DEAD-box proteins, characterized by the conserved motif Asp-Glu-Ala-Asp, are putative RNA helicases implicated in several cellular processes involving modifications of RNA secondary structure and ribosome/spliceosome assembly. Based on their distribution patterns, some members of this family may be involved in embryogenesis, spermatogenesis, and cellular growth and division. DDX15 (DEAH-box protein 15), also known as DHX15, DBP1 or HRH2, is a nuclear ATP-dependent RNA helicase that is involved in pre-mRNA splicing and is a member of the DEAH-box subfamily of DEAD-box proteins. Expressed throughout the body, DDX15 is a pre-mRNA processing factor that plays a role in spliceosome disassembly after the release of mature mRNA. When localized to the nucleoli, DDX15 is thought to interact with the La/SSB autoantigen, an RNA chaperone that functions in various intracellular processes. DDX15 is 795 amino acids in length and is the human ortholog of the S. cerevisiae protein Prp43. helicase 2.

RORET, also known as RING finger protein 15 (RNF15) or zinc finger protein RoRet, is a 465 amino acid member of the TRIM family, also known as the RING-B-box coiled-coil (RBCC) family. Members of the RBCC family have an N-terminal RING finger, followed by one or two zinc-binding domains (B-box domains), a leucine coiled-coil region and a variable C-terminal domain. Found in all eukaryotes, members of the RBCC family typically function within a larger protein complex and possess ubiquitin-protein isopeptide ligase activity.The protein encoded by this gene is a member of the tripartite motif (TRIM) family. The TRIM motif includes three zinc-binding domains, a RING, a B-box type 1 and a B-box type 2, and a coiled-coil region. The function of this protein has not been identified.

Interleukin 15 (IL-15) is a widely expressed proinflammatory cytokine that is structurally and functionally related to interleukin 2 (IL-2). IL-15 signals through JAK kinases to activate STAT3, STAT5, and STAT6 transcription factors. IL-15 regulates the activation of T cells, neutrophils, and macrophages, and is a stimulatory cytokine promoting dendritic cell function. IL-15 expression is dysregulated in chronic inflammatory disorders, including rheumatoid arthritis, psoriasis, and pulmonary inflammatory diseases. Therefore, IL-15 may serve as an effective therapeutic target, due to the beneficial outcomes of IL-15 neutralisation in models of psoriasis and diabetes. Human IL-15 shows activity on mouse cells.

Modulation of the chromatin structure plays an important role in the regulation of transcription in eukaryotes. The nucleosome, made up of four core histone proteins (H2A, H2B, H3 and H4), is the primary building block of chromatin. The N-terminal tail of core histones undergoes different posttranslational modifications including acetylation, phosphorylation and methylation. These modifications occur in response to cell signal stimuli and have a direct effect on gene expression. In most species, the histone H2B is primarily acetylated at lysines 5, 12, 15 and 20. Histone H3 is primarily acetylated at lysines 9, 14, 18 and 23. Acetylation at lysine 9 appears to have a dominant role in histone deposition and chromatin assembly in some organisms. Phosphorylation at Ser10 of histone H3 is tightly correlated with chromosome condensation during both mitosis and meiosis.

Recognizes an oncofetal antigen of 220 kDa, identified as a tumor-associated glycoprotein (TAG-72) with properties of a mucin. This MAb defines the mucin-carried sialylated-Tn epitope. TAG-72 is usually expressed by adenocarcinomas, but is negative in mesotheliomas. Studies have reported that this antibody has 80% sensitivity and 93% specificity for pulmonary adenocarcinoma. Therefore, TAG-72 is a useful marker to distinguish between mesothelioma and adenocarcinoma. However, false positive reactions can occur so results must be interpreted with the utmost caution. This antibody may be useful in the differentiation of non-small cell carcinomas from small cell carcinomas of the lung. The combined use of anti-TAG-72 and anti-GCDFP-15 is valuable in the diagnosis of apocrine carcinoma.
