SCUBE3 is a novel secreted 993 amino acid cell-surface osteoblast protein that plays an important role in bone cell biology. While highly expressed in osteoblasts, SCUBE3 expression is unobservable or very low in non-bone tissues. SCUBE3 forms homo-oligomers and hetero-oligomers with SCUBE1, and may undergo C-terminal proteolytic cleavage or become N-glycosylated following translation. Two SCUBE3 isoforms exist as a result of alternative splicing events. SCUBE3 contains one CUB domain and nine EGF-like domains, and is encoded by a gene which maps to human chromosome 6p21.31, a region associated with a rare form of metabolic bone disease known as Paget's disease. Chromosome 6 contains 170 million base pairs and comprises nearly 6% of the human genome.

In the case of Src p60, Y527 is this tyrosine; however, a mutant form of c-Src in which Y527 is replaced by phenylalanine is transforming and displays 5- to 10-fold elevated kinase activity compared to its normal counterpart. Csk has been identified as a Src-related tyrosine kinase having both SH2 and SH3 domains and a catalytic domain but lacking sequences amino terminal to the SH3 domain as well as carboxy terminal regulatory sequences. LRFN1 is a 771 amino acid single-pass type I membrane protein that belongs to the LRFN family. Containing a fibronectin type-III domain, an Ig-like (immunoglobulin-like) domain, a LRRCT domain, a LRRNT domain and seven LRR (leucine-rich) repeats, LRFN1 is thought to promote neurite outgrowth in hippocampal neurons and is involved in the regulation and maintenance of excitatory synapses. LRFN1 forms heteromeric complexes with LRFN2, LRFN3, LFRN4 and LFRN5, but does not have the ability to form homomeric complexes across cell junctions of adjacent cells like other LRFN family members. The PDZ-binding motif of LRFN1 is required for neurite outgrowth promotion and for SAP 97-, NE-dlg- and PSD-95-binding. LRFN1 is encoded by a gene located on human chromosome 19q13.2 and mouse chromosome 7 A3.
