This MAb recognizes a single chain, transmembrane, heavily glycosylated protein of 90-120 kDa, which is identified as CD34. On the basis of differential sensitivity to degradation by specific enzymes, epitopes of monoclonal antibodies to CD34 are classified intothree main categories, class I, class II and class III. It is a class II antibody whose epitope is resistant to neuraminidase but sensitive to glycoprotease and chymopapain.CD34 expression is a hallmark for identifying pluripotent hematopoietic stem or progenitor cells. Its expression is gradually lost as lineage committed progenitors differentiate. CD34 is a marker of choice for staining blasts in acute myeloid leukemia. In addition, CD34 is expressed by soft tissue tumors, such as solitary fibrous tumor and gastrointestinal stromal tumor. Its expression is also found in vascular endothelium. Additionally, it appears that proliferating endothelial cells express this molecule more than the non-proliferating endothelial cells. Anti-CD34 labels > 85% of angiosarcoma and Kaposi's sarcoma, but with a lower specificity.

NCAM2 is an 837 amino acid protein encoded by the human gene NCAM2. NCAM2 contains five immunoglobulin-like domains, two Fibronectin type III domains, a transmembrane domain and a cytoplasmic domain. The gene is expressed most strongly in human adult and fetal brain. NCAM2 is a member of the neural cell adhesion molecule (NCAM) family. NCAMs are closely related cell surface glycoproteins involved in cell to cell interactions during growth and are thought to play an important role in embryogenesis and development. NCAM2 is a considered a good candidate for involvement in certain Down syndrome phenotypes because a slight overexpression of NCAMs increases many-fold the homotypic adhesion properties of cells. Stat5 regulates NCAM2 in vivo by binding to the NCAM2 intron in the NKL natural killer cell line; this binding is induced by cytokines that activate Stat5. Neither Stat1 nor Stat3 bind to this region, despite sharing a consensus binding sequence with Stat5.

Fe65L2 is a 486 amino acid protein that contains one WW domain and two PID domains. Binding to the intracellular domain of the -Amyloid precursor protein, Fe65L2 is thought to modulate the internalisation and, therefore, the accessibility and function of -Amyloid. Via its ability to control the intracellular accumulation of -Amyloid, Fe65L2 is thought to play a role in the pathogenesis of Alzheimer's disease. Fe65L2 exists as four alternatively spliced isoforms designated isoform I, isoform II, isoform III and isoform IV. Fe65L2 interacts with Amyloid-like protein and is encoded by a gene located on human chromosome 5, which contains 181 million base pairs and comprises nearly 6% of the human genome. Deletion of the p arm of chromosome 5 leads to Cri du chat syndrome, while deletion of the q arm or of chromosome 5 altogether is common in therapy-related acute myelogenous leukaemias and myelodysplastic syndrome.

Ro autoantigens are of clinical significance because directed against them are found in most patients with primary Sjqgren syndrome, subacute cutaneous lupus erythematosus (SLE), neonatal lupus erythematosus, ANA-negative lupus erythematosus, and systemic lupus erythematosus-like disease secondary to homozygous C2 or C4 complement deficiency (1). Ro/SSA is a ribonucleoprotein that binds to auto in 35 to 50% of patients with SLE and in up to 97% of patients with Sjqgren syndrome (2). The Ro/SSA particle consists of a single immunoreactive protein noncovalently bound with one of four small RNA molecules (2). Most anti-Ro/SSA-positive sera detect not only the main protein, but also a smaller Ro/SSA protein (2). The genes which encode the smaller and larger proteins map to human chromosomes 11p15.5 and 1q31, respectively (3?). The family of voltage-dependent chloride channels (CLCs) regulate cellular trafficking of chloride ions, a critical component of all living cells. CLCs regulate excitability in muscle and nerve cells, aid in organic solute transport, and maintain cellular volume. CLC-KA is a kidney-specific chloride channel that mediates transepithelial chloride transport in the thin ascending limb of the Henle loop in the inner medulla. CLC-KA plays a crucial role in urine concentration. The gene encoding human CLC-KA maps to chromosome 1p36. Mutations in this gene may be associated with nephrogenic diabetes insipidus in those cases where mutations in the vasopressin V2 receptor and the AQP2 water channel are lacking. CLC-KB mediates basolateral chloride ion efflux in the thick ascending limb and in more distal nephron segments. The gene encoding human CLC-KB maps to chromosome 1p36. Mutations in this gene cause type III Barter’s syndrome which is characterized by renal salt-wasting and low blood pressure.

Midline-1 (Tripartite motif-containing protein 18, Putative transcription factor XPRF, RING finger protein 59) is a 667 amino acid protein encoded by the human gene MID1. Midline-1 belongs to the TRIM/RBCC family and contains two B box-type zinc fingers, one B30.2/SPRY domain, one COS domain, one fibronectin type-III domain and one RING-type zinc finger. Midline-1 is believed to have E3 ubiquitin ligase activity which targets the catalytic subunit of protein phosphatase 2 for degradation. It is a cytoplasmic protein found as a homodimer or heterodimer with Midline-2. It also interacts with IGBP1 (Lymphocyte signaling protein A4). Defects in MID1 are the cause of Opitz syndrome type I (OS-I). OS-I is an X-linked recessive disorder characterized by hypertelorism, genital-urinary defects such as hypospadias in males and splayed labia in females, lip-palate-laryngotracheal clefts, imperforate anus, developmental delay and congenital heart defects. OS-I mutations produce proteins with a decreased affinity for microtubules.
