This gene encodes a member of the mitogen-activated protein kinase (MAPK) family. Members of this family act as an integration point for multiple biochemical signals and are involved in a wide variety of cellular processes such as proliferation, differentiation, transcription regulation, and development. They form a three-tiered signaling module composed of MAPKKKs, MAPKKs, and MAPKs. This protein is phosphorylated at serine and threonine residues by MAPKKKs and subsequently phosphorylates downstream MAPK targets at threonine and tyrosine residues. A similar protein in mouse has been reported to play a role in liver organogenesis. A pseudogene of this gene is located on the long arm of chromosome X. Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDH21 (protocadherin 21), also known as PRCAD, is an 859 amino acid single-pass membrane protein that localizes to the outer segments of photoreceptor cells and contains six cadherin domains. Existing as multiple alternatively spliced isoforms, PCDH21 functions as a calcium-dependent cell adhesion protein that is thought to be required for the structural integrity of photoreceptor cells and may be involved in the formation and maintenance of neuronal networks.

A family of integral membrane proteins are involved in taste perception and include T1R, which is involved in sweet taste perception and T2R, which is involved in bitter taste perception. Both types of taste receptors couple to various G proteins to initiate signal transduction cascades. Specifically, T2R50 is expressed in subsets of taste receptor cells of the tongue and exclusively in gustducin-positive cells.

Adenosine is involved in a variety of processes, including the synthesis of urea, the anti-inflammatory response, and the inhibition of protein synthesis. The Adenosine receptors, including Adenosine A1-R, Adenosine A2A-R, Adenosine A2B-R and Adenosine A3-R, are integral membrane proteins that are members of the G protein-coupled receptor family. Adenosine A1-R mediates ureagenesis in a partially calcium-dependent manner. Adenosine is known to mediate coronary vasodilation via Adenosine A2A-R. Collagen synthesis and total protein synthesis are inhibited in certain cells by Adenosine, acting via the A2B receptors. Activation of Adenosine A3-R inhibits the induction of TNF?and blocks the endotoxin CD14 receptor signal transduction pathway.

The correlation of anti-sperm with cases of unexplained infertility implicates a role for these in blocking fertilization. Improved diagnosis and treatment of immunologic infertility, as well as identification of proteins for targeted contraception, are dependent on the identification and characterisation of relevant sperm antigens. The protein expressed by this gene is recognised by anti-sperm from an infertile man. This protein localizes to the tail of permeabilized human sperm and contains eight contiguous armadillo repeats, a motif known to mediate protein-protein interactions. Studies in mice suggest that this protein is involved in sperm flagellar motility and maintenance of the structural integrity of mature sperm. Several transcript variants encoding different isoforms have been found for this gene.

Nidogens are highly conserved proteins present in vertebrate and invertebrate basement membranes. Nidogens connect the Laminin and Collagen IV networks and integrate other proteins into the membrane. In mammals, two Nidogen proteins, Nidogen and Nidogen-2, interact at comparable levels with Collagen I, IV and Perlecan, serving to stabilize basement membranes and playing a major role in embryogenesis. The two isoforms have a similar shape, consisting of three globular domains, and co-localize in vessel walls and other basement membrane zones. Nidogen-2 is a cell adhesion protein glycosylated at nitrogen and oxygen sites, and is widely distributed in basement membranes in heart, placenta, bone and, to a lesser extent, in pancreas, kidney and skeletal muscle.
