcript variants encoding different isoforms have been described for this gene.
