This gene encodes a member of the Ced-4 family of apoptosis proteins. Ced-family members contain a caspase recruitment domain (CARD) and are known to be key mediators of programmed cell death. The encoded protein contains a distinct N-terminal pyrin-like motif, which is possibly involved in protein-protein interactions. This protein interacts strongly with caspase 2 and weakly with caspase 9. Overexpression of this gene was demonstrated to induce apoptosis in cells. Multiple alternatively spliced transcript variants encoding distinct isoforms have been found for this gene, but the biological validity of some variants has not been determined. Tect3 is a 607 amino acid single-pass type I membrane protein that belongs to the tectonic family and exists as four alternatively spliced isoforms. Tect3 interacts with MKS1 and may be involved in apoptosis regulation. The gene that encodes Tect3 contains approximately 31,560 bases and maps to human chromosome 10q24.1. Spanning nearly 135 million base pairs and encoding nearly 1,200 genes, chromosome 10 makes up approximately 4.5% of the human genome. Several protein-coding genes, including those that encode chemokines, cadherins, excision repair proteins, early growth response factors (Egrs) and fibroblast growth receptors (FGFRs), are located on chromosome 10. Defects in some of the genes that map to chromosome 10 are associated with Charcot-Marie Tooth disease, Jackson-Weiss syndrome, Usher syndrome, nonsyndromatic deafness, Wolman’s syndrome, Cowden syndrome, Cockayne syndrome, multiple endocrine neoplasia type 2 and porphyria. Tetrahydrobiopterin deficiency and a number of syndromes involving defective skull and facial bone fusion are also linked to chromosome 10.

These seven transmembrane proteins include the adrenergic, serotonin and dopamine receptors. Like the a2-adrenergic receptor, serotonin receptor functions are also mediated by G proteins that inhibit the activity of adenylyl cyclase. These subunits, IKK alpha and Ikk beta, are protein kinases whose function is needed for NF kappa B activation by pro inflammatory stimuli. IKK is composed of similar amounts of IKK alpha, Ikk beta, which are differentially processed forms of a third subunit, IKK gamma. IKK gamma interacts preferentially with IKK beta and is required for the activation of the IKK complex.

Esophagin, also known as small proline-rich protein 3 (SPR3) or Cornifin ∫, belongs to the cornifin family of cornified-envelope structural proteins. It is expressed in mucosal epithelia such as esophagus and tongue and is strongly induced during epidermal keratinocyte differentiation. Due to its highly inducible nature, Esophagin is considered a marker of squamous differentiation. Esophagin serves as a cross-linking protein within the cornified cell envelope and may play a role in the maintenance of normal esophageal epithelial homeostasis. It shares significant homology with the related proteins, SPRR1 and SPRR2. Esophagin is typically not expressed in healthy human epithelium, but its expression is upregulated in numerous hyperproliferative disorders of the skin. Contrastly, its expression is dramatically downregulated in esophageal squamous cell carcinoma.

Defects in the gene encoding GALK1 can cause galactosemia II, an autosomal recessive disorder characterized by congenital cataracts during infancy, often within the first two weeks of life. In the adult population, galactosemia II can cause presenile cataracts that are secondary to accumulation of galactitol in the lens of the eye. A second gene, GALK2, encodes an enzyme with greater activity against GalNAc than galactose. GALK2 has been implicated in the salvage pathway for the reutilization of free GalNAc derived from the degradation of complex carbohydrates.

Neuromedin U is a neuropeptide with high activity on smooth muscle. It is widely expressed in gastrointestinal systems and central nervous system (CNS). Peripheral activities of neuromedin U include smooth muscle stimulation, ion transport alterations in the gut and the regulation of local blood flow and adrenocortical function. Neuromedin U receptors 1 and 2 (NMUR1 and NMUR2) are multi-pass membrane proteins that belong to the G-protein coupled receptor 1 family of proteins. Both NMUR1 and NMUR2 act as receptors for the neuromedin U neuropeptide. NMUR1 is detected in peripheral organs, particularly in urogenital and gastrointestinal systems, with highest levels in testis. It’s expression in CNS is low, but the protein has been detected in cerebellum, hippocampus, dorsal root ganglia and spinal cord. NMUR2 is predominantly detected in central nervous system with highest levels detected in medulla oblongata, spinal cord and thalamus. It may also be detected in testis but has low levels of expression in peripheral tissues.
