PCDH12 Antibody: Protocadherins comprise the largest group within the cadherin family of calcium-dependent cell-cell adhesion molecules. Protocadherin 12 (PCDH12) was initially identified through PCR screening of mouse heart microvascular endothelial cell RNA; further experiments revealed its mRNA to be strongly expressed in highly vascularized organs such as lung and kidney, in addition to glycogen-rich trophoblasts in the placenta. PCDH12-null mice are viable and fertile, but show reduced placental and embryonic sizes when compared to wild-type mice. Further studies showed significant expression changes in 2, 289 genes, including those involved in tissue morphogenesis, angiogenesis, cell-matrix adhesion and migration, immune response and chromatin remodeling.

Hairy/enhancer of split-related proteins, such as HEY1, are basic helix-loop-helix (bHLH) transcription factors implicated in cell fate decision and boundary formation. HEY genes are direct transcriptional targets of the Notch signaling pathways in Drosophila and vertebrates. This gene encodes an estrogen receptor, a ligand-activated transcription factor composed of several domains important for hormone binding, DNA binding, and activation of transcription. The protein localizes to the nucleus where it may form a homodimer or a heterodimer with estrogen receptor 2. Estrogen and its receptors are essential for sexual development and reproductive function, but also play a role in other tissues such as bone. Estrogen receptors are also involved in pathological processes including breast cancer, endometrial cancer, and osteoporosis. Alternative splicing results in several transcript variants, which differ in their 5' UTRs and use different promoters.

Fibroblast growth factor 21 (FGF-21) is a member of the fibroblast growth factor family and has been identified as an important regulator of energy metabolism connecting nutrition, growth, reproduction and longevity. FGF-21 lacks the heparin-binding domain of most FGF proteins, allowing its secretion. FGF-21 is abundantly expressed in adipose tissues, liver and pancreas. In adipose tissue, FGF-21 promotes glucose uptake and oxidation and in liver it replenishes on fasting the tissues with fuel on low nutritional supply. It also induces lipolysis and ketogenesis. FGF-21 acts through FGF receptors (FGFR) associated with the auxiliary protein beta-Klotho. Recently, FGF-21 has been reported to interact directly with the brain circadian clock to coordinate activity and reproduction as part of the adaptation to fasting. Due to its multiple functions of normalizing glucose, lipid and energy homeostasis, FGF-21 represents an attractive novel therapy for type 2 diabetes mellitus and obesity.
