Epithelial Cell Adhesion Molecule (EpCAM) is a signal type I transmembrane glycoprotein that belongs to the EPCAM family. EpCAM is composed of an extracellular domain with one thyroglobulin type-1 domain, a transmembrane domain and a cytoplasmic domain. EpCAM is found on the surface of adenocarcinoma, but not on mesodermal or neural cell membranes. The EpCAM molecule has been shown to function as a homophilic Ca2+ independent adhesion molecule. It may act as a physical homophilic interaction molecule between intestinal epithelial cells (IECs) and intraepithelial lymphocytes (IELs) at the mucosal epithelium as an immunological barrier providing the first line of defence against infection. Defects in EPCAM are a cause of hereditary non-polyposis colorectal cancer type 8 (HNPCC8) and diarrhoea type 5 (DIAR5). EpCAM plays a role in embryonic stem cells proliferation and differentiation; it up-regulates the expression of FABP5, MYC and Cyclin A and Cyclin E. It is highly and selectively expressed by undifferentiated embryonic stem cells.

The progression of cells through the cell cycle is regulated by a family of protein kinases known as cyclin-dependent kinases (Cdks). The sequential activation of individual members of this family and their consequent phosphorylation of critical substrates promotes orderly progression through the cell cycle. The cyclins function as differentially expressed positive regulators of Cdks. Negative regulators of the cycle include the p53-inducible 21 kDa WAF1/Cip1 protein designated p21, Kip1 p27 and p16. The complexes formed by Cdk4 and the D-type cyclins have been strongly implicated in the control of cell proliferation during the G1 phase. It has recently been shown that p16 binds to Cdk4 and inhibits the catalytic activity of the Cdk4/cyclin D complex. Moreover, the gene encoding p16 exhibits a high frequency of homozygous deletions and point mutations in established human tumor cell lines.

The individual APC/C components mRNA and protein levels are expressed at approximately the same levels in most tissues and cell lines, suggesting that they perform their functions as part of a complex. It has been suggested that APC10 plays a role to regulate the binding of specific substrates to the APC/C complex, similar to that of coactivators.

STEAP1 Antibody: The six-transmembrane epithelial antigen of prostate 1 (STEAP1) was the first member of a family of metalloreductases identified as cell-surface antigens in prostate tissue. The normal function of STEAP is still uncertain; unlike other members of the STEAP family, STEAP1 does not promote iron or copper reduction or uptake and lacks the FNO-like reductase domain critical for activity. However, its expression is highly increased in multiple cancer cell lines, including prostate, bladder, colon, and ovarian cancers. Supporting this is evidence that STEAP1 peptides can be used to stimulate CD8+ T cells from healthy donors, enabling them to recognize STEAP1-positive human tumor cells, suggesting that STEAP1 may a potential target for cancer immunotherapy. At least three isoforms of STEAP1 are known to exist.

MTA1 encodes a protein that was identified in a screen for genes expressed in metastatic cells, specifically, mammary adenocarcinoma cell lines. Expression of this gene has been correlated with the metastatic potential of at least two types of carcinomas although it is also expressed in many normal tissues. The role it plays in metastasis is unclear. MTA1 was initially thought to be the 70kD component of a nucleosome remodeling deacetylase complex, NuRD, but it is more likely that this component is a different but very similar protein. These two proteins are so closely related, though, that they share the same types of domains. These domains include two DNA binding domains, a dimerization domain, and a domain commonly found in proteins that methylate DNA. The profile and activity of this gene product suggest that it is involved in regulating transcription and that this may be accomplished by chromatin remodeling.

DEDAF Antibody: Apoptosis is related to many diseases and induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain (DD) death effector domain (DED), and caspase recruitment domain (CARD) containing molecules. Several molecules including caspases and adaptor FADD contain DEDs. A novel protein that interacts with DED of caspase-8 and 10, and FADD was identified recently and designated DEDAF for DED associated factor. DEDAF is identical to the transcriptional repressor RYBP. DEDAF/RYBP is expressed in multiple tissues and cell lines. DEDAF interacts with FADD and augments the formation of CD95/FADD/capase-8 complexes at the cell membrane, and interacts with DED-containing DNA biding protein (DEDD) in the nucleus indicating it is involved in the regulation of both cytoplasmic and nuclear events of apoptosis.

CD45RA is an isoform of the human leukocyte common antigen (CD45). Human CD45 contains three exons which encode peptide segments designated A, B and C, respectively. The differential splicing of the exons generates at least five isoforms, ABC, AB, BC, B and O. This antibody reacts with ABC and BC isoforms. CD45RA is expressed on 40-50% of peripheral CD4+ T-cells, 50% of peripheral CD8+ T-cells, B-cells, and leukemic B-cell lines. T-cells expressing CD45RA are naive or virgin T-cells. T-cells expressing CD45RO are memory T-cells. CD45RA and CD45RO define complementary, predominantly non-overlapping populations of resting peripheral T-cells. This MAb is useful in study on the subpopulation of CD4+ or CD8+ T-cells. It can especially be used to differentiate T-cell lymphomas (CD45RO +ve) from B cell lymphomas (CD45RA +ve).

IL-32 Antibody: Interleukin-32 (IL-32) was initially identified as a transcript (NK4) that is selectively expressed in lymphocytes and NK cells and whose expression is increased following activation by IL-2. It was later re-isolated from an IL-18-treated lung carcinoma cell line and re-named IL-32. IL-32 is unusual in that it does not share sequence homology with known cytokine families and is highly expressed in immune tissues, existing in at least four differentially spliced isoforms. Because treatment of human monocytic and mouse macrophage cells with IL-32 induces several proinflammatory cytokines such as TNF-alpha , IL-8 and MIP-2, and because it is also induced in human peripheral lymphocyte cells after mitogen stimulation and in epithelial cells by IFN gamma , it has been suggested that IL-32 may play a role in autoimmune and inflammatory diseases such as rheumatoid arthritis.

Phosphorylation of these three tyrosine residues is necessary for kinase activation.Insulin receptors (IRs) share significant similarity with IGF1 receptors in both structure and function,including an equivalent triple tyrosine cluster within the activation loop of the kinase domain (Tyr1146, Tyr1150 and Tyr1151).Tyrosine autophosphorylation of insulin receptor is one of the earliest cellular responses to insulin stimulation. Autophosphorylation begins with phosphorylation of Tyr1146 and either Tyr1150 or Tyr1151. Full kinase activation requires the triple tyrosine phosphorylation.

GFR alpha 1 Antibody: Glial cell line-derived neurotrophic factor (GDNF) is a potent survival factor for central and peripheral neurons and is essential for the development of kidneys and the enteric nerves system. Physiological responses to GDNF require the presence of a novel glycosylphosphadidylinositol linked protein GDNFRalpha, which is a cell surface receptor for GDNF. The cDNAs encoding GDNFRalpha from human, rat, chicken and mouse have been cloned recently. GDNFRalpha was also termed Ret ligand 1 (RETL1) or TGF-beta-related neurotrophic factor receptor 1 (TrnR1) and nominated as GFR alpha -1 recently. GFR alpha -1 binds GDNF specifically and mediates activation of the Ret protein tyrosine kinase (PTK). Thus, GDNF, GFR alpha and the Ret PTK form a complex to transduce GDNF signal and to mediate GDNF function.

RET (ret proto-oncogene) is a member of the cadherin superfamily and a receptor tyrosine kinase, which are cell-surface molecules that transduce signals for cell growth and differentiation. It can undergo oncogenic activation in vivo and in vitro by cytogenetic rearrangement. Ligands that bind the Ret receptor include the glial cell line-derived neurotropic factor (GDNF) and its congeners neurturin, persephin and artemin. Alterations in the corresponding Ret gene are associated with diseases including papillary thyroid carcinoma, multiple endocrine neoplasia (type 2A and 2B), familial medullary thyroid carcinoma and a congenital developmental disorder known as Hirschsprung’s disease. The Tyr905 residue located in the Ret kinase domain plays a crucial role in Ret catalytic and biological activity. Substitution of Phe for Tyr905 dramatically inhibits Ret autophosphorylation activity.
