Hemagglutinin Monoclonal Antibody: Influenza A virus is a major public health threat, killing more than 30,000 people per year in the USA. Novel influenza virus strains caused by genetic drift and viral recombination emerge periodically to which humans have little or no immunity, resulting in devastating pandemics. Influenza A can exist in a variety of animals, but it is in birds that all subtypes can be found. These subtypes are classified based on the combination of the virus coat glycoproteins hemagglutinin (HA) and neuraminidase (NA) subtypes. HA interacts with host cell surface proteins containing oligosaccharides with terminal sialyl residues. Its extracellular region has two domains (HA1 and HA2); HA1 is cleaved from the main hemagglutinin protein by the host immune system. During 1997, an H5N1 avian influenza virus was determined to be the cause of death in 6 of 18 infected patients in Hong Kong. This more recent virulent strain of H5N1 is now seen in Africa and Europe, as well as in Southeast Asia. There is some evidence of human to human spread of this virus, but it is thought that the efficiency of this type of transmission is low. Virus isolated from a human infected with the H5N1 strain in 1997 could bind to oligosaccharides from human as well as avian sources, indicating its species-jumping ability.

The Human Serpin superfamily consists of at least 35 members that target not only serine proteases, but also selected cysteine proteases and non-protease proteins. As protease inhibitors, serpins have an array of functions including regulating blood clotting, the complement pathway, extracellular matrix remodeling, and cell motility. Serpin G1 is a serine protease inhibitor protein. It is the largest member among the serpin class of proteins. Remarkably, Serpin G1 has a 2-domain structure, unlike most family members. The C-terminal serpin domain is similar to other serpins, and this part of Serpin G1 provides the inhibitory activity. The N-terminal domain is not essential for Serpin G1 to inhibit proteinases and has no similarity to other proteins. The main function of Serpin G1 is the inhibition of the complement system to prevent spontaneous activation. Serpin G1 is an acute phase protein and circulates in blood at levels of around 0.25g/L, whose levels rise 2-fold during inflammation. Although named after its complement inhibitory activity, Serpin G1 also inhibits proteinases of the fibrinolytic, clotting, and kinin pathways. Most notably, Serpin G1 play a potentially crucial role in regulating important physiological pathways including complement activation, blood coagulation, fibrinolysis and the generation of kinins. It is also the most important physiological inhibitor of fXIIa, chymotrypsin and plasma kallikrein.

RFC, also named activator 1, is a protein complex consisting of five distinct subunits of 140, 40, 38, 37, and 36 kD. RFC5 is the 36 kD subunit. This subunit can interact with the C-terminal region of PCNA. It forms a core complex with the 38 and 40 kDa subunits. Membrane Receptors Transforming growth factor beta is a multifunctional cytokine known to modulate several tissue development and repair processes, including cell differentiation, cell cycle progression, cellular migration, adhesion, and extracellular matrix production. There are 3 forms encoded by separate genes TGFB1, TGFB2, and TGFB3. The diverse effects of TGF beta are mediated by the TGF beta receptors and cell surface binding proteins. In addition to type I TGF beta receptor (TGFBR1) and type II (TFGBR2), type III (TGF beta III receptor) has been identified. It is a glycoprotein that binds TGF beta and exists in both a membrane bound and a soluble form. It may serve as a receptor accessory molecule in both the TGF beta and fibroblast growth factor systems. TGF beta III receptor lacks a recognizable signaling domain and has no clearly defined role in TGF beta signaling. Endothelial cells undergoing epithelial mesenchymal transformation express TGF beta III receptor, and TGF beta III receptor specific antisera inhibits mesenchyme formation and migration. Misexpression of TGF beta III receptor in nontransforming ventricular endothelial cells conferrs transformation in response to TGFB2. These results support a model where TGF beta III receptor localizes transformation in the heart and plays an essential, nonredundant role in TGF beta signaling. TGF beta III receptor, or beta glycan, can function as an inhibin coreceptor with ActRII. TGF beta III receptor binds inhibin with high affinity and enhances binding in cells coexpressing ActRII and TGF beta III receptor. Inhibin forms crosslinked complexes with both recombinant and endogenously expressed TGF beta III receptor and ActRII. TGF beta III receptor confers inhibin sensitivity to cell lines that otherwise respond poorly to this hormone.
