IL-32 Antibody: Interleukin-32 (IL-32) was initially identified as a transcript (NK4) that is selectively expressed in lymphocytes and NK cells and whose expression is increased following activation by IL-2. It was later re-isolated from an IL-18-treated lung carcinoma cell line and re-named IL-32. IL-32 is unusual in that it does not share sequence homology with known cytokine families and is highly expressed in immune tissues, existing in at least four differentially spliced isoforms. Because treatment of human monocytic and mouse macrophage cells with IL-32 induces several proinflammatory cytokines such as TNF-alpha , IL-8 and MIP-2, and because it is also induced in human peripheral lymphocyte cells after mitogen stimulation and in epithelial cells by IFN gamma , it has been suggested that IL-32 may play a role in autoimmune and inflammatory diseases such as rheumatoid arthritis.

AU-specific RNA-binding enoyl-CoA hydratase (AUH) protein binds to the AU-rich element (ARE), a common element found in the 3' UTR of rapidly decaying mRNA such as c-fos, c-myc and granulocyte/ macrophage colony stimulating factor. ARE elements are involved in directing RNA to rapid degradation and deadenylation. AUH is also homologous to enol-CoA hydratase, an enzyme involved in fatty acid degradation, and has been shown to have intrinsic hydratase enzymatic activity. AUH is thus a bifunctional chimera between RNA binding and metabolic enzyme activity. A possible subcellular localization in the mitochondria has been demonstrated for the mouse homolog of this protein which shares 92% identity with the human protein. It has been suggested that AUH may have a novel role as a mitochondrial located AU-binding protein. SPB is an amine-reactive and photoreactive heterobifunctional crosslinking reagent. The photoreactive psoralen group provides much greater covalent insertion yields than phenyl azide-containing compounds, which makes this reagent a superior alternative to typical photoreactive NHS-ester crosslinkers. The psoralen tricyclic planar ring system intercalates into double-stranded, and to a lesser extent, single-stranded, DNA, RNA, PCR products, cDNA or oligonucleotides. It also can be used with agarose gel-purified oligonucleotides because the presence of agarose does not interfere with the coupling reaction. The nucleic acid labeling site of the psoralen group (the 5,6 double bond in thymine residues) does not interfere with subsequent hybridization reactions, creating a highly-sensitive direct labeling alternative to radioactive probes.

SMPT is a heterobifunctional sulfhydryl- and amine-reactive cross-linker. SMPT is used often for conjugating a toxin molecule to a monoclonal antibody directed against a cell-surface antigen. These immunotoxins produced with SMPT are highly potent as the cleavable disulfide imparts increased cytotoxicity compared to conjugates without a cleavable bond, such as those produced with SPDP. Also, next to the pyridine-2-thione in the spacer arm are a benzene ring and a methyl group adjacent to a carbon that hinders the disulfide bond, allowing exceptional conjugate stability in vivo. SMPT has the added benefit of being more stable to hydrolysis in aqueous solutions than other NHS cross-linkers.

Novel influenza virus strains caused by genetic drift and viral recombination emerge periodically to which humans have little or no immunity, resulting in devastating pandemics. Influenza A can exist in a variety of animals; however it is in birds that all subtypes can be found. These subtypes are classified based on the combination of the virus coat glycoproteins hemagglutinin (HA) and neuraminidase (NA) subtypes. During 1997, an H5N1 avian influenza virus was determined to be the cause of death in 6 of 18 infected patients in Hong Kong. There was some evidence of human to human spread of this virus, but it is thought that the transmission efficiency was fairly low. HA interacts with cell surface proteins containing oligosaccharides with terminal sialyl residues. Virus isolated from a human infected with the H5N1 strain in 1997 could bind to oligosaccharides from human as well as avian sources, indicating its species-jumping ability.

The PDPH is a heterobifunctional crosslinker containing sulfhydryl-reactive pyridyldithiol and carbonyl-reactive hydrazide moieties. Pyridyldithiols react with free sulfhydryls (-SH) to form disulfide bonds. Hydrazide groups react with carbonyls (aldehydes and ketones) to form stable hydrazone bonds. Aldehyde groups can be created by periodate-oxidation of sialic acid and other sugar components of glycoprotein polysaccharides. Thus, PDPH is useful for conjugating glycoproteins and sulfhydryl-containing peptides or proteins. Likewise, PDPH is useful as a sulfhydryl-addition reagent for glycoproteins and other carbohydrates; after reaction of the hydrazide to an oxidized carbohydrate, the pyridyldithiol group can be cleaved by a reducing agent to expose a sulfhydryl group. Yet another application for PDPH is to react the primary amine of the hydrazide moiety to a carboxyl group using the crosslinker EDC.

IL-33 Antibody: Interleukin-33 (IL-33) is a recently identified member of the IL-1 family of cytokines whose other members include IL-1 alpha /beta, IL-1Ra and IL-18. Its receptor has been shown to be ST2, an IL-1 receptor family member that also acts as a negative regulator of TLR-IL-1R signaling and IL-1R accessory protein (IL-1RAcP). Receptor binding of IL-33 activates NF-kappa B and MAP kinases and induces the expression of TH2-associated cytokines such as IL-4, IL-5 and IL-6. Prolonged IL-33 treatment of mice led to the development of eosinophilia, splenomegaly, and severe pathological changes in mucosal organs such as lungs, esophagus and small intestine. Recent experiments have shown that IL-33 can also co-localize with heterochromatin and possesses transcriptional repressor activities, indicating that IL-33 may function as both a proinflammatory cytokine and an intracellular nuclear factor with transcriptional regulatory properties.
