The DPF3 protein, also known as Cerd4, is a member of the d4 gene family of transcription modulators that also includes DPF1/Neud4 and DPF2/Requiem. DPF3 has been shown to be a epigenetic key factor for heart and muscle development and can bind to methylated and acetylated lysine residues of histone 3 and 4, suggesting that DPF3 may play a role in recruiting chromatin remodeling complexes to acetylated histones. Two isoforms of DPF3, DPF3a and DPF3b, are required as transcriptional co-activators in SWI/SNF complex-dependent activation of the NF-kappaB RelA/p50 heterodimer.

The CD4 antigen is highly expressed on human T helper cells and thymocytes, and at lower levels on monocytes and dendritic cells. This integral membrane glycoprotein of approximately 58 kDa contains four external domains (D1 to D4) that show homology to members of the immunoglobulin (Ig) superfamily. It is responsible for the recognition of the MHC class II antigen. The CD4 antibody recognizes most thymocytes and about 65% of peripheral blood T cells.

UBE2D4 regulate of protein metabolic process. UBE2D4 accepts ubiquitin from the E1 complex and catalyzes its covalent attachment to other proteins. In vitro, UBE2D4 able to promote polyubiquitination using all 7 ubiquitin Lys residues, but may prefer 'Lys-11' and 'Lys-48'-linked poly-ubiquitination.

Lymphocyte-activation gene 3 (LAG3), also known as CD223, is a type I transmembrane protein with four extracellular Ig-like domains, designated D1 to D4 and belongs to the immunoglobulin superfamily. The gene for LAG3 lies adjacent to the gene for CD4 on human chromosome 12p13.32 and shares approximately 20% identical to the CD4 gene. LAG3 is expressed on activated T cells, natural killer cells, B cells and plasmacytoid dendritic cells. LAG3 binds with high affinity to MHC class II molecules, and it interferes competitively with the binding of CD4 to MHC class II and thereby blocks the transduction of stimulatory signals mediated by this interaction. LAG3 negatively regulates cellular proliferation, activation, and homeostasis of T cells, and plays an important role in Treg suppressive function. LAG3 is the target of various drug development programs to develop new treatments for cancer and autoimmune disorders. The soluble form, sLAG-3, is being developed as a cancer drug.
