ACAA1 is a 424 amino acid member of the thiolase family of enzymes and is involved in lipid metabolism. Localized to the peroxisome, ACAA1 catalyzes the conversion of acyl-CoA and acetyl-CoA to 3-oxoacyl-CoA in the fatty acid oxidation pathway. ACAA1 shows high enzymatic activity in liver, kidney, intestine and white adipose tissue in rats, where it exists as two types, namely type A and type B. Human ACAA1 shares 86% amino acid identity with its rat counterpart, suggesting a conserved function for ACAA1 among different species.

ARHI contains a highly conserved GTP-binding domain, a putative effector domain distinct from that of RAS and RAP proteins, and a C-terminal membrane localization motif. ARHI mRNA is detected in all normal breast and ovarian epithelial cell cultures tested, as well as in normal ovary, heart, liver, pancreas and brain however ARHI expression is down-regulated in breast and ovarian tumors. Reactivation of ARHI expression in breast cancer cells is associated with increased histone H3 acetylation and decreased lysine 9 methylation of histone H3.

This gene encodes a transcription factor that is a member of the leucine zipper family of DNA binding proteins. This protein binds to the cAMP-responsive element (CRE), an octameric palindrome. It forms a homodimer or a heterodimer with c-Jun and stimulates CRE-dependent transcription. This protein is also a histone acetyltransferase (HAT) that specifically acetylates histones H2B and H4 <i>in vitro</i>; thus it may represent a class of sequence-specific factors that activate transcription by direct effects on chromatin components.
