Penicillin refers to any member of beta-lactam antibiotics group. These agents are identified by a beta-lactam ring within their molecular structure. As the most widely used group of antibiotics available, beta-lactams are used for the treatment of bacterial infections usually caused by gram-positive organisms. Beta-lactam antibiotics are bactericidal, functioning to inhibit the synthesis of the peptidoglycan layer of bacterial cell walls. Bacterial penicillin-binding proteins and beta-lactamases constitute a large family of serine proteases that perform essential functions in the synthesis and maintenance of peptidoglycan cell wall. Notably, beta-lactamases cleave beta-lactams, therefore providing the bacteria with resistance to the antibiotic. Homologues of beta-lactamases occur in many species, including human, rat, cow, rabbit, pig, xenopus, zebrafish, and C. elegans. The human homologues, LACTB and LACTB2, are active-site-serine enzymes thought to be involved in metabolism.

This is mediated through serine and/or threonine phosphorylation of a range of downstream substrates. Over 100 substrate candidates have been reported so far, but for most of them, no isoform specificity has been reported. AKT3 is the least studied AKT isoform. It plays an important role in brain development and is crucial for the viability of malignant glioma cells. AKT3 isoform may also be the key molecule in up-regulation and down-regulation of MMP13 via IL13. Required for the coordination of mitochondrial biogenesis with growth factor-induced increases in cellular energy demands. Down-regulation by RNA interference reduces the expression of the phosphorylated form of BAD, resulting in the induction of caspase-dependent apoptosis.

Serine/threonine-protein kinase that plays an essential role in the regulation of actin filament dynamics. Acts downstream of several Rho family GTPase signal transduction pathways. Activated by upstream kinases including ROCK1, PAK1 and PAK4, which phosphorylate LIMK1 on a threonine residue located in its activation loop. LIMK1 subsequently phosphorylates and inactivates the actin binding/depolymerizing factors cofilin-1/CFL1, cofilin-2/CFL2 and destrin/DSTN, thereby preventing the cleavage of filamentous actin (F-actin), and stabilizing the actin cytoskeleton. In this way LIMK1 regulates several actin-dependent biological processes including cell motility, cell cycle progression, and differentiation. Phosphorylates TPPP on serine residues, thereby promoting microtubule disassembly. Stimulates axonal outgrowth and may be involved in brain development. Isoform 3 has a dominant negative effect on actin cytoskeletal changes. Required for atypical chemokine receptor ACKR2-induced phosphorylation of cofilin (CFL1).

Human SerpinB1, also designated monocyte/neutrophil elastase inhibitor (M/NEI) or leukocyte elastase inhibitor (LEI), is a cytoplasmic protein which acts as a fast-acting stoichiometric proteinase inhibitor that regulates the activity of neutrophil elastase (NE), cathepsin-G and proteinase-3. There are four homologous genes in mouse designated SerpinB1a, SerpinB1b, SerpinB1c and the pseudogene, Serpinb1-ps1. The three protein-coding genes share significant sequence identity, however SerpinB1a (also designated EIA) has been characterized as the functional ortholog of human SerpinB1.

A group of proteins that play a major role in this process are the serine/threonine protein phosphatases. Protein phosphatase (PP) holoenzyme is a trimeric complex that contains a regulatory subunit, a variable subunit and a catalytic subunit. Families of PP catalytic subunits include PP1, PP2A, PP2B, PP2C, PPX and PP5. Regulatory subunits include nuclear inhibitor of PP1 (NIPP1), PP1 nuclear targeting subunit (PNUTS), PP2A-A, PP2A-B, PP2A-B56, PP2A-C, PP2B-B and PR48. PNUTS, also designated CAT53 or FB19, is encoded by the gene PPP1R10. PNUTS acts as an inhibitor for the phosphatase activity of PP1 Alpha and PP1 Gamma. It is a nuclear protein primarily detected in nucleoplasmic bodies and within nucleoli. PNUTS expression levels are highest in brain, heart, lung, placenta, liver, kidney, pancreas and skeletal muscle.

This MAb recognizes a protein of ~17 kDa, identified as CELA3B (Chymotrypsin like elastase family member 3B). Elastases form a subfamily of serine proteases that hydrolyze many proteins in addition to elastin. Humans have six elastase genes which encode the structurally similar proteins elastase 1, 2, 2A, 2B, 3A, and 3B. Unlike other elastases, elastase 3B has little elastolytic activity. Like most of the human elastases, elastase 3B is secreted from the pancreas as a zymogen and, like other serine proteases such as trypsin, chymotrypsin and kallikrein; it has a digestive function in the intestine. Elastase 3B preferentially cleaves proteins after alanine residues. Elastase 3B may also function in the intestinal transport and metabolism of cholesterol. Both elastase 3A and elastase 3B have been referred to as protease E and as elastase 1, and excretion of this protein in fecal material is frequently used as a measure of pancreatic function in clinical assays.
