Nidogens are highly conserved proteins present in vertebrate and invertebrate basement membranes. Nidogens connect the Laminin and Collagen IV networks and integrate other proteins into the membrane. In mammals, two Nidogen proteins, Nidogen and Nidogen-2, interact at comparable levels with Collagen I, IV and Perlecan, serving to stabilize basement membranes and playing a major role in embryogenesis. The two isoforms have a similar shape, consisting of three globular domains, and co-localize in vessel walls and other basement membrane zones. Nidogen-2 is a cell adhesion protein glycosylated at nitrogen and oxygen sites, and is widely distributed in basement membranes in heart, placenta, bone and, to a lesser extent, in pancreas, kidney and skeletal muscle.

EYA3 is a 573 amino acid protein that localizes to both the nucleus and the cytoplasm and is one of several mammalian homologs of the Drosophila Eya (eyes absent) protein. Existing as two alternatively spliced isoforms, EYA3 possesses magnesium-catalyzed phosphatase activity and is thought to play a role in transcriptional regulation during organogenesis. Specifically, EYA3 interacts with proteins such as Six1 and, via this interaction, functions to activate the expression of genes that are involved in cellular proliferation and organ development. Upon DNA damage, EYA3 may be phosphorylated by ATM or ATR. The gene encoding EYA3 maps to chromosome 1, which spans about 260 million base pairs and comprises nearly 8% of the human genome.

The metabolism of glutathione releases free glutamate and the dipeptide, cysteinyl-glycine, which is hydrolysed to cysteine and glycine by dipeptidases. In the presence of high concentrations of dipeptides and some amino acids, can also catalyse a transpeptidation reaction, transferring the gamma-glutamyl moiety to an acceptor amino acid to form a new gamma-glutamyl compound. Initiates extracellular glutathione (GSH) breakdown, provides cells with a local cysteine supply and contributes to maintain intracellular GSH level. CTBS is an evolutionarily conserved member of the glycosyl hydrolase 18 family of proteins. Localizing to the lysosome, CTBS plays a role in the degradation of asparagine-linked (Asn-linked) glycoproteins. Glycoproteins are translocated to lysosomes via endocytosis or autophagy where they are broken down by proteases and glycosidases. The catabolism of glycoproteins is an important step in the regular turnover of cellular contents and in maintaining the homeostasis of glycosylation. CTBS functions as a glycosidase that cleaves the reducing end GlcNAc from the core chitobiase unit of oligosaccharides. Before this reaction can occur, AGA (the lysosomal glycosylasparaginase) must first remove the Asn from the Asn-linked glycoprotein to expose the reducing end GlcNAc, thereby allowing CTBS to access the exposed moiety.

MSK1 is a mitogen and stress activated protein kinase 1 which belongs to the AGC family of kinases and is related in structure to the ribosomal p70 S6 kinase subfamily. MSK1 can be activated by ERK1/2 and SAPK2/p38 MAP kinase. It is also known to be required for the phosphorylation of CREB, ATF1 H3 and HMG14 in response to mitogen and stress. Similar to RSK, MSK1 contains two kinase domains (N term and a C term). Once phosphorylated on Thr581 and Ser360 by ERK1/2 and SAPK2/p38, MSK1 autophosphorylate on at least 5 sites. Of these autophosphorylation sites Ser212 and Ser376 get phosphorylated by the C terminal kinase domain of MSK1 which is essential for the catalytic activity of the N terminal kinase domain.
