The 3-HAO family of proteins consists of intramolecular dioxygenases that contain non-heme ferrous iron. HAAO (3-hydroxyanthranilate 3,4-dioxygenase), also known as HAO or 3-HAO, is a 286 amino acid monomeric cytoplasmic protein that belongs to the 3-HAO family. Utilizing Fe2+ ions as a cofactor, HAAO participates in the formation of quinolinic acid (QUIN), and excitotoxin, by catalyzing the oxidative ring opening of 3-hydroxyanthranilate to 2-amino-3-carboxymuconate semialdehyde. HAAO may play a role in disorders that are associated with varying levels of QUIN expression, and is widely expressed in peripheral organs, including kidney and liver. Existing as two alternatively spliced isoforms, the gene encoding HAAO maps to human chromosome 2p21 and mouse chromosome 17 E4.

Recognizes a protein of 80 kDa-90 kDa, identified as CD36. It is expressed on platelets, monocytes and macrophages, microvascular endothelial cells, erythrocyte precursors, mammary epithelial cells, and some macrophage derived dendritic cells. CD36 acts as a receptor for thrombospondin (TSP), collagen types I, IV and V, P. falciparum malaria-infected erythrocytes, and sickle erythrocytes. It also functions as a scavenger receptor, mediating macrophage uptake of oxidized low-density lipoprotein (LDL) and recognition of apoptotic polymorphonuclear leukocytes (PMN). CD36 plays a role in platelet aggregation, macrophage foam cell development, inflammation, and the tissue ischemia observed in sickle cell disease and cerebral malaria. Note that 1-4% of Japanese and East Asia population lack CD36.

Friedreich ataxia is a progressive neurodegenerative disorder caused by loss of function mutations in the frataxin gene. The human frataxin gene maps to chromosome 9q13.The frataxin gene encodes a mitochondrial protein of the same name. Frataxin assembles into a stable homopolymer with iron-binding capabilities. When expressed in E. Coli human frataxin binds iron atoms at a rate of 10 iron atoms per 1 molecule of the frataxin polymer. Thus, frataxin appears to function in some capacity for iron-storage for the mitochondria. Frataxin may also function as an activator of oxidative phosphorylation to increase mitochondrial membrane potential and elevate cellular ATP. Frataxin is expressed in tissues with high metabolic activity including heart, liver and brown fat.

ADH6 (alcohol dehydrogenase 6), also known as ADH-5, is a 368 amino acid member of the class V zinc-containing alcohol dehydrogenase family. This family of enzymes functions to metabolize a wide variety of substrates such as retinol, hydroxysteroids, ethanol, aliphatic alcohols and lipid peroxidation products. Localized to the cytoplasm and expressed in the stomach and liver, ADH6 catalyzes the reversible oxidation of alcohols to their corresponding aldehydes or ketones and is able to bind two zinc ions as cofactors. ADH6 contains a glucocorticoid response element upstream of its 5' UTR which is thought to be a steroid binding site, suggesting that expression of ADH6 may be under hormonal control. Multiple isoforms of ADH6 exist due to alternative splicing events.

Environmental stressors to K562 cells such as heat shock, a higher temperature than the ideal body temperature of the organism from which the cell line was derived, and oxidative stress, the production of oxygen-centered free radicals, regulate and increase the expres-sion of JWA. This response to environmental stressors suggests similiarity of JWA to heat shock protein 70 (HSP70), which is upregulated by heat stress and toxic chemicals.

17beta-HSD8 belongs to the 17beta-HSD family of proteins that regulate the availability of steroids within a tissue. 17beta-HSD8 converts active steroids to their inactive form through its oxidative activity. It is a key player in the inactivation of Estradiol and Testosterone. 17beta-HSD8 is predominantly expressed in placenta, endometrium and prostate but can also be found in liver, and pancreas, with lowest levels found in testis, ovary and kidney. It has been proposed that a reduction in the levels of 17beta-HSD8 may lead to abnormal elevations in the local level of sex steroids, which can lead to recessive renal cystic disease. It has also been suggested that low levels of 17beta-HSD proteins may result in an underdeveloped urogenital system.

Initiates repair of AP sites in DNA by catalyzing hydrolytic incision of the phosphodiester backbone immediately adjacent to the damage, generating a single-strand break with 5'-deoxyribose phosphate and 3'-hydroxyl ends. Does also incise at AP sites in the DNA strand of DNA/RNA hybrids, single-stranded DNA regions of R-loop structures, and single-stranded RNA molecules. Has a 3'-5' exoribonuclease activity on mismatched deoxyribonucleotides at the 3' termini of nicked or gapped DNA molecules during short-patch BER. Possesses a DNA 3' phosphodiesterase activity capable of removing lesions (such as phosphoglycolate) blocking the 3' side of DNA strand breaks. May also play a role in the epigenetic regulation of gene expression by participating in DNA demethylation. Acts as a loading factor for POLB onto non-incised AP sites in DNA and stimulates the 5'-terminal deoxyribose 5'-phosphate (dRp) excision activity of POLB. Plays a role in the protection from granzymes-mediated cellular repair leading to cell death. Also involved in the DNA cleavage step of class switch recombination (CSR). On the other hand, APEX1 also exerts reversible nuclear redox activity to regulate DNA binding affinity and transcriptional activity of transcriptional factors by controlling the redox status of their DNA-binding domain, such as the FOS/JUN AP-1 complex after exposure to IR.
