Major histocompatibility complex (MHC) molecules form an integral part of the immune response system. They are cell-surface receptors that bind peptides and present them to T lymphocytes. Human leukocyte antigens (HLAs) are polymorphic members of the MHC family that are specifically involved in the presentation of antigens to the T cell receptor. There are two classes of HLA antigens: class I (HLA-A, HLA-B and HLA-C) and class II (HLA-D). Class I molecules are expressed in nearly all cells and play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum. The differential structural properties of MHC class I and class II molecules account for their respective roles in activating different populations of T lymphocytes. HLA-B encodes a membrane anchored heavy chain which hetero-dimerizes with a light chain (∫-2-Microglobulin) to form MHC-I. Polymorphisms yield hundreds of HLA-B alleles. The HLA-B27 allele appears with increased frequency in uveitis patients.

There are two forms of glutamic acid decarboxylases (GADs) that exist in brain: GAD-65 (also known as GAD2) and GAD-67 (also known as GAD1, GAD or SCP). GAD-65 and GAD-67 are members of the group II decarboxylase family of proteins and are responsible for catalyzing the rate limiting step in the production of GABA (gamma-aminobutyric acid) from L-glutamic acid. Although both GADs are found in brain, GAD-65 localizes to synaptic vesicle membranes in nerve terminals, while GAD-67 is distributed throughout the cell. GAD-67 is responsible for the basal levels of GABA synthesis. In the case of a heightened demand for GABA in neurotransmission, GAD-65 transiently activates to assist in GABA production. As a member of the group II decarboxylase family, GADL1 (Glutamate decarboxylase-like protein 1) is a 521 amino acid protein that utilizes pyridoxal phosphate as a cofactor for its carboxylase activity. There are two isoforms of GADL1 that exist as a result of alternative splicing events.

Proteins internalized into the endocytic pathway are usually degraded. Efficient proteolysis requires denaturation, induced by acidic conditions within lysosomes, and reduction of inter- and intrachain disulfide bonds. Cytosolic reduction is mediated enzymatically by thioredoxin. In the endocytic pathway, reduction of protein disulfide bonds is important for the generation of MHC class II-peptide complexes. This process is catalyzed by a gamma-interferon-inducible thiol reductase (GILT). GILT is synthesized as a precursor, and following delivery to MHC class II-containing compartments (MIICs), is processed to the mature form via cleavage of amino- and carboxy-terminal propeptides. A lysosomal thiol reductase, GILT, is optimally active at low pH and capable of catalyzing disulfide bond reduction both in vivo and in vitro. GILT is expressed constitutively in antigen-presenting cells and is induced by g-interferon in other cell types, suggesting a potentially important role in antigen processing. Additionally, T cell recognition of select exogenous and endogenous epitopes is dependent on tumor cell expression of GILT. The absence of GILT in melanomas alters antigen processing and the hierarchy of immunodominant epitope presentation.

The GK1.5 antibody reacts with mouse CD4, a 55 kDa protein which acts as a co-receptor for the T cell receptor (TCR) in its interaction with MHC Class II molecules on antigen-presenting cells. The extracellular domain of CD4 binds to the beta-2 domain of MHC Class II, while its cytoplasmic tail provides a binding site for the tyrosine kinase lck, facilitating the signaling cascade that initiates T cell activation. CD4 is typically expressed on thymocytes, certain mature T cell populations such as Th17 and T regulatory (Treg) cells, as well as on dendritic cells. The GK1.5 antibody is widely used as a phenotypic marker for CD4 expression. If used together, the GK1.5 antibody and an alternative antibody, Anti-Mouse CD4 clone RM4-5, will "compete" for binding, i.e. RM4-5 is able to block GK1.5 binding to cells. In contrast, the Anti-Mouse CD4 clone RM4-4 does not block binding of the GK1.5 antibody to cells (Arora S et al. 2006. Infect. Immun. 74: 4339-4348). The GK1.5 antibody is also reported to be cross-reactive with Syrian hamster CD4.

The protein encoded by this gene belongs to the class-II aminoacyl-tRNA synthetase family. Aminoacyl-tRNA synthetases play critical roles in mRNA translation by charging tRNAs with their cognate amino acids. The encoded protein is a mitochondrial enzyme that specifically aminoacylates alanyl-tRNA. Mutations in this gene are a cause of combined oxidative phosphorylation deficiency 8.
