Recognises an oncofetal antigen of 220 kDa, identified as a tumor-associated glycoprotein (TAG-72) with properties of a mucin. This mAb defines the mucin-carried sialylated-Tn epitope. TAG72 is usually expressed by adenocarcinomas, but is negative in mesotheliomas. Studies have reported that this antibody has 80% sensitivity and 93% specificity for pulmonary adenocarcinoma. Therefore, TAG72 is a useful marker to distinguish between mesothelioma and adenocarcinoma. However, false positive reactions can occur so results must be interpreted with the utmost caution. This antibody may be useful in the differentiation of non-small cell carcinomas from small cell carcinomas of the lung. The combined use of anti-TAG72 and anti-GCDFP-15 is valuable in the diagnosis of apocrine carcinoma.

TNF ligand-related molecule 1 is an anti-angiogenic cytokine that is also called Tumor necrosis factor superfamily, member 15 (TNFSF15) and Vascular endothelial growth inhibitor (VEGI/VEGI192A). This protein is abundantly expressed in endothelial cells, but is not expressed in either B or T cells. The expression of this protein is inducible by TNF and IL-1 alpha. This cytokine is a ligand for receptor TNFRSF25 and decoy receptor TNFRSF21/DR6. It can activate NF-kappaB and MAP kinases, and acts as an autocrine factor to induce apoptosis in endothelial cells. This cytokine is also found to inhibit endothelial cell proliferation, and thus may function as an angiogenesis inhibitor. Reduced expression of TL1A has been reported as a marker of poor prognosis in breast cancer.

The receptor for IL-21 (IL-21R, also termed NILR for novel Interleukin receptor) is a new member of the class I cytokine receptor family. IL-21R forms a complex with the common cytokine receptor gamma chain, gammac, and mediates IL-21 signaling. IL-21 and its receptor activate JAK-STAT signaling pathway. IL-21 is expressed in activated T cells, and HL-60 and THP-1 cell lines. IL-21 plays a role in the proliferation and maturation of NK, B and T cell populations.
