Glutathione is the major low molecular weight thiol compound of the living plant or animal cell. It is a tripeptide with a gamma peptide linkage between the amine group of cysteine (which is attached by normal peptide linkage to a glycine) and the carboxyl group of the glutamate side-chain. It is an antioxidant, preventing damage to important cellular components caused by reactive oxygen species such as free radicals and peroxides. The sulfhydryl (thiol) group (SH) of cysteine serves as a proton donor and is responsible for the biological activity of glutathione. Glutathione suppresses human immunodeficiency virus expression in chronically infected monocytic cells. It is a useful tripeptide involved in many aspects of metabolism, including transport of g-glutanyl amino acids and reductive cleavage of disulfide bonds. Endogenous antioxidant that plays a major role in reducing reactive oxygen species formed during cellular metabolism and the respiratory burst. Glutathione-S-transferase catalyzes the formation of glutathione thioethers with xenobiotics, leukotrienes, and other molecules that have an electrophilic center. Glutathione also forms disulfide bonds with cysteine residues in proteins. Via these mechanisms, it can have the paradoxical effect of reducing the efficacy of anti-cancer agents.

KEAP1 Antibody: KEAP1 (kelch-like ECH-associated protein 1) is a stress sensing adaptor for the Cullin3 (Cul3)-dependent E3 ubiquitin ligase complex that negatively regulates NRF2 (NF-E2-related factor 2) and plays a role in the oxidative stress response. It targets NFE2L2/NRF2 for ubiquitination and degradation by the proteasome. KEAP1 contains an amino terminal BTB/POZ domain and a carboxyl terminal KELCH domain which are required for interaction with NRF2, and in binding Cul3-E3 ubiquitin ligase. Altered expression of NRF2 is associated with chronic obstructive pulmonary disease (COPD). KEAP1 also targets the down regulation of NF-kappa B activity by targeting IKK beta degradation. Mutation of the KEAP1 gene is found in lung cancer.

GRASP (GRP1-associated scaffold protein, tamalin) is a 395 amino acid proteinencoded by the human gene GRASP. GRASP is a scaffold protein that com-prises multiple protein-interacting domains, including a 95 kDa postsynapticdensity protein (PSD-95)/discs-large/ZO-1 (PDZ) domain, a leucine-zipperregion and a carboxyl-terminal PDZ-binding motif. GRASP is involved withintracellular trafficking and contributes to the macromolecular organizationof group 1 metabotropic glutamate receptors (mGluRs) at synapses. GRASPforms a heteromer composed of GRASP , PSCD2 and at least one mGluR-1. Italso interacts with PSCD3, mGluR-2, mGluR-3 and mGluR-5. GRASP is highlyexpressed in brain and has lower levels of expression in lung, heart, embryo, kidney and ovary.
