The LIM-only (LMO) proteins, LMO1 and LMO2, are nuclear factors that are characterized by a conserved LIM domain (1). The LIM domain consists of a cysteine-rich zinc-binding motif that is present in a variety of transcription factors, including the LIM homeobox (LHX) proteins expressed in the central nervous system and involved in cell differentiation (2). NLI, known also as CLIM-1, and the related protein CLIM-2 facilitate the formation of heteromeric LIM complexes and also enhance the nuclear retention of LIM proteins (5). LMO2 and the related protein LMO4 are expressed in thymic precursor cells (6). LMO4 is also expressed in mature T cells, cranial neural crest cells, somite, dorsal limb bud mesenchyme, motor neurons, and Schwann cell progenitors (7).

Programmed death-ligand 1 (PD-L1), also known as CD274 or B7-H1, is a protein that in humans is encoded by the CD274 gene. It is mapped to 9p24.1. PD-L1 is a 40kDa type 1 transmembrane protein that has been speculated to play a major role in suppressing the immune system during particular events such as pregnancy, tissue allografts, autoimmune disease and other disease states such as hepatitis. It has been concluded that upregulation of PD-L1 on tumor MDCs downregulates T-cell immunity and that PD-L1 blockade may represent an approach for cancer immunotherapy. Additionally, PD-L1 can provide positive costimulatory signals for innate and adaptive immunity and for protection against intracellular bacterial infection. What’s more, it has been found that PD1/PDL1 pathway may be a good target for restoring antitumor immunity in ovarian cancer.

Mouse Interleukin 36 beta (IL-36B)is a member of the IL-1 family of proteins. It is a cytokine that binds to and signals through the IL1RL2/IL-36R receptor which in turn activates NF-kappa-B and MAPK signalling pathways in target cells linked to a pro-inflammatory response. IL-36B is synthesised in several cells including resting and activated monocytes, and B cells. The receptor for IL-36 beta is thought to be a combination of IL-1 Rrp2 and IL-1 RAcP. Interleukin 36 beta is one part of the IL-36 signalling system that is thought to be present in epithelial barriers and to take part in local inflammatory response; similar to the IL-1 system with which it shares the coreceptor IL1RAP. Interleukin 36 beta are involved in a number of fundamental biological processes such as stimulating production of interleukin-6 and interleukin-8 in synovial fibrobasts, articular chondrocytes and mature adipocytes, inducing expression of a number of antimicrobial peptides including beta-defensin 4 and beta-defensin 103 as well as a number of matrix metalloproteases , inducing the production of proinflammatory cytokines in bone marrow-derived dendritic cells (BMDCs), including IL-12, Il-1 beta, IL-6, TNF-alpha and IL-23, and activating p38 MAPK phosphorylation in BMDCs.Moreover, interleukin 36 beta may be involved in skin inflammatory response by acting on keratinocytes, dendritic cells, and indirectly on T cells to drive tissue infiltration, cell maturation and cell proliferation. It plays an important role in dendritic cell maturation by stimulating the surface expression of CD80, CD86 and MHC class II and inducing the production of IFN-gamma, IL-4 and IL-17 by T helper 1 (Th1) cells, cultured CD4+ T cells and splenocytes.

IGSF11 is also known as BTIGSF (brain and testis-specific immunoglobulin superfamily protein) or VSIG3 (V-set and immunoglobulin domain-containing protein 3) and is a 431 amino acid protein that is expressed as three isoforms. IGSF11 is highly expressed in testis and ovary and is also expressed in brain, kidney and skeletal muscle, localized to the cellular membrane as a single-pass membrane protein. IGSF11 is an immunoglobulin with V-type and C2-type domains that function in molecular recognition. When IGSF11 is in the trans position, it plays an important role in cell-cell adhesion via both homophilic and heterophilic interactions with other molecules. These cell–cell interactions are also thought to be important for neuronal cell interactions, such as neuron–neuron or neuron–glia interactions, which are important for the development and function of the central nervous system. In addition, IGSF11 might also be involved interactions between Sertoli cells and spermatocytes, which are important associations during spermatogenesis. The IGSF11 gene is commonly upregulated in gastric cancer and IGSF11 is highly expressed in many types of human tumors, indicating that it may be useful as a target for immunotherapy.

Eukaryotic ABC transporters are largely responsible for trafficking hydrophobic compounds either within the cell as part of a metabolic process, outside the cell for transport to other organs, or for secretion from the body. The cholesterol-responsive transporter, ABCA7, maps to human chromosome 19 and mouse chromosome 10 and has been reported as a candidate regulator of ceramide transport in epidermal lipid reorganization. High expression levels of ABCA7 have been reported in myelolymphatic tissues, reticuloendothelial cells, peripheral leukocytes, thymus, spleen and bone marrow. This expression pattern of the two alternatively-spliced isoforms also indicates an involvement in lipid homeostasis in cells of the immune system, though the complete role of ABCA7 is not yet known. Full-length type I ABCA7 has shown plasma membrane localization, while the type II splicing variant has shown expression predominantly in the endoplasmic reticulum.

Fibrinogen is the main protein of blood coagulation system. It is a large protein and it consists of two identical subunits that contain three polypeptide chains: alpha, beta and gamma. All chains are connected with each other by a number of disulfide bonds. Fibrinopeptides A (1 to 16 amino acids) and B (1 to 17 amino acids) are released by thrombin from the N terminal parts of alpha and beta chains, respectively. In this way fibrinogen is converted into fibrin, which by means of polymerization forms a fibrin clot. Fibrinogen clotting underlies pathogenesis of MI, thromboembolism and thromboses of arteries and veins, since fibrin is the main substrate for thrombus formation. Fibrinogen activation is also involved in pathogenesis of inflammation, tumor growth and many other diseases. The normal fibrinogen concentration in plasma is about 3 mg/ml. The elevated level of fibrinogen in patient's blood is regarded as an independent risk factor for cardiovascular diseases. An increase in blood fibrinogen concentration was shown to be a strong predictor of coronary heart disease (Sonel A. et al, and Rapold H.J. et al). All these facts make fibrinogen an important parameter in the diagnosis of cardiovascular diseases.

Serotonin (also designated 5-hydroxytryptamine or 5-HT) is a molecule that functions as a neurotransmitter, a hormone and a mitogen, and it is predominantly expressed in the gut, platelets and central nervous system (CNS). In the CNS, serotonin modulates several processes, including anxiety, sleep, appetite, behavior and drug abuse. In platelets and gut, serotonin plays a major role in cardiovascular function and motility of the gastrointestinal tract, respectively. Serotonin mediates its effects through several of G protein coupled receptors, designated 5-HT receptors or alternatively SR receptors. The SR-2 receptors are comprised of three subtypes, SR-2A, SR-2B and SR-2C, which activate phospholipase C and release intracellular stores of calcium in response to serotonin. SR-2A has a specific role in tracheal smooth muscle contraction, bronchoconstriction and mediating aldosterone production, and it is also thought to play a role in several psychiatric disorders, including depression and schizophrenia. SR-2B is expressed in embryonic and adult cardiovascular tissues, gut and brain and plays an important role in the pathology of cardiac disorders. SR-2C is thought to mediate the effects of atypical antipsychotic drugs.

The cystatin superfamily is a well-established family of cysteine protease inhibitors. Cystatins A and B (type 1) are mainly intracellular; cystatins C, D, E/M, F, G, S, SN and SA cystatins are extracellular (type 2); and the kininogens are type 3 cystatins which are intravascular proteins. All true cystatins inhibit cysteine peptidases of the papain family, such as cathepsins, and some also inhibit legumain family enzymes. Cystatin SA, cystatin S and cystatin SN are found primarily in saliva. Cystatin S and SN can also be expressed in tears, urine and seminal fluid. Cystatin C is a related protein which is expressed in brain, thymus, ovary, epididymis and vas deferens. Cystatin D protects against proteinases in the oral cavity, while Cystatin E/M and F moderate the inhibition of cathepsin proteins. The fetuins, part of the cystatin superfamily, are secretable proteins that influence osteogenesis and bone resorption, regulation of the insulin and hepatocyte growth factor receptors and the response to systemic inflammation. High molecular weight kininogen (Kininogen HC) and low molecular weight kininogen (Kininogen LC) have varied roles, though they both inhibit the thrombin- and plasmin-induced aggregation of thrombocytes.

PIST Antibody: Autophagy, the process of bulk degradation of cellular proteins through an autophagosomic-lysosomal pathway is important for normal growth control and may be defective in tumor cells. It is involved in the preservation of cellular nutrients under starvation conditions as well as the normal turnover of cytosolic components and is negatively regulated by TOR (Target of rapamycin). PIST, a PDZ-containing protein, was discovered in a yeast two-hybrid system as a binding partner to Beclin-1, a Bcl-2-interacting protein homologous to the yeast autophagy gene apg6. Experiments with mutant PIST proteins lacking the PDZ domain showed that PIST interaction with Beclin-1 through its coiled-coil domain can modulate Beclin-1 activity and suggest that PIST interactions with other proteins through its PDZ domain may regulate the activity of PIST and Beclin-1.
