APC5 Antibody: Cell cycle regulated protein ubiquitination and degradation within subcellular domains is thought to be essential for the normal progression of mitosis. APC5 is a highly conserved component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle. APC/C is responsible for degrading anaphase inhibitors, mitotic cyclins, and spindle-associated proteins ensuring that events of mitosis take place in proper sequence. The individual APC/C components mRNA and protein levels are expressed at approximately the same levels in most tissues and cell lines, suggesting that they perform their functions as part of a complex. While little is known of APC5, it is thought that APC5 associates with other APC/C components APC1, APC4, and CDC23 interdependently, such that loss of any one subunit reduces binding between the remaining three.

Nanos1 Antibody: Nanos1 is one of three known mammalian homologs to the Drosophila gene nanos. Nanos1 is an RNA-binding protein containing a zinc-finger motif and is expressed in the developing nervous system and continues in the adult brain. Interestingly, unlike mice deficient in either nanos2 or nanos3, mice lacking the nanos1 gene develop normally with no sign of abnormalities. Recently it has been found that expression of nanos1 mRNA is down-regulated by E-cadherin in a human breast cancer cell line and the amino-terminal domain on Nanos1 interacts with the E-cadherin-binding protein p120ctn. Furthermore, overexpression of Nanos1 in human colorectal DLD1 cancer cells functionally abolished cell-cell adhesion, allowing the cancer cells to develop strong migratory and invasive properties. These results suggest that targeting Nanos1 might prove an effective strategy in the treatment of E-cadherin-negative tumors.

Programmed death ligand 1 (PD-L1, B7-H1 or CD274) is a member of the growing B7 family of immune proteins that provide signals for both stimulating and inhibiting T cell activation. CD274 has been identified as one of two ligands for programmed death 1 (PD-1), a member of the CD28 family of immunoreceptors. CD274 is widely expressed in several organs such as heart, skeletal muscle, placenta and lung, and in lower amounts in thymus, spleen, kidney and liver. CD274 expression is upregulated in a small fraction of activated T and B cells and a much larger fraction of activated monocytes. CD274 expression is also induced in dendritic cells and keratinocytes after IFN-gamma stimulation. CD274 expression is also upregulated in a variety of tumour cell lines. Interaction of CD274 with PD-1 results in inhibition of TCR mediated proliferation and cytokine production, suggesting an inhibitory role in regulating immune responses. The CD274 - PD-1 pathway is involved in the negative regulation of some immune responses and may play an important role in the regulation of peripheral tolerance.

Gamma-aminobutyric acid (GABA) is the primary inhibitory neurotransmitter in the central nervous system. There are two major classes of GABA receptors: the GABAA and the GABAB subtype of receptors. GABAA-Rs are important therapeutic targets for a range of sedative, anxiolytic, and hypnotic agents and are implicated in several diseases including epilepsy, anxiety, depression, and sub-stance abuse. The GABAA-R is a multimeric subunit complex. To date six alphas, four betas and four gamma;s, plus alternative splicing variants of some of these subunits, have been identified (Olsen and Tobin, 1990; Whiting et al., 1999; Ogris et al., 2004). Injection in oocytes or mammalian cell lines of cRNA coding for alpha- and beta-subunits results in the expression of functional GABAA-Rs sensitive to GABA. However, coexpression of a gamma-subunit is required for benzodiazepine modulation. It has recently been suggested that PKCepsilon regulates the sensitivity of GABAA alpha1beta2gamma2 receptors to ethanol and benzodiazepines through phosphorylation of serine 327 in the large intracellular loop of gamma2 (Qi et al., 2007)
