SERPINB10 is a serine proteinase inhibitor of the ovalbumin like B clade of serpins. It was first discovered in human bone marrow in a search for serpins similar to SerpinB6. SERPINB10 was identified in lymphocytes, and was elevated in chronic myeloid leukemia and chronic myelomonocytic leukemia cells. The monocytic cell lines THP-1 and AML-193 also showed elevated SERPINB10 expression.

This mAb recognises an extracellular epitope of an integral membrane glycoprotein of 150 kDa, identified as CD13. This antigen is present on most cells of myeloid origin including granulocytes, monocytes, mast cells, and GM-progenitor cells. It is also expressed by the majority of Aml, Cml in myeloid blast crisis, and in a smaller fraction of lymphoid leukemias. It is absent from normal lymphocytes, platelets and erythrocytes. CD13 is also present on fibroblasts; endothelial cells, epithelial cells from renal proximal tubules and intestinal brush border, bone marrow stromal cells, osteoclasts, and cells lining bile duct canaliculi. CD13 is identical to aminopeptidase N (APN), a prominent membrane-bound metalloprotease present on the surface of intestinal brush border and renal tubules. CD13 plays a role in metabolism of biologically active peptides, in phagocytosis, and in bactericidal/tumoricidal activities. It also serves as a receptor for human coronaviruses (HCV). The lineage-restricted pattern of expression of CD13 within the hemopoietic compartment suggests that it may be important in myeloid cell differentiation.

Interleukins (ILs) are a large group of cytokines that are produced mainly by leukocytes, although some are produced by certain phagocytes and auxiliary cells. ILs have a variety of functions, but most function to direct other immune cells to divide and differentiate. Each IL acts on a specific, limited group of cells through a receptor specific for that IL. Human IL10 is a non glycosylated polypeptide consisting of 178 amino acids. There is 73% homology between the human and mouse IL10 proteins, however, the human IL10 acts on both human and mouse target cells, while the mouse IL10 has species specific activity. The cellular sources of IL10 are CD4+ T cells and T cell clones, thymocytes, B cells and B cell lymphomas, macrophages, mast cell lines and keratinocytes. IL10 will stimulate the growth of stem cells, mast cells and thymocytes. IL10 enhances cytotoxic T cell development, and costimulates B cell differentiation and immunoglobulin secretion. IL10 inhibits cytokine production by macrophages and suppresses macrophage class II MHC expression. The human IL10 gene is on human chromosome 1.

IL-22 Receptor Antibody: A novel cytokine, designated IL-TIF for IL-10 related T cell-derived inducible factor and IL-22, was recently identified. The receptor for IL-22 (IL-22R, also termed CRF2-9 and IL-TIF-R1 chain) is a new member of the class II cytokine receptor family. IL-22R forms a complex with IL-10 receptor beta chain and mediates IL-22 signaling. IL-22 and its receptor activate JAK-STAT signaling pathway. IL22R is expressed in normal liver and kidney and their cell lines HepG2 and TK-10. A soluble form of IL-22 receptor, also termed IL-22 binding protein (IL-22BP) and IL-22 receptor-alpha 2 (IL-22RA2), was identified very recently. IL-22BP prevents binding of IL-22 to the functional cell surface IL-22R complex and neutralizes IL-22 activity. LPS induces IL-22 expression, which indicates the role of IL-22 in inflammatory response.

HLA-A, with HLA-B and HLA-C, belongs to major histocompatibility complex (MHC) class I antigens and expresses constitutively on all nucleated cells. MHC class I antigens play a role in class I MHC-associated antigen presentation, inhibition of NK cell cytotoxicity, tumor surveillance, and tissue allotransplantation. This mAb is useful for HLA molecular typing of peripheral blood leukocytes as well as a large number of leukemic cell lines. It reacts with an intralocus determinant present on a limited number of HLA-A locus-encoded gene products (HLA-A2, -A3, -A28, -A29, -A30, -A31 and Aw33). Its epitope maps between aa65-to-aa80 of the alpha1 domain of the HLA-A. This mAb recognises an intralocus determinant present on a limited number of HLA-A locus-encoded gene products (HLA-A2, -A3, A28, -A29, -A30, -A31 and -Aw33). Furthermore, by testing its reactivity with HLA-A2 natural variants and mutants, the importance of amino acid residues 79 and/or 80 of the alpha1 domain was demonstrated in the formation of an intralocus HLA-A determinant.
