Beschreibung:   Primordial germ cell (PGCs)-specific protein involved in epigenetic chromatin reprogramming in the zygote following fertilization. In zygotes, DNA demethylation occurs selectively in the paternal pronucleus before the first cell division, while the adjacent maternal pronucleus and certain paternally-imprinted loci are protected from this process. Participates in protection of DNA methylation in the maternal pronucleus by preventing conversion of 5mC to 5hmC: specifically recognizes and binds histone H3 dimethylated at 'Lys-9' (H3K9me2) on maternal genome, and protects maternal genome from TET3-mediated conversion to 5hmC and subsequent DNA demethylation. Does not bind paternal chromatin, which is mainly packed into protamine and does not contain much H3K9me2 mark. Also protects imprinted loci that are marked with H3K9me2 in mature sperm from DNA demethylation in early embryogenesis. Beschreibung:   JMJD2A Antibody: Members of the Jumonji domain 2 (JMJD2) family contain a JmjN domain, a JmjC domain, a JD2H domain, two TUDOR domains, and two PHD-type zinc fingers. The first member of this group, JMJD2A, is widely expressed in human tissues and cell lines and functions as a trimethylation-specific demethylase, converting specific trimethylated histone residues to the dimethylated form, and as a transcriptional repressor. JMJD2A can also form a complex with the androgen receptor (AR), a transcription factor that is pivotal for the development of prostate cancer. Overexpression of JMJD2A stimulates AR function and this stimulation is dependent on JMJD2A catalytic activity, suggesting that JMJD2A might be a critical protein with roles in cell proliferation and oncogenesis.

Beschreibung:   Modulation of the chromatin structure plays an important role in the regulation of transcription in eukaryotes. The nucleosome, made up of four core histone proteins (H2A, H2B, H3 and H4), is the primary building block of chromatin. The N-terminal tail of core histones undergoes different posttranslational modifications including acetylation, phosphorylation and methylation. These modifications occur in response to cell signal stimuli and have a direct effect on gene expression. In most species, the histone H2B is primarily acetylated at lysines 5, 12, 15 and 20. Histone H3 is primarily acetylated at lysines 9, 14, 18 and 23. Acetylation at lysine 9 appears to have a dominant role in histone deposition and chromatin assembly in some organisms. Phosphorylation at Ser10 of histone H3 is tightly correlated with chromosome condensation during both mitosis and meiosis.

Expressed at low levels in prostate tissue, AIP1 functions as a Ras GTPase-activating protein that interacts with ASK 1 and, via this interaction, disrupts the association of ASK 1 with an inhibitory 14-3-3 complex, thereby allowing free ASK 1 to function within the cell. AIP1 exists as multiple alternatively spliced isoforms and is down-regulated in prostate cancer tissue, suggesting a role in tumor suppression. Chromosomal aberrations in the gene encoding AIP1 are associated with acute myeloid leukemia (AML), implicating a role for AIP1 fusion proteins in tumorigenesis.

Containing 6 C2H2-type zinc fingers which function to bind DNA, ZNF219 is thought to be involved in transcriptional regulation. Specifically, ZNF219 has been identified as a transcriptional repressor that downregulates the transcription of the HMG-14 promoter.
