Dtk, also called Tyro3, belongs to the TAM receptor family of receptor protein tyrosine kinases (RPTKs) composed of three receptors Tyro3, Axl, and Mer. These receptors share a characteristic molecular structure of two immunoglobulin-like and two fibronectin type III repeats and have been best characterised for their roles in immune regulation, fertility, thrombosis and phagocytosis. Gas6 and protein S have been identified as ligands for these receptors. Gas6 binding induces tyrosine phosphorylation and downstream signalling pathways that can lead to cell proliferation, migration, or the prevention of apoptosis. Tyro3 and Axl play important regulatory roles in a variety of tissues, including the central nervous, reproductive, immune, and vascular systems. Tyro3 is widely expressed during embryonic development and preferentially expressed during neurogenesis in the central nervous system.

Human Chemokine (C-X-C motif) Ligand 7 (CXCL7), also known as neutrophil activating peptide 2 (NAP-2), is a member of the CXC chemokines containing an ELR domain (Glu-Leu-Arg tripeptide motif). Similar to other ELR domain containing CXC chemokines, such as IL-8 and the GRO proteins, CXCL7 binds CXCR2, chemoattracts and activates neutrophils. CXCL7, Connective Tissue Activating Protein III (CTAPIII) and beta thrombogulin ( beta TG), are proteolytically processed carboxylterminal fragments of platelet basic protein (PBP) which is found in the alphagranules of human platelets. Although CTAPIII, beta TG, and PBP represent amino-terminal extended variants of NAP2 and possess the same CXC chemokine domains, these proteins do not exhibit CXCL7/NAP2 activity. CXCL7 induces cell migration through the G-protein-linked receptor CXCR-2.

BAT1 is a member of the DEAD protein family of ATP-dependent RNA helicases. Members of this family are involved in a number of cellular functions including initiation of translation, RNA splicing, and ribosome assembly. A cluster of genes, BAT1-BAT5, has been localized in the vicinity of the genes for TNF alpha and TNF beta. These genes are all within the human major histocompatibility complex class III region. This protein is a negative regulator of inflammation. It is also thought to be a translation initiation factor. This gene is a strong candidate gene for rheumatoid arthritis. There are multiple alternatively spliced transcript variants known for this gene but only two have been fully described. Both of these variants encode the same isoform. This gene has been found to have multiple polyadenylation sites.

FLT3 encodes a class III receptor tyrosine kinase that regulates hematopoiesis. The receptor consists of an extracellular domain composed of five immunoglobulin-like domains, one transmembrane region, and a cytoplasmic kinase domain split into two parts by a kinase-insert domain. The receptor is activated by binding of the fms-related tyrosine kinase 3 ligand to the extracellular domain, which induces homodimer formation in the plasma membrane leading to autophosphorylation of the receptor. The activated receptor kinase subsequently phosphorylates and activates multiple cytoplasmic effector molecules in pathways involved in apoptosis, proliferation, and differentiation of hematopoietic cells in bone marrow. Mutations that result in the constitutive activation of this receptor result in acute myeloid leukemia and acute lymphoblastic leukemia.

NIPSNAP3A Antibody: The NIPSNAP proteins comprise a family of evolutionarily well-conserved proteins with strong sequence similarity to the central portion of a protein encoded by C. elegans chromosome III between a 4-nitrophenylphosphatase (NIP) domain and non-neuronal SNAP25-like protein. NIPSNAP2, a novel gene encoding a protein with tyrosine phosphorylation sites and a transmembrane domain, is co-amplified with EGFR in approximately 40% of glioblastomas, the most common and malignant form of central nervous system tumors. While NIPSNAP3B is highly expressed skeletal muscle, NIPSNAP3A mRNA levels are low. NIPSNAP3A protein is associated with plasma membrane and partially localized in rafts. NIPSNAP proteins have been suggested to be important in vesicular transport.

Glial Fibrillary Acidic Protein (GFAP) was discovered by Amico Bignami and co-workers as a major fibrous protein of multiple sclerosis plaques. It was subsequently found to be a member of the 10 nm or intermediate filament (IF) family, specifically the IF family Class III, which also includes peripherin, desmin and vimentin. GFAP is strongly and specifically expressed in astrocytes and certain other astroglia in the CNS, in satellite cells, peripheral ganglia, and in non-myelinating Schwann cells in peripheral nerves. In many damage and disease states GFAP expression is heavily upregulated in astrocytes. In addition, neural stem cells frequently strongly express GFAP. Point mutations in the protein coding region of the GFAP gene lead to Alexander disease which is characterized by the presence of abnormal astrocytes containing GFAP protein aggregates known as Rosenthal fibers.

This antibody recognises a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). cytokeratin 10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of cytokeratin 10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, cytokeratin 10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), cytokeratin 10 is detected very frequently. Expression of cytokeratin 10 is related to maturation of malignant keratinocytes, being preferentially detected in more-differentiated parts.

It also supports survival, proliferation, and function of the differentiated macrophages and regulates differentiation of mononuclear phagocytes to osteoclasts (Pixley and Stanley).
