Beschreibung:   Cadherins comprise a family of Ca++-dependent adhesion molecules that function to mediate cell-cell binding critical to the maintenance of tissue structure and morphogenesis. Cadherins each contain a large extracellular domain at the amino-terminus, which is characterised by a series of five homologous repeats, the most distal of which is thought to be responsible for binding specificity. The relatively short carboxy-terminal, intracellular domain interacts with a variety of cytoplasmic proteins, including beta-catenin, to regulate cadherin function. Cadherin-26 is a 852 amino acid single-pass transmembrane protein containing 4 cadherin domains through which it interacts with other cadherins in a homophilic manner. There are 4 named isoforms of cadherin-26 that are produced as a result of alternative spicing events.

Beschreibung:   Glut4 is a twelve pass transmembrane protein (12TM) whose carboxy-terminus may dictate its cellular localization. Aberrant Glut4 expression has been suggested to contribute to such maladies as obesity and diabetes. Glut4 null mice have shown that while functional Glut4 protein is not required for maintaining normal glucose levels, it is necessary for sustained growth, normal cellular glucose, fat metabolism and prolonged longevity. TUG (ASPL in humans) regulates the trafficking of glucose via Glut4. Full-length TUG forms a complex with Glut4 and in 3T3-L1 adipocytes and this complex is present in unstimulated cells and is disassembled by insulin. TUG acts by trapping endocytosed Glut4 and tethering it intracellularly. Insulin mobilizes this pool of retained Glut4 by releasing the tether.

Beschreibung:   HP1 alpha is a component of heterochromatin. It recognises and binds histone H3 tails methylated at Lys-9, leading to epigenetic repression. HP1 alpha may interact with lamin B receptor (LBR). This interaction can contribute to the association of the heterochromatin with the inner nuclear membrane. HP1 proteins are relatively small proteins (~25 kDa) with a conserved amino-terminal chromo domain and a structurally related carboxy-terminal motif, the chromo shadow domain. Both domains of HP1 are required for binding to native chromatin in vivo, but they contribute differentially to binding in euchromatin and heterochromatin.

Beschreibung:   EDD is a progestin-regulated gene that was isolated from T-47D human breast cancer cells. Based on sequence homology, EDD appears to be a human homolog of the Drosophila hyperplastic discs (hyd) gene, a tumor suppressor gene that is required for control of imaginal disc growth. EDD contains a HECT domain in the carboxy terminus. HECT domain-containing proteins function as ubiquitin-protein ligases, or E3 enzymes. EDD has been shown to bind to ubiquitin, and like other HECT family proteins, may function as an E3 ubiquitin-protein ligase.

Beschreibung:   FAKD3 is a member of a small family of Fas-activated serine/threonine kinase domain (FASTKD) containing proteins that share an amino terminal mitochondrial targeting domain and multiple carboxy terminal FAST domains as well as a putative RNA-binding RAP domain. The members of this family are ubiquitously expressed and are generally most abundant in mitochondria-enriched tissues such as heart, skeletal muscle and brown-adipose tissue. Some members of this protein family may play a role in apoptosis. The protein encoded by this gene interacts with components of the mitochondrial respiratory and translation networks. A pseudogene of this gene is also present on chromosome 5. Alternative splicing results in multiple transcript variants.

Beschreibung:   The P2X receptor family is comprised of ligand-gated ion channels that allow for the increased permeability of calcium into the cell in response to extracellular ATP. The seven P2X receptors, P2X1-P2X7, form either homomeric or heteromeric channels or both. They are characterised by intracellular amino- and carboxy-termini. P2X receptors are expressed in a wide variety of tissues, including neurons, prostate, bladder, pancreas, colon, testis and ovary. The major function of the P2X receptors is to mediate synaptic transmissions between neurons and to other tissues via the binding of extracellular ATP, which acts as a neurotransmitter. The P2X receptors may be involved in the onset of necrosis or apoptosis after prolonged exposure to high concentrations of extracellular ATP.

Beschreibung:   LPP (LIM containing lipoma preferred partner), is a scaffolding protein which contains three LIM domains at its carboxy terminus, preceded by a proline rich pre LIM region containing a number of protein interaction domains. LPP localizes to sites of cell adhesion, such as focal adhesions and cell-cell contacts and may be involved in cell-cell adhesion and cell motility. LPP also shuttles through the nucleus and may function as a transcriptional co-activator. The human LPP gene maps to chromosomal location 3q28, and preferentially translocates to the HMGIC gene in a subclass of human benign mesenchymal tumors known as lipomas. Alternate splicing results in multiple transcript variants.

Beschreibung:   First is a 297 amino acid protein that contains a putative carboxy-terminal coiled-coil domain. With highest expression in embryonic dorsal retina and, during later embryonic stages, the anterior epithelial cells of the lens, it is suspected that EURL may play a role in cell determination and differentiation. The gene encoding EURL maps to the long arm of chromosome 21, which houses approximately 300 genes and comprises nearly 1.5% of the human genome. Chromosome 21-associated disorders include Alzheimer's Disease, amyotrophic lateral sclerosis and, most notably, Down Syndrome (also known as trisomy 21).
