Phosphorylase b kinase catalyzes the phosphorylation of serine in certain substrates, including troponin I. Polymer of 16 chains, four each of alpha, beta, gamma, and delta. Alpha and beta are regulatory chains, gamma is the catalytic chain, and delta is calmodulin. The two calmodulin-binding domains appear to act in concert to bind a single molecule of calmodulin and are pseudosubstrate/autoinhibitory domains.

TM4-B, also known as TSPAN16 (tetraspanin-16) or TM4SF16 (transmembrane 4 superfamily member 16), is a 245 amino acid multi-pass membrane protein that belongs to the tetraspanin (TM4SF) family. Members of the TM4SF family mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. TM4-B is broadly expressed in most human tissues and cell lines including neural and bone marrow derived tissues. The gene that encodes TM4-B contains nearly 31,000 bases and maps to human chromosome 19p13.2. Chromosome 19 consists of over 63 million bases, houses approximately 1,400 genes and is recognized for having the greatest gene density of the human chromosomes. It is the genetic home for a number of immunoglobulin (Ig) superfamily members, including the killer cell and leukocyte Ig-like receptors, a number of ICAMs, the CEACAM and PSG families and Fc receptors (FcRs).

Poly(ADP-ribosylation) is a method of DNA damage-dependent posttranslational modification that helps to rescue injured proliferating cells from cell death. The PARP (poly(ADP-ribose) polymerase) proteins comprise a superfamily of enzymes that functionally modify histones and other nuclear proteins, thereby preventing cell death. PARPs use NAD+ as a substrate to catalytically transfer ADP-ribose residues onto protein acceptors; a process that, when repeated multiple times, leads to the formation of poly(ADPribose) chains on the protein. The presence of these chains alters the function of the target protein and promotes cell survival. PARP proteins are implicated in a variety of diseases, including cancer, neurodegenerative and inflammatory disorders. PARP-16 is a 322 amino acid poly (ADP-ribose) polymerase protein localized to the membrane. Expressed as three isoforms produced by alternative splicing, PARP-16 contains one PARP catalytic domain.

Members of the EF-CBP (N-terminal EF-hand calcium binding protein)/NECAB (neuronal calcium-binding protein) family participate in neuronal calcium signaling. EF-CBP2, also known as NECAB2 (N-terminal EF-hand calcium binding protein 2), neuronal calcium-binding protein 2 or synaptotagmin-interacting protein 2 (Stip-2), is a 386 amino acid cytoplasmic protein that contains one antibiotic biosynthesis monooxygenase (ABM) domain and two EF-hand domains. Expressed in brain, EF-CBP2 is suggested to bind metabotropic glutamate receptor 5 (mGluR-5) in a calcium-regulated manner. The gene encoding EF-CBP2 maps to human chromosome 16, which encodes over 900 genes and comprises nearly 3% of the human genome. The rare disorder Rubinstein-Taybi syndrome is also associated with chromosome 16, as is Crohn's disease, which is a gastrointestinal inflammatory condition.
