Beschreibung:   The downstream of kinase family (Dok-1-7) are members of a class of docking proteins that interact with receptor tyrosine kinases and, via this interaction, mediate biological responses within the body. Dok-4 (Downstream of kinase-4) is a 326 amino acid protein that contains one PH domain and one IRS-type PTB domain and belongs to the Dok family of interacting proteins. Expressed in a variety of tissues with highest expression in liver, heart, kidney and skeletal muscle, Dok-4 plays an important role in Ret-mediated neurite outgrowth and may link Ret with downstream effectors during neuronal differentiation. Additionally, Dok-4 is thought to play a positive role in the activation of MAPK pathways and may participate in T-cell induced immune system regulation. Overexpression of Dok-4 is associated with clear cell renal cell carcinoma, suggesting a role for Dok-4 in tumorigenesis.

Beschreibung:   AKAP associated sperm protein (encoded by the gene ROPN1L) is a sperm protein which interacts with A-kinase anchoring protein, AKAP3, through the amphipathic helix region of AKAP3. The Type II regulatory subunit of cAMP-dependent protein kinase (PKARII) also binds to this helical domain of AKAP3, allowing PKARII to be targeted to specific subcellular compartments. It is suggested that sperm contains several proteins that bind to AKAPs in a manner similar to PKARII, and this encoded protein may be one of them.

Beschreibung:   PDK1 (3 Phosphoinositide Dependent Protein Kinase 1) phosphorylates AGC kinases. PDK1 activates conventional PKC and PKC zeta through phosphorylation of critical threonine residues in the activation loop. PDK1 also phosphorylates Protein Kinase B (PKB) at threonine 308 in the presence of phosphatidylinositol-3,4,5-trisphosphate. Active Akt inactivates Glycogen Synthase Kinase 3 (GSK3), eventually leading to the dephosphorylation and activation of glycogen synthase and the stimulation of glycogen synthesis. Because of the role that PDK plays in insulin-induced glycogen synthesis and PKC activation it is a potentially important target for metabolic drug research. There are three named isoforms.

Beschreibung:   Heterotrimeric G protein-mediated signal transduction is a dynamically regulated process with the intensity of signal decreasing over time despite the continued presence of the agonist (1,2). G protein-coupled receptor kinases (GRKs) are activated by activated G protein-coupled receptors, and they function to phosphorylate and inactivate cell surface receptors in the heterotrimeric G protein signaling cascade (3,4). GIT1 (for GRK-interactor 1) and GIT2 are GTPase-activating proteins (GAP) for members of the ADP ribosylation factor (ARF) family of small GTP-binding proteins, which are involved in vesicular trafficking (5,6). GIT1 overexpression results in reduced internalization and resensitization of b2-adrenergic receptor, thus reducing b2-adrenergic receptor signaling (5).

Beschreibung:   ATF2 is a member of the ATF/CREB family of basic region leucine zipper DNA binding proteins that regulates transcription by binding to a consensus cAMP response element (CRE) in the promoter of various viral and cellular genes. Many of these genes are important in cell growth and differentiation, and in stress and immune responses. ATF2 is a nuclear protein that binds DNA as a dimer and can form dimers with members of the ATF/CREB and Jun/Fos families. It is a stronger activator as a heterodimer with cJun than as a homodimer. Several isoforms of ATF2 arise by differential splicing. The stable native full length ATF2 is transcriptionally inactive as a result of an inhibitory direct intramolecular interaction of its carboxy terminal DNA binding domain with the amino terminal transactivation domain. Following dimerization ATF2 becomes a short lived protein that undergoes ubiquitination and proteolysis, seemingly in a protein phosphatase-dependent mechanism. Stimulation of the transcriptional activity of ATF2 occurs following cellular stress induced by several genotoxic agents, inflammatory cytokines, and UV irradiation. This activation requires phosphorylation of two threonine residues in ATF2 by both JNK/SAP kinase and p38 MAP kinase. ATF2 is abundantly expressed in brain.

Beschreibung:   G-protein coupled opioid receptor that functions as receptor for the endogenous neuropeptide nociceptin. Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of down-stream effectors. Signaling via G proteins mediates inhibition of adenylate cyclase activity and calcium channel activity. Arrestins modulate signaling via G proteins and mediate the activation of alternative signaling pathways that lead to the activation of MAP kinases. Plays a role in modulating nociception and the perception of pain. Plays a role in the regulation of locomotor activity by the neuropeptide nociceptin.

Beschreibung:   The ProSAP family of proteins contain many protein-protein interaction domains and serve as scaffolding mediators within the post-synaptic density (PSD) of excitatory brain synapses. The PSD is an electron-dense structure underneath the post-synaptic plasma membrane of excitatory synapses that anchors and clusters glutamate receptors opposite to the pre-synaptic neurotransmitter release site. Shank proteins contain PDZ modular domains that coordinate the synaptic localization of ion channels, receptors, signaling enzymes, and cell adhesion molecules. The PDZ domain mediates protein-protein interactions via the recognition of a conserved sequence motif at the C-terminus of their target protein(s). ProSAPiP1 (proline rich synapse associated protein interacting protein 1) is a 673 amino acid protein that interacts with the PDZ domain of Shank 3. ProSAPiP1 expression is brain-specific with highest expression within the cerebellum, hippocampus and cerebral cortex.

Beschreibung:   This gene belongs to the superfamily of acid-sensing ion channels, which are proton-gated, amiloride-sensitive sodium channels. These channels have been implicated in synaptic transmission, pain perception as well as mechanoperception. This gene is predominantly expressed in the pituitary gland, and was considered a candidate for paroxysmal dystonic choreoathetosis (PDC), a movement disorder, however, no correlation was found between mutations in this gene and PDC. Beschreibung:   Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. The protocadherein gamma cluster consists of three subfamilies (A, B and C). As a member of the gamma subfamily B, PCDHGB1 (Protocadherin gamma B1) is a 927 amino acid protein that is one of 22 proteins encoded by the protocadherin gamma cluster. Typical of gamma protocadherins, PCDHGB1 contains six cadherin motifs and is a type I transmembrane receptor expressed in the central nervous system. With localization to synapses, members of the gamma cluster of protocadherins are essential for neuronal survival. There are two isoforms of PCDHGB1 that are produced as a result of alternative splicing events.
